Project description:Annually, agricultural activity produces an enormous amount of plant biomass by-product. Many studies have reported the biomethane potential of agro-industrial wastes, but only a few studies have investigated applying the substrates in both batch and continuous mode. Tomato is one of the most popular vegetables globally; its processing releases a substantial amount of by-product, such as stems and leaves. This study examined the BMP of tomato plant (Solanum lycopersicum Mill. L. cv. Alfred) waste. A comparative test revealed that the BMPs of corn stover, tomato waste,and their combination were approximately the same, around 280 mL methane/g Volatile Solid. In contrast, the relative biogas production decreased in the presence of tomato waste in a continuous mesophilic anaerobic digestion system; the daily biogas productions were 860 ± 80, 290 ± 50, and 570 ± 70 mL biogas/gVolatile Solid/day in the case of corn stover, tomato waste, and their mixture, respectively. The methane content of biogas was around 46-48%. The fermentation parameters of the continuous AD experiments were optimal in all cases; thus, TW might have an inhibitory effect on the microbial community. Tomato plant materials contain e.g. flavonoids, glycoalkaloids (such as tomatine and tomatidine), etc. known as antimicrobial and antifungal agents. The negative effect of tomatine on the biogas yield was confirmed in batch fermentation experiments. Metagenomic analysis revealed that the tomato plant waste caused significant rearrangements in the microbial communities in the continuously operated reactors. The results demonstrated that tomato waste could be a good mono-substrate in batch fermentations or a co-substrate with corn stover in a proper ratio in continuous anaerobic fermentations for biogas production. These results also point to the importance of running long-term continuous fermentations to test the suitability of a novel biomass substrate for industrial biogas production.

Project description:Viruses represent major disease transmitting agents carried by human excreta and animal manure. Understanding virus inactivation is therefore essential in preventing microbial spread due to inadequate treatment of these materials. Here, we investigated the inactivation kinetics of the single-stranded (ss) RNA phage MS2, DNA phages T4 and ΦX174, andthe double-stranded DNA human adenovirus in stored human urine, sludge, and animal manure, at temperatures and pH valuestypical of storage under naturally occurring conditions or mesophilic anaerobic digestion (<40 °C). The ssRNA phage MS2 was most readily inactivated in all samples compared to the other viruses tested. This is consistent with previous findings in wellcontrolled buffer solutions of similar composition, where inactivation was found to be governedby bases (NH3, carbonate, hydroxide) that catalyze the transesterification and cleavage of the ssRNA. Correspondingly, MS2 inactivation kinetics in real matrices could be adequately modelled by only taking into account the effects of temperature, pH, carbonate and ammonia on the integrity of ssRNA. DNA viruses were more persistent compared to MS2;however, inactivation in selected sludge and manure samples proceeded at faster rates compared to well-controlled buffersolutions of similar composition. This indicates a contribution of microbial or enzymatic activity to inactivation of DNA viruses. Overall, this study identifies the most important factors contributing to inactivation of viruses in human excreta and manure, and highlights the differences in inactivation kinetics and mechanisms between ssRNA and DNA viruses.

Project description:Sulfur reduction at hyperthermoacidophilic conditions represents a promising opportunity for metal sulfide precipitation from hot acidic metallurgical streams, avoiding costly cooling down. The suitability of mesophilic anaerobic sludges as the inoculum for sulfur-reducing bioreactors operated at high temperature and low pH was explored. We examined sludges from full-scale anaerobic reactors for sulfur-reducing activity at pH 2.0-3.5 and 70 or 80 °C, with H2 as an electron donor. At pH 3.5 in batch experiments, sulfidogenesis started within 4 days, reaching up to 100-200 mg·L-1 of dissolved sulfide produced after 19-24 days, depending on the origin of the sludge. Sulfidogenesis resumed after removing H2S by flushing with nitrogen gas, indicating that sulfide was limiting the conversion. The best performing sludge was used to inoculate a 4 L gas-lift reactor fed with H2 as the electron donor, CO2 as the carbon source, and elemental sulfur as the electron acceptor. The reactor was operated in semibatch mode at a pH 3.5 and 80 °C, and stable sulfide production rates of 60-80 mg·L-1·d-1 were achieved for a period of 24 days, without formation of methane or acetate. Our results reveal the potential of mesophilic anaerobic sludges as seed material for sulfur-reducing bioprocesses operated at hyperthermoacidophilic conditions. The process needs further optimization of the volumetric sulfide production rate to gain relevance for practice.

Project description:Anaerobic digestion (AnD) is a microbiological process that converts organic waste materials into biogas. Because of its high methane content, biogas is a combustible energy source and serves as an important environmental technology commonly used in the management of animal waste generated on large animal farms. Much work has been done on hardware design and process engineering for the generation of biogas. However, little is known about the complexity of the microbiology in this process. In particular, how microbes interact in the digester and eventually breakdown and convert organic matter into biogas is still regarded as a "black box." We used 16S rRNA sequencing as a tool to study the microbial community in laboratory hog waste digesters under tightly controlled conditions, and systematically unraveled the distinct interaction networks of two microbial communities from mesophilic (MAnD) and thermophilic anaerobic digestion (TAnD). Under thermophilic conditions, the well-known association between hydrogen-producing bacteria, e.g., Ruminococcaceae and Prevotellaceae, and hydrotrophic methanogens, Methanomicrobiaceae, was reverse engineered by their interactive topological niches. The inferred interaction network provides a sketch enabling the determination of microbial interactive relationships that conventional strategy of finding differential taxa was hard to achieve. This research is still in its infancy, but it can help to depict the dynamics of microbial ecosystems and to lay the groundwork for understanding how microorganisms cohabit in the anaerobic digester.

Project description:The potential of periodate (PI) in sludge anaerobic digestion is not tapped, although it has recently attracted great research interest in organic contaminants removal and pathogens inactivation in wastewater treatment. This is the first work to demonstrate significant improvement in methane generation from waste activated sludge (WAS) with PI pretreatment and to provide underlying mechanisms. Biochemical methane potential tests indicated that methane yield enhanced from 100.2 to 146.3 L per kg VS (VS, volatile solids) with PI dosages from 0 to 100 mg per g TS (TS, total solids). Electron spin resonance showed PI could be activated without extra activator addition, which might be attributed to the native transition metals (e.g., Fe2+) in WAS, thereby generating hydroxyl radical (•OH), superoxide radicals (•O2 -), and singlet oxygen (1O2). Further scavenging tests demonstrated all of them synergistically promoted WAS disintegration, and their contributions were in the order of •O2 - > •OH > 1O2, leading to the release of substantial biodegradable substances (i.e., proteins and polysaccharides) into the liquid phase for subsequent biotransformation. Moreover, fluorescence and ultraviolet spectroscopy analyses indicated the recalcitrant organics (especially lignocellulose and humus) could be degraded by reducing their aromaticity under oxidative stress of PI, thus readily for methanogenesis. Microbial community analysis revealed some microorganisms participating in hydrolysis, acidogenesis, and acetoclastic methanogenesis were enriched after PI pretreatment. The improved key enzyme activities and up-regulated metabolic pathways further provided direct evidence for enhanced methane production. This research was expected to broaden the application scope of PI and provide more diverse pretreatment choices for energy recovery through anaerobic digestion.
