Project description:We successfully develop a two-in-one approach to generate non-viral genome specific targeted CAR T cells through CRISPR/Cas9. In the adoptive therapy for relapsed/refractory aggressive B-cell non-Hodgkin lymphoma, we observed durable responses without serious adverse events and complete remission in patients treated with these PD1 knockout CAR T cells. Here we applied single-cell sequencing to analyze the characteristcis of T cells prepared by different methods.

Project description:The rapidly developing miniaturization in numerous fields require low-demanding but robust methods of nanomaterial production. Colloidal synthesis provides great flexibility in product material, size, and shape. Gold nanoparticle synthesis has been thoroughly studied, however, recent reports on mechanistic insights of crystal formation have been hindered by the numerous procedures and parameter optimization works. With every new study, scientists fill another blank space on the map of understanding anisotropic growth and find out the critical parameters. In the current work, we highlight the choice importance for surfactant supplier in achieving the gold nanotriangle formation. We systematically study the variation in the shape yield when utilizing five batches of cetyltrimethylammonium chloride (CTAC) from varied suppliers. Using analytical techniques, we search for deviations causing such variation, e.g. different impurity content. We found only a marginal effect of iodine contamination on the studied system, excluding this factor as decisive in contrast to what was proposed earlier in the literature, and leaving the high dependency of the yield to originate from yet unknown reagent characteristics. A deeper understanding of these factors would provide highly effective protocols lowering the reagent consumption and increasing the accessibility of nanomaterials manufactured in a sustainable manner.

Project description:In the past decade, there has been considerable interest in radiosensitization using gold nanoparticles that accumulate specifically in cancerous tissue while sparing normal tissues. Despite this interest, it remains unclear which nanoparticle morphologies, cellular uptake, or cytoplasmic distribution elicit optimal radiosensitization. We introduce gold nanotriangles (AuNTs) as a possible X-ray radiotherapy sensitizer. In this study, we first explored a large-scale synthetic method for the production of high quality monodisperse AuNTs. Second, we conducted in vitro and in vivo experiments to evaluate the effect of PEGylated AuNTs (pAuNTs) on cellular uptake, cytotoxicity, bio-distribution, and radiosensitization on radiation-resistant human Glioblastoma Multiforme (GBM) cells. Our results suggest that the new scale up synthesis methodology consistently produced high quality AuNTs and pAuNTs which had nonspecific cellular uptake without any obvious cytotoxicity and exhibited excellent radiosensitization.

Project description:INTRODUCTION: Fibrin tissue adhesive, which has applications in several areas of medicine, can be prepared by different methods. AIM: To compare fibrin tissue adhesives prepared by 3 different methods. METHOD: In this prospective experimental laboratory study, fibrin tissue adhesives prepared by the use of plasma fibrinogen (group 1), cryoprecipitation (group 2), and precipitation by ammonium sulfate (group 3) were tested on 15 rabbits and 10 fragments of dura mater. The quality of the clots was assessed in terms of the success of the healing process, local toxicity, graft adhesion capacity, and degree of adhesion of 2 fragments of dura mater produced. RESULTS: All methods produced a clot with high adhesion and no toxicity, but tensile strength testing revealed that the glue produced from the ammonium sulfate-precipitated clot (group 3) was the strongest, requiring 39 g/cm(2) to separate the fragments as opposed to 23 g/cm(2) for group 2 and 13 g/cm(2) for group 1. CONCLUSION: All methods produced good results as far as clot formation and non-toxicity, but ammonium sulfate precipitation produced the best tensile strength and was thus the most effective method of preparing fibrin tissue adhesive.

