Solvent-induced polymorphism in dipodal N-donor ligands containing a biphenyl core.

ABSTRACT: Polymorph screenings for two related dipodal N-donor ligands containing a biphenyl core, namely 4,4'-bis(pyridin-4-ylmethyl)-1,1'-biphenyl (1) and 4,4'-bis(1H-imidazol-1-ylmethyl)-1,1'-biphenyl (2) were performed, and the new phases were isolated and their crystal structures analysed. Profiling included methods such as PXRD and thermal analysis. Hirshfeld surface analyses, as well as crystal lattice energy calculations provided deeper insight in the interplay of the intermolecular forces and the stability of the isolated phases. Furthermore, our studies revealed the presence of solvent-induced polymorphism, whereby the metastable phase is dominant upon crystallisation from THF (1a) and EtOH (2c). Upon heating, these phases transform into a more stable form, whereby the transformations were followed by PXRD studies (1, 2).

Project description:Cp*IrIII and CpIrIII complexes have attracted interest as catalysts for oxidative transformations, and highly oxidizing iridium species are postulated as key intermediates in both catalytic water and C-H bond oxidation. Strongly electron-donating ligand sets have been shown to stabilize IrIV complexes. We describe the synthesis and reactivity of complexes containing the CpIr(biphenyl-2,2'-diyl) moiety stabilized by a series of strong donor carbon-based ligands. The oxidation chemistry of these complexes has been characterized electrochemically, and a singly oxidized IrIV species has been observed by X-band EPR for the complex CpIr(biph)(p-CNCH2SO2C6H4CH3).

New tetrahydroisoquinoline-based P-glycoprotein modulators: decoration of the biphenyl core gives selective ligands.

Project description:P-glycoprotein (P-gp, MDR1) is a membrane transporter expressed in several regions of our body. It plays a crucial defense role as it mediates the efflux of hundreds of potentially toxic substances. However, P-gp is one of the main causes of failure in cancer chemotherapy, as a number of chemotherapeutic agents are P-gp substrates. Another interesting implication concerns the correlation between P-gp expression impairment and the onset of several central nervous system pathologies such as Alzheimer's and Parkinson's diseases. In view of these considerations, in the present study, a new series of P-gp modulators have been designed, synthesized and evaluated for their activity towards P-gp and two other sister proteins (BCRP and MRP1). The compounds, structurally correlated to the potent but non-selective P-gp inhibitor MC70 [4'-(6,7-dimethoxy-3,4-dihydro-1H-isoquinolin-2-ylmethyl)biphenyl-4-ol], proved fairly selective towards P-gp, with a potency in the micromolar range. Compounds 5a, 5d and 12d proved capable of restoring doxorubicin toxicity in resistant cancer cells.

Synthesis and reactivity of palladium(II) fluoride complexes containing nitrogen-donor ligands.

Project description:This article describes the synthesis, characterization, and reactivity of palladium(II) fluoride complexes containing sp(2) and sp(3) nitrogen-containing supporting ligands. Both cis and trans complexes of general structure (N)(N')Pd(II)(R)(F) (R = Ar or CH(3)) as well as cis-(N)(2)Pd(II)(F)(2) are reported. Crystallographic characterization of these molecules has allowed structural comparisons to related phosphine-ligated species. Furthermore, these studies have revealed that nitrogen-donor ligands support some of the longest and the shortest Pd-F bonds reported to date. The thermal decomposition of (N)(N')Pd(II)(R)(F) has also been examined, and no products of C-F bond-forming reductive elimination were obtained in any case.

Unusual polymorphism in new bent-shaped liquid crystals based on biphenyl as a central molecular core.

