Project description:: The synthesis of metal nanoparticles using plant extracts is a very promising method in green synthesis. The medicinal value of Moringa oleifera leaves and the antimicrobial activity of metallic copper were combined in the present study to synthesize copper nanoparticles having a desirable added-value inorganic material. The use of a hydroalcoholic extract of M. oleifera leaves for the green synthesis of copper nanoparticles is an attractive method as it leads to the production of harmless chemicals and reduces waste. The total phenolic content in the M. oleifera leaves extract was 23.0 ± 0.3 mg gallic acid equivalent/g of dried M. oleifera leaves powder. The M. oleifera leaves extract was treated with a copper sulphate solution. A color change from brown to black indicates the formation of copper nanoparticles. Characterization of the synthesized copper nanoparticles was performed using ultraviolet-visible light (UV-Vis) spectrophotometry, Fourier-transform infrared (FTIR) spectrometry, high-resolution transmission electron microscopy (HRTEM), scanning electron microscopy (SEM), and X-ray diffraction (XRD). The synthesized copper nanoparticles have an amorphous nature and particle size of 35.8-49.2 nm. We demonstrate that the M. oleifera leaves extract and the synthesized copper nanoparticles display considerable antioxidant activity. Moreover, the M. oleifera leaves extract and the synthesized copper nanoparticles exert considerable anti-bacterial activity against Escherichia coli, Klebsiella pneumoniae, Staphylococcus aureus, and Enterococcus faecalis (MIC values for the extract: 500, 250, 250, and 250 µg/mL; MIC values for the copper nanoparticles: 500, 500, 500, and 250 µg/mL, respectively). Similarly, the M. oleifera leaves extract and the synthesized copper nanoparticles exert relatively stronger anti-fungal activity against Aspergillusniger, Aspergillus flavus, Candida albicans, and Candida glabrata (MIC values for the extract: 62.5, 62.5, 125, and 250 µg/mL; MIC values for the copper nanoparticles: 125, 125, 62.5, and 31.2 µg/mL, respectively). Our study reveals that the green synthesis of copper nanoparticles using a hydroalcoholic extract of M. oleifera leaves was successful. In addition, the synthesized copper nanoparticles can be potentially employed in the treatment of various microbial infections due to their reported antioxidant, anti-bacterial, and anti-fungal activities.

Project description:BackgroundHerein, we first time used the gum Moringa oleifera as reducing and capping agent for successful synthesis of silver nitrate and zinc oxide nanoparticles(NPs) through green synthesis approach. This study was aimed to check antibacterial activities of synthesized NPs against multidrug resistant bacteria methicillin-resistant Staphylococcus aureus (MRSA).MethodsAqueous solutions of AgNO3 and purified gum powder were mixed with 1:1 ratio, autoclaved at 120oC for 2 min. NPs pellet collected after centrifugation at 10,000 g for 20 min. ZnO NPs were prepared by mixing purified gum powder and metal salt with1:1 ratio, heated (70oC) and stirred at 100 rpm for 4 h followed by centrifugation at 10,000 g for 20 min. Pellet was washed and calcinated at 400oC for 4 h. Antibacterial potential against E. coli, S. aureus and methicillin-resistant Staphylococcus aureus (MRSA) was assessed by widely used Kirby-Bauer antibiotic susceptibility test.ResultsOptical observation of colour change from transparent to dark and UV-Visible analysis confirmed the synthesis of NPs. Fourier transform infrared spectroscopy (FTIR) of prepared nonmaterial revealed the characteristic AgNPs and ZnO stretch vibrations at wave number of 523 cm- 1 and 471 cm- 1resectively. Crystalline nature of AgNPs and ZnO NPs was confirmed by x-ray diffraction pattern with clear sharp Peaks. Scanning electron microscopy (SEM) revealed good surface morphology of AgNPs and ZnO NPs with 50nm and 60nm size respectively. AgNPs and ZnO NPs exhibited excellent antibacterial activity against E. coli (with zone of inhibition of 21 ± 02mm and 22 ± 03mm) and S.aureus ( with zone of inhibition of 20 ± 03mm and 21 ± 02mm) while good activity was observed against "super bug" methicillin-resistant Staphylococcus aureus (MRSA) with 16 ± 03mm ad 17 ± 02mm zone if inhibitions respectively.ConclusionsThis novel addition of Moringa Gum based nanoparticles will open new dimensions in the field of nanomedicine and pharmaceutics especially against MDR bacterial strains.

