Project description:Understanding the initial and flow conditions of contemporary flows in Martian gullies, generally believed to be triggered and fluidized by CO2 sublimation, is crucial for deciphering climate conditions needed to trigger and sustain them. We employ the RAMMS (RApid Mass Movement Simulation) debris flow and avalanche model to back calculate initial and flow conditions of recent flows in three gullies in Hale crater. We infer minimum release depths of 1.0-1.5 m and initial release volumes of 100-200 m3. Entrainment leads to final flow volumes that are ∼2.5-5.5 times larger than initially released, and entrainment is found necessary to match the observed flow deposits. Simulated mean cross-channel flow velocities decrease from 3-4 m/s to ∼1 m/s from release area to flow terminus, while flow depths generally decrease from 0.5-1 to 0.1-0.2 m. The mean cross-channel erosion depth and deposition thicknesses are ∼0.1-0.3 m. Back-calculated dry-Coulomb friction ranges from 0.1 to 0.25 and viscous-turbulent friction between 100 and 200 m/s2, which are values similar to those of granular debris flows on Earth. These results suggest that recent flows in gullies are fluidized to a similar degree as are granular debris flows on Earth. Using a novel model for mass flow fluidization by CO2 sublimation we are able to show that under Martian atmospheric conditions very small volumetric fractions of CO2 of ≪1% within mass flows may indeed yield sufficiently large gas fluxes to cause fluidization and enhance flow mobility.

Project description:Xanthoria parietina (L.) Th. Fr. is a widely spread foliose lichen showing high tolerance against UV-radiation thanks to parietin, a secondary lichen substance. We exposed samples of X. parietina under simulated Martian conditions for 30 days to explore its survivability. The lichen's vitality was monitored via chlorophyll a fluorescence that gives an indication for active light reaction of photosynthesis, performing in situ and after-treatment analyses. Raman spectroscopy and TEM were used to evaluate carotenoid preservation and possible variations in the photobiont's ultrastructure respectively. Significant differences in the photo-efficiency between UV irradiated samples and dark-kept samples were observed. Fluorescence values correlated with temperature and humidity day-night cycles. The photo-efficiency recovery showed that UV irradiation caused significant effects on the photosynthetic light reaction. Raman spectroscopy showed that the carotenoid signal from UV exposed samples decreased significantly after the exposure. TEM observations confirmed that UV exposed samples were the most affected by the treatment, showing chloroplastidial disorganization in photobionts' cells. Overall, X. parietina was able to survive the simulated Mars conditions, and for this reason it may be considered as a candidate for space long-term space exposure and evaluations of the parietin photodegradability.

Project description:Microbial pathogens have evolved exquisite mechanisms to interfere and intercept host biological processes, often through molecular mimicry of specific host proteins. Ubiquitination is a highly conserved eukaryotic post-translational modification essential in determining protein fate, and is often hijacked by pathogenic bacteria. The conserved SKP1/CUL1/F-box (SCF) E3 ubiquitin ligase complex plays a key role in ubiquitination of proteins in eukaryotic cells. The F-box protein component of the SCF complex provides specificity to ubiquitination by binding to specific cellular proteins, targeting them to be ubiquitinated by the SCF complex. The bacterial pathogens. Legionella pneumophila, Agrobacterium tumefaciens, and Ralstonia solanacearum utilize type III or IV translocation systems to inject into the host cell eukaryotic-like F-box effectors that interact with the host SKP1 component of the SCF complex to trigger ubiquitination of specific host cells targets, which is essential to promote proliferation of these pathogens. Our bioinformatic analyses have identified at least 74 genes encoding putative F-box proteins belonging to 22 other bacterial species, including human pathogens, plant pathogens, and amebal endosymbionts. Therefore, subversion of the host ubiquitination machinery by bacterial F-box proteins may be a widespread strategy amongst pathogenic bacteria. The findings that bacterial F-box proteins harbor Ankyrin repeats as protein-protein interaction domains, which are present in F-box proteins of primitive but not higher eukaryotes, suggest acquisition of many bacterial F-box proteins from primitive eukaryotic hosts rather than the mammalian host.