Project description:Bent-shaped mesogens possessing a biphenyl as a central core have been synthesized and the role of the terminal chain and the orientation of the ester as a linkage group have been investigated. For the studied molecular core we have established that both parameters play an important role for the mesomorphic properties. The polyfluoroalkyl terminal chain supports the formation of mesophases, and the introduction of a chiral lactate terminal chain destabilizes mesophases for the first type of mutual orientation of ester groups, attached to the central core. On the contrary, for the opposite orientation of esters, the terminal chain has no effect on the mesomorphic properties, and columnar phases have been found for all compounds. A unique phase sequence has been found for the mesogen with the fluorinated chain. A generalized tilted smectics, SmCG, have been observed in a temperature interval between two different lamellar SmCP phases and characterized by X-ray and dielectric measurements. The dielectric spectroscopy data are unique and presented for the first time in the SmCG phase providing new information about the molecular dynamics.

Solvent-Induced Polymorphism of Iron(II) Spin Crossover Complexes.

Project description:Two new mononuclear iron(II) compounds (1) and (2) of the general formula [Fe(L)₂](BF₄)₂·nCH₃CN (L = 4-(2-bromoethyn-1-yl)-2,6-bis(pyrazol-1-yl)pyridine, n = 1 for (1) and n = 2 for compound (2)), were synthesized. The room temperature crystallization afforded concomitant formation of two different solvent analogues: compound (1) exhibiting triclinic P-1 and compound (2) monoclinic C2/c symmetry. Single-crystal X-ray studies confirmed the presence of the LS (low-spin) state for both compounds at 180 K and of the HS (high-spin) state for compound (2) at 293 K, in full agreement with the magnetic investigations for both solvent polymorphs. Compound (1) exhibits spin transition above 293 K followed by subsequent solvent liberation, while the spin transition of (2) takes already place at 237 K. After complete solvent removal from the crystal lattice, compound (1d) (the desolvated polymorph derived from (1)) exhibits spin transition centered at 342 K accompanied by a thermal hysteresis loop, while the analogous compound (2d) (the desolvated derivate of compound (2)) remains blocked in the HS state over all the investigated temperature range.

Screening of different interactions in oxo-manganese porphyrin dimers containing axial N-donor ligands: a theoretical study.

Project description:A theoretical analysis for describing the dimeric assemblies of high-valent manganese(v)-oxo meso-tetraphenylporphyrin (TPP) ([(TPP)MnVO]2 2+) and meso-tetrakis(pentafluorophenyl)porphyrin (TPFPP) ([(TPFPP)MnVO]2 2+) in the presence of axial N-donor ligands is presented. Our theoretical results revealed two types interactions in dimers: a sandwich-like interaction between phenyl rings of porphyrin molecules, and a non-bonded T-shape interaction between nitrogen donors attached to Mn centers. The curvature in the geometry of porphyrin in the [(TPP)MnVO]2 2+/N-donor system is significantly smaller than that of [(TPFPP)MnVO]2 2+/N-donor system. Moreover, the Mn-N(ax) distances in [(TPFPP)MnVO]2 2+/N-donor system are shorter than those of [(TPP)MnVO]2 2+/N-donor system. Also, the donor-acceptor interaction between the imidazoles and the Mn centers are stronger than those of the other ligands in both porphyrins. These results are supported by atoms in molecules (AIM) and natural bond orbital (NBO) analysis.

Development of diverse adjustable axially chiral biphenyl ligands and catalysts.

Project description:Development of highly efficient and practical chiral ligands and catalysts is an eternal theme in asymmetric synthesis. Here, we report the design, synthesis, and evaluation of a new kind of adjustable axially chiral biphenyl ligands and catalysts, in which six model reactions including asymmetric additions of diethylzinc or alkynes to aldehydes in the presence of axially chiral [1,1'-biphenyl]-2,2'-diol ligands, palladium-catalyzed asymmetric cycloadditions in the presence of phosphoramidite ligands, and chiral phosphoric acid-catalyzed asymmetric synthesis of 1,1'-spirobiindane-7,7'-diol derivative and [4 + 3] cyclization were attempted. The results showed that variation of 2,2'-substituent groups could provide different types of ligands and catalysts, and adjustment of substituent groups at the 3,3', 5,5', 6,6'-positions could make ligands and catalysts more efficient in the asymmetric catalytic synthesis. Therefore, our present research should provide a new and useful strategy for development of diverse axially chiral ligands and catalysts.