Project description:Moringa oleifera (M. oleifera) is globally recognized for its medicinal properties and offers high-quality, protein-rich seeds. This study aimed to explore the potential of M. oleifera seeds as a significant source of protein-based nanoparticles (PBNPs) using the ultrasonication technique after desolvation and to evaluate their cytotoxicity in the human leukemia cell line (THP-1) for the first time. The properties of the PBNPs were confirmed by dynamic light scattering (DLS), transmission electron microscopy (TEM), scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDX), X-ray diffraction (XRD), and Fourier-transform infrared spectroscopy (FT-IR). The extracted protein from moringa seed cake flour had a significant protein content of 54.20%, and the resulting PBNPs had an average size of 134.3 ± 0.47 nm with a robust zeta potential of -43.15 mV. Notably, our study revealed that PBNPs exhibited cytotoxic potential at high concentrations, especially against the THP-1 human leukemia cell line, which is widely used to study immunomodulatory properties. The inhibitory effect of PBNPs was quantitatively evidenced by a cytotoxicity assay, which showed that a concentration of 206.5 μg mL-1 (log conc. 2.315) was required to inhibit 50% of biological activity. In conclusion, our findings highlight the potential of M. oleifera seeds as a valuable resource in the innovative field of eco-friendly PBNPs by combining traditional medicinal applications with contemporary advancements in protein nanotechnology. However, further studies are required to ensure their biocompatibility.

Project description:An ultrahigh-pressure supercritical fluid extraction method was optimized and applied to extract seed oil lipids from two moringa species, namely Moringa oleifera (MO) and Moringa peregrina (MP). A full-factorial design was used to investigate the direct and interaction influence of pressure and temperature in the range of 40 to 80 MPa and 40 to 70 °C, respectively, on the extracted amount of oil from crushed seeds. The results revealed that pressure has a significant positive influence on the extracted amount of oil. The best extraction condition using neat CO2 was found at 80 MPa and 57 °C, yielding 396 ± 23 and 529 ± 26 mg oil per gram of seeds for MO and MP, respectively. An extraction kinetics study revealed a mainly solubility-controlled extraction of oil, and 28 g of CO2 was required to extract 400 mg of oil per gram of seeds of MO using the developed method. Addition of ethanol to the sample prior to the extraction increased the proportion of extractable polar lipids as well as the total amount of extracted oil. The developed method increased the extracted amount of oil twofold compared to a reference method based on solvent sonication. The obtained oil consisted mainly of glycerolipids, sterol esters, and phospholipids. Phospholipids, campesterol, and stigmasterol ester concentrations were found to be higher in MO while cholesterol ester was more abundant in MP.

Project description:Moringa oleifera Lam. is a tropical plant, used for centuries as food and traditional medicine. The aim of this study was to develop, validate and biochemically characterize an isothiocyanate-enriched moringa seed extract (MSE), and to compare the anti-inflammatory effects of MSE-containing moringa isothiocyanate-1 (MIC-1) with a curcuminoid-enriched turmeric extract (CTE), and a material further enriched in its primary phytochemical, curcumin (curcumin-enriched material; CEM). MSE was prepared by incubating ground moringa seeds with water to allow myrosinase-catalyzed enzymatic formation of bioactive MIC-1, the predominant isothiocyanate in moringa seeds. Optimization of the extraction process yielded an extract of 38.9% MIC-1. Phytochemical analysis of MSE revealed the presence of acetylated isothiocyanates, phenolic glycosides unique to moringa, flavonoids, fats and fatty acids, proteins and carbohydrates. MSE showed a reduction in the carrageenan-induced rat paw edema (33% at 500 mg/kg MIC-1) comparable to aspirin (27% at 300 mg/kg), whereas CTE did not have any significant effect. In vitro, MIC-1 at 1 μM significantly reduced the production of nitric oxide (NO) and at 5 μM, the gene expression of LPS-inducible nitric oxide synthase (iNOS) and interleukins 1β and 6 (IL-1β and IL-6), whereas CEM did not show any significant activity at all concentrations tested. MIC-1 (10μM) was also more effective at upregulating the nuclear factor (erythroid-derived 2)-like 2 (Nrf2) target genes NAD(P)H:quinone oxidoreductase 1 (NQO1), glutathione S-transferase pi 1 (GSTP1), and heme oxygenase 1 (HO1) than the CEM. Thus, in contrast to CTE and CEM, MSE and its major isothiocyanate MIC-1 displayed strong anti-inflammatory and antioxidant properties in vivo and in vitro, making them promising botanical leads for the mitigation of inflammatory-mediated chronic disorders.