Project description:Sulfur has been considered to be a predominant light element in the Martian core, and thus the sound velocity of Fe-S alloys at relevant high pressure and temperature is of great importance to interpret its seismological data. Here we measured the compressional sound velocity (VP) of liquid Fe, Fe80S20 and Fe57S43 using ultrasonic pulse-echo overlap method combined with a Kawai-type multi-anvil apparatus up to 20 GPa, likely corresponding to the condition at the uppermost core of Mars. The results demonstrate that the VP of liquid iron is least sensitive to its sulfur concentration in the Mars' whole core pressure range. The comparison of seismic wave speeds of Fe-S liquids with future observations will therefore tell whether the Martian core is molten and contains impurity elements other than sulfur.

Project description:Microorganisms growing at atmospheric pressures of 0.7 kPa may have a significant impact on the search for life on Mars. Data on their nutrient requirements in a simulated Martian environment are required to ascertain both the potential risk of forward contamination and the potential of past or present habitability of Mars. Serratia liquefaciens can grow at concomitant conditions of low pressure, low temperature, and anoxic atmosphere. Changes in the metabolic fingerprint of S. liquefaciens grown under varying physical conditions including diverse atmospheric pressures (0.7 kPa to 101.3 kPa), temperatures (30 °C or 0 °C), and atmospheric gas compositions (Earth or CO2) were investigated using Biolog GN2 assays. Distinct patterns for each condition were observed. Above 10 kPa S. liquefaciens performed similar to Earth-normal pressure conditions (101.3 kPa) whereas below 10 kPa shifts in metabolic patterns were observed. The differences indicated a physiological alteration in which S. liquefaciens lost its ability to metabolize the majority of the provided carbon sources at 0.7 kPa with a significant decrease in the oxidation of amino acids. By measuring the physiological responses to different carbon sources we were able to identify nutritional constraints that support cellular replication under simulated shallow Mars subsurface conditions.

Project description:Mars, with its ancient history of long-lived habitable environments, continues to captivate researchers exploring the potential for extant life. This study investigates the biosignature potential of Martian methane by assessing the viability of hydrogenotrophic methanogenesis in Methanosarcina barkeri MS under simulated Martian surface conditions. We expose M. barkeri to sustained hypobaria (7-12 mbar), low temperature (0˚C), and a CO2-dominated gas mixture mimicking the Martian atmosphere. The results demonstrate statistically quantifiable CH4 production under all tested conditions, including at 7-12 mbar. Transcriptomics reveal that low total pressure and temperature did not significantly impact gene expression, highlighting the resilience of M. barkeri. However, atmospheric gas composition, specifically Mars gas with 2.9% pH2, led to significant down-regulation of methanogenesis genes, hindering growth over 14 days. Notably, CH4 production scaled with the partial pressure of H2, revealing that hydrogen uptake affinity is a stronger predictor of habitability and methanogenic potential than favorable Gibbs free energy of reaction. Our findings suggest that Mars' subsurface could harbor habitable refugia capable of supporting methanogenesis, sustaining microbial life at low metabolic steady states. These insights challenge assumptions about Martian habitability and have implications for astrobiological exploration, planetary protection, and in situ resource utilization for future human missions.

Project description:The waters that were present on early Mars may have been habitable. Characterising environments analogous to these waters and investigating the viability of their microbes under simulated martian chemical conditions is key to developing hypotheses on this habitability and potential biosignature formation. In this study, we examined the viability of microbes from the Anderton Brine Springs (United Kingdom) under simulated martian chemistries designed to simulate the chemical conditions of water that may have existed during the Hesperian. Associated changes in the fluid chemistries were also tested using inductively coupled plasma-optical emission spectroscopy (ICP-OES). The tested Hesperian fluid chemistries were shown to be habitable, supporting the growth of all of the Anderton Brine Spring isolates. However, inter and intra-generic variation was observed both in the ability of the isolates to tolerate more concentrated fluids and in their impact on the fluid chemistry. Therefore, whilst this study shows microbes from fluctuating brines can survive and grow in simulated martian water chemistry, further investigations are required to further define the potential habitability under past martian conditions.