Enantioselectivity Induced by Stereoselective Interlocking: A Novel Core Motif for Tropos Ligands.

Project description:Well-defined supramolecular interactions are a powerful tool to control the stereochemistry of a catalytic reaction. In this paper, we report a novel core motif for fluxional 2,2'-biphenyl ligands carrying (S)-amino acid-derived interaction sites in 5,5'-position that cause spontaneous enrichment of the Rax rotamer. The process is based on strong non-covalent interlocking between interaction sites, which causes diastereoselective formation of a supramolecular ligand dimer, in which the axial chirality of the two subunits is dictated by the stereochemical information in the amino acid residues. The detailed structure of the dimer was elucidated by NMR spectroscopy and single-crystal X-ray analysis. Three different phosphorus-based ligand types, namely a bisphosphine, a bisphosphinite and a phosphoramidite were synthesized and characterized. Whereas the first one was found to exist in a strongly weighted equilibrium, the two others each exhibited stereoconvergent behavior transforming into the diastereopure Rax rotamer. Enriched ligands were used in rhodium-mediated asymmetric hydrogenation reactions of prochiral olefins in which very high enantioselectivities of up to 96:4 were achieved.

Project description:Crystal structures for a series of bis-(acetyl-acetonato)oxovanadium(IV) complexes containing N-donor pyridyl ligands are reported, namely, bis-(acetyl-acetonato-κ2 O,O')oxido(pyridine-κN)vanadium(IV), [V(C5H7O2)2O(C5H5N)], 1, bis-(acetyl-acetonato-κ2 O,O')oxido(pyridine-4-carbo-nitrile-κN)vanadium(IV), [V(C5H7O2)2O(C6H4N2)], 2, and bis-(acetyl-acetonato-κ2 O,O')(4-meth-oxy-pyridine-κN)oxidovanadium(IV), [V(C5H7O2)2O(C6H7NO)], 3, Compounds 1-3 have the formulae VO(C5H7O2)2 L, where L = pyridine (1), 4-cyano-pyridine (2), and 4-meth-oxy-pyridine (3). Compound 1 was previously reported [Meicheng et al. (1984 ▸). Kexue Tongbao, 29, 759-764 and DaSilva, Spiazzi, Bortolotto & Burrow (2007). Acta Crystallogr., E63, m2422] and redetermined here at cryogenic temperatures. Compounds 1 and 2 as pyridine and 4-cyano-pyridine adducts, respectively, crystallize as distorted octa-hedral structures with the oxo and pyridyl ligands trans to one another. A crystallographic twofold axis runs through the O-V-N bonds. Compound 3 containing a 4-meth-oxy-pyridine ligand crystallizes as a distorted octa-hedral structure with the oxo and pyridyl ligands cis to one other, removing the twofold symmetry seen in the other complexes.

The Anticancer Activities of Some Nitrogen Donor Ligands Containing bis-Pyrazole, Bipyridine, and Phenanthroline Moiety Using Docking Methods.

Project description:The anticancer study of nitrogen-chelating ligands can be of tremendous help in choosing ligands for the anticancer metal complexes design especially with ruthenium(II). The inhibitory anticancer activities of some nitrogen-chelating ligands containing bis-pyrazole, bipyridine, and phenanthroline were studied using experimental screening against cancer cell and theoretical docking methods. In vitro anticancer activities showed compound 11 as the most promising inhibitor, and the computational docking further indicates its strong inhibitory activities towards some cancer-related receptors. Among the twenty-one modelled ligands, pyrazole-based compounds 7, 11, and 15 are the most promising inhibitors against the selected receptors followed by 18 and 21 which are derivatives of pyridine and phenanthroline, respectively. The presence of the carboxylic unit in the top five ligands that displayed stronger inhibitory activities against the selected receptors is an indication that the formation of noncovalent interactions such as hydrogen bonding and a strong electron-withdrawing group in these compounds are very important for their receptor interactions. The thermodynamic properties, the polarizabilities, and the LUMO energy of the compounds are in the same patterns as the observed inhibitory activities.