Project description:The study analyzed the aqueous leaf extracts of Moringa oleifera and Musa sps. for phytochemical components, including flavonoids, sterols, saponins, tannins, and glycosides. The LC-MS analysis revealed gingerol, vicenin-2, caffeic acid, quercetin, and other compounds in the extracts. The synthesized MO- and MS- CuNPs (copper nanoparticles) exhibited plasmon resonance at 320 and 480 nm respectively, and Cu(II)-O stretching vibrations, manifested by their Fourier Transform Infrared (FT-IR) spectra. Thermogravimetric analysis showed that MO-/MS-CuNPs experienced a total weight loss of 80.2% and 68% respectively. Further, the EDS analysis revealed that MO-CuNPs have a copper content of 20%, while MS-CuNPs is 32%. The X-ray diffraction (XRD) patterns of the annealed MO and MS-CuNPs indicate the presence of both the cubic phase of copper (Cu) and the monoclinic tenorite phase of copper oxide (CuO). The percent DPPH scavenging activity of MO- and MS-CuNPs is 34.4 and 28.8, and by KMnO4 is 63.8 and 47.3% respectively. The behavior of CuNPs exhibited an asymmetrical response, with greater inhibition of Gram-negative bacteria compared to Gram-positive bacteria. CuNPs and medicinal plant-derived carbon dots were integrated into nanofibrous electrospun biopolymer scaffolds for wound care, demonstrating 100% wound healing in Yorkshire pigs full thickness excision wounds after 34 days.

Project description:Silver nanoparticles (AgNPs), widely recognized for their nanoscale geometric size and unique properties, such as large specific surface area, high permeability, and high safety, were synthesized using the endophytic fungus Penicillium polonicum PG21 through a green approach. Four key synthesis factors-48 h, 45 °C, pH 9.0, and 80 mM AgNPs concentration-were optimized. Characterization via ultraviolet-visible spectroscopy, transmission electron microscopy, Fourier-transform infrared spectroscopy, and X-ray diffraction revealed the AgNPs as approximately 3-25 nm spherical particles with numerous functional groups ensuring stability. AgNPs were tested against various fungal and bacterial plant pathogens, including Botrytis cinerea (EB-1), Alternaria alternata (EB-2, EB-3), Fusarium solani (RG-1), Williamsia serinedens (SL-1), Sphingopyxis macrogoltabida (SL-2), Bacillus velezensis (SL-3), and Pseudomonas mediterranea (SL-4), causing agricultural challenges. PG21-synthesized AgNPs exhibited inhibition rates against all tested fungi, with 60 μg/mL AgNPs demonstrating optimal inhibition rates. Notably, EB-1 experienced a significant growth inhibition, reaching an inhibition rate reached of 74.22 ± 1.54%. Conversely, RG-1 exhibited the smallest inhibitory effect at 48.13 ± 0.92%. The effect of AgNPs on safflower seed germination and growth revealed notable increases in shoot length, fresh weight, stem length, and number of lateral roots-1.4, 1.4, 1.33, and 10.67 times higher than the control, respectively, at an AgNPs concentration of 80 μg/mL. In conclusion, green-synthesized AgNPs demonstrate pathogen toxicity, showcasing potential applications in disease management for industrial crops and promoting plant growth.