Project description:We report an efficient, seed-mediated method for the synthesis of gold nanotriangles (NTs) which can be used for controlled self-assembly. The main advantage of the proposed synthetic protocol is that it relies on using stable (over the course of several days) intermediate seeds. This stability translates into increasing time efficiency of the synthesis and makes the protocol experimentally less demanding ('fast addition' not required, tap water can be used in the final steps) as compared to previously reported procedures, without compromising the size and shape monodispersity of the product. We demonstrate high reproducibility of the protocol in the hands of different researchers and in different laboratories. Additionally, this modified seed-mediated method can be used to produce NTs with edge lengths between ca. 45 and 150 nm. Finally, the high 'quality' of NTs allows the preparation of long-range ordered assemblies with vertically oriented building blocks, which makes them promising candidates for future optoelectronic technologies.

Project description:Gold nanotriangles (AuNTs) functionalized with dithiolated oligonucleotides have been employed to develop an amplification-free electrochemical biosensor for SARS-CoV-2 in patient samples. Gold nanotriangles, prepared through a seed-mediated growth method and exhaustively characterized by different techniques, serve as an improved electrochemical platform and for DNA probe immobilization. Azure A is used as an electrochemical indicator of the hybridization event. The biosensor detects either single stranded DNA or RNA sequences of SARS-CoV-2 of different lengths, with a low detection limit of 22.2 fM. In addition, it allows to detect point mutations in SARS-CoV-2 genome with the aim to detect more infective SARS-CoV-2 variants such as Alpha, Beta, Gamma, Delta, and Omicron. Results obtained with the biosensor in nasopharyngeal swab samples from COVID-19 patients show the possibility to clearly discriminate between non-infected and infected patient samples as well as patient samples with different viral load. Furthermore, the results correlate well with those obtained by the gold standard technique RT-qPCR, with the advantage of avoiding the amplification process and the need of sophisticated equipment.

Project description:Turbostratic graphene is a multilayer graphene, which has exotic electrical properties similar to those of monolayer graphene due to the low interlayer interaction. Additionally, the stacking structure of the turbostratic multilayer graphene can decrease the effect of attachment of charge impurities and surface roughness. This paper explores the growth of high-purity and high-quality turbostratic graphene with different interlayer spacings by calcining ferric chloride and sucrose at 1000 °C for 1 h under an argon atmosphere. X-ray diffraction patterns and Raman results imply that the turbostratic graphene contains two different interlayer spacings: 3.435 and 3.55 Å. The 3.55 Å turbostratic graphene is on top of the 3.435 Å turbostratic graphene, and there is an AB stacking pattern between the topmost graphene layer of 3.435 Å turbostratic graphene and the first graphene layer of the 3.55 Å turbostratic graphene, with an interlayer spacing of 3.35 Å. The two different interlayer spacings of turbostratic graphene arise from different cooling rates between the higher temperature ranges (>700 °C) and lower temperatures (<700 °C).

Project description:Four methods of preparing makgeolli, a traditional Korean turbid rice wine, were reported in this study. The four processing routes include single-stage simultaneous saccharification and fermentation of glutinous rice with nuruk - a Korean starter culture (1SF-N), single-stage fermentation with nuruk and yeast (1SF-YN), two-stage fermentation (2SF) and three-stage fermentation (3SF). Chemical analysis was used to determine how the different processing routes could affect the rice wine's properties in terms of alcohol content, pH, colour, mineral content, proximate composition, antioxidant activity, total phenolic content, sugar, free amino acid, and organic acid profile. Sensory analysis using polarised projective mapping (PPM) and 62 participants found that sweetness is the most desirable attribute for makgeolli among New Zealand consumers with sourness and bitterness as less desirable. The 2SF makgeolli sample had the highest concentration of glucose (8.2 mg/mL) and maltose (107 mg/mL) and in the PPM experiment was the most preferred out of the four processing methods. The 1SF-N makgeolli sample had the highest alcohol (13% ABV), crude protein (4.9%), antioxidant activity, total phenolic (621 mg GAE/L) and free amino acids content, however, it was the least overall liked makgeolli sample. Overall, the novelty of this research includes formulating a traditional Korean turbid rice wine in a Western country environment and evaluating consumer perception of makgeolli beyond the normal clientele in South Korea. From these results it is suggested that the properties of makgeolli can be manipulated via processing to suit the brewer's sensory needs that best fits the consumer market.