Project description:The martian surface environment exhibits extremes of salinity, temperature, desiccation, and radiation that would make it difficult for terrestrial microbes to survive. Recent evidence suggests that martian soils contain high concentrations of MgSO₄ minerals. Through warming of the soils, meltwater derived from subterranean ice-rich regolith may exist for an extended period of time and thus allow the propagation of terrestrial microbes and create significant bioburden at the near surface of Mars. The current report demonstrates that halotolerant bacteria from the Great Salt Plains (GSP) of Oklahoma are capable of growing at high concentrations of MgSO₄ in the form of 2 M solutions of epsomite. The epsotolerance of isolates in the GSP bacterial collection was determined, with 35% growing at 2 M MgSO₄. There was a complex physiological response to mixtures of MgSO₄ and NaCl coupled with other environmental stressors. Growth also was measured at 1 M concentrations of other magnesium and sulfate salts. The complex responses may be partially explained by the pattern of chaotropicity observed for high-salt solutions as measured by agar gelation temperature. Select isolates could grow at the high salt concentrations and low temperatures found on Mars. Survival during repetitive freeze-thaw or drying-rewetting cycles was used as other measures of potential success on the martian surface. Our results indicate that terrestrial microbes might survive under the high-salt, low-temperature, anaerobic conditions on Mars and present significant potential for forward contamination. Stringent planetary protection requirements are needed for future life-detection missions to Mars.

Project description:Iron oxide-hydroxide minerals in Martian dust provide crucial insights into Mars' past climate and habitability. Previous studies attributed Mars' red color to anhydrous hematite formed through recent weathering. Here, we show that poorly crystalline ferrihydrite (Fe5O8H · nH2O) is the dominant iron oxide-bearing phase in Martian dust, based on combined analyses of orbital, in-situ, and laboratory visible near-infrared spectra. Spectroscopic analyses indicate that a hyperfine mixture of ferrihydrite, basalt and sulfate best matches Martian dust observations. Through laboratory experiments and kinetic calculations, we demonstrate that ferrihydrite remains stable under present-day Martian conditions, preserving its poorly crystalline structure. The persistence of ferrihydrite suggests it formed during a cold, wet period on early Mars under oxidative conditions, followed by a transition to the current hyper-arid environment. This finding challenges previous models of continuous dry oxidation and indicates that ancient Mars experienced aqueous alteration before transitioning to its current desert state.

Project description:The habitability of Mars is determined by the physical and chemical environment. The effect of low water availability, temperature, low atmospheric pressure and strong UV radiation has been extensively studied in relation to the survival of microorganisms. In addition to these stress factors, it was recently found that silicates exposed to simulated saltation in a Mars-like atmosphere can lead to a production of reactive oxygen species. Here, we have investigated the stress effect induced by quartz and basalt abraded in Mars-like atmospheres by examining the survivability of the three microbial model organisms Pseudomonas putida, Bacillus subtilis, and Deinococcus radiodurans upon exposure to the abraded silicates. We found that abraded basalt that had not been in contact with oxygen after abrasion killed more than 99% of the vegetative cells while endospores were largely unaffected. Exposure of the basalt samples to oxygen after abrasion led to a significant reduction in the stress effect. Abraded quartz was generally less toxic than abraded basalt. We suggest that the stress effect of abraded silicates may be caused by a production of reactive oxygen species and enhanced by transition metal ions in the basalt leading to hydroxyl radicals through Fenton-like reactions. The low survivability of the usually highly resistant D. radiodurans indicates that the effect of abraded silicates, as is ubiquitous on the Martian surface, would limit the habitability of Mars as well as the risk of forward contamination. Furthermore, the reactivity of abraded silicates could have implications for future manned missions, although the lower effect of abraded silicates exposed to oxygen suggests that the effects would be reduced in human habitats.