Project description:The aim of the study was to investigate the antibacterial and anticoagulant activity of Moringa (Moringa oleifera) seed extracts and coagulant protein for their potential application in water treatment. Pathogenic microorganisms were obtained from Ramachandra Hospital, Chennai, India. Bacterial cell aggregation and growth kinetics studies were employed for six bacterial strains with different concentrations of seed extracts and coagulant protein. Moringa seed extract and coagulant protein showed cell aggregation against six bacterial strains, whereas seed extract alone showed growth inhibition of all six bacterial strains for up to 6 h, compared to that of control. Escherichia coli and Salmonella para typhi B did not develop resistance against coagulant protein. The results imply that Moringa oleifera is likely an efficient low-molecular bioactive peptide (with <7.5 kDa plant-based coagulant and antimicrobial peptides, confirmed by applying amino acid sequences), using liquid chromatography-mass spectrometry and HPLC, with the corresponding sequences from Napin-1A peptide posing different degrees of antibacterial activity against different pathogenic organisms.

Project description:The employment of plant extracts in the synthesis of metal nanoparticles is a very attractive approach in the field of green synthesis. To benefit from the potential synergy between the biological activities of the Moringa oleifera and metallic bismuth, our study aimed to achieve a green synthesis of phytochemical encapsulated bismuth nanoparticles using a hydroalcoholic extract of M. oleifera leaves. The total phenolic content in the M. oleifera leaves extract used was 23.0 ± 0.3 mg gallic acid equivalent/g of dried M. oleifera leaves powder. The physical properties of the synthesized bismuth nanoparticles were characterized using UV-Vis spectrophotometer, FT-IR spectrometer, TEM, SEM, and XRD. The size of the synthesized bismuth nanoparticles is in the range of 40.4-57.8 nm with amorphous morphology. Using DPPH and phosphomolybdate assays, our findings revealed that the M. oleifera leaves extract and the synthesized bismuth nanoparticles possess antioxidant properties. Using resazurin microtiter assay, we also demonstrate that the M. oleifera leaves extract and the synthesized bismuth nanoparticles exert potent anti-bacterial activity against Escherichia coli, Klebsiella pneumoniae, Staphylococcus aureus, and Enterococcus faecalis (estimated MIC values for the extract: 500, 250, 250, and 250 µg/mL; estimated MIC values for the bismuth nanoparticles: 500, 500, 500, and 250 µg/mL, respectively). Similarly, the M. oleifera leaves extract and the synthesized bismuth nanoparticles display relatively stronger anti-fungal activity against Aspergillus niger, Aspergillus flavus, Candida albicans, and Candida glabrata (estimated MIC values for the extract: 62.5, 62.5, 125, and 250 µg/mL; estimated MIC values for the bismuth nanoparticles: 250, 250, 62.5, and 62.5 µg/mL, respectively). Thus, green synthesis of bismuth nanoparticles using M. oleifera leaves extract was successful, showing a positive antioxidant, anti-bacterial, and anti-fungal activity. Therefore, the synthesized bismuth nanoparticles can potentially be employed in the alleviation of symptoms associated with oxidative stress and in the topic treatment of Candida infections.

Project description:Effect of ultrasonic power on the structure and functional properties of water-soluble protein extracted from defatted Moringa oleifera seed were explored. The results showed that ultrasonic treatment could reduce β-sheet and β-turn content of water-soluble protein from Moringa oleifera seed (MOWP) and increase the content of random coil and α-helix. Changes in intrinsic fluorescence spectra, surface hydrophobicity (H0) and thermal behaviors indicated that ultrasonic had significant effect on the tertiary structure of MOWP. The results of SEM and SDS-PAGE showed that the MOWP was aggregated but not significantly degraded by ultrasound. The solubility, foaming properties and emulsifying properties of MOWP increased firstly and then decreased with the increase of ultrasonic power. Ultrasonic treatment altered the functional properties of MOWP, which might be attributed to the exposure of hydrophilic group and the change of and secondary and tertiary structure.
