Tag and Capture Flow Hydrogen Exchange Mass Spectrometry with a Fluorous-Immobilized Probe.

Project description:Analysis of complex mixtures of proteins by hydrogen exchange (HX) mass spectrometry (MS) is limited by one's ability to resolve the protein(s) of interest from the proteins that are not of interest. One strategy for overcoming this problem is to tag the target protein(s) to allow for rapid removal from the mixture for subsequent analysis. Here we illustrate a new solution involving fluorous conjugation of a retrievable probe. The appended fluorous tag allows for facile immobilization on a fluorous surface. When a target protein is passed over the immobilized probe molecule, it can be efficiently captured and then exposed to a flowing stream of deuterated buffer for hydrogen exchange. The utility of this method is illustrated for a model system of the Elongin BC protein complex bound to a peptide from HIV Vif. Efficient capture is demonstrated, and deuteration when immobilized was identical to deuteration in conventional solution-phase hydrogen exchange MS. Protein captured from a crude bacterial cell lysate could also be deuterated without the need for separate purification steps before HX MS. The advantages and disadvantages of the method are discussed in light of miniaturization and automation.

Hydrogen sulfide sensing using an aurone-based fluorescent probe.

Project description:Hydrogen sulfide detection and sensing is an area of interest from both an environmental and a biological perspective. While many methods are currently available, the most sensitive and biologically applicable ones are fluorescence based. In general, these fluorescent probes are based upon large, high-molecular weight, well-characterized fluorescent scaffolds that are synthetically demanding to prepare and difficult to tune and modify. In this study, we have reported a new reduction-based, rationally designed and synthesized turn-on fluorescent probe (Z)-2-(4'-azidobenzylidene)-5-fluorobenzofuran-3(2H)-one (6g) utilizing a low molecular weight aurone fluorophore. During these studies, the modular nature of the synthesis was used to quickly overcome problems with solubility, overlap of excitation of the probe and reduced product, and rate of reaction, resulting in a final compound that is efficient and sensitive for the detection of hydrogen sulfide. The limitation of slow reaction and the reduced fluorescence in a biologically relevent medium was solved by employing cationic surfactant cetyltrimethyl ammonium bromide (CTAB). The probe features a high fluorescence enhancement, fast response (10-30 min), and good sensitivity (1 μm) and selectivity for hydrogen sulfide.

A Visible Colorimetric Fluorescent Probe for Hydrogen Sulfide Detection in Wine.

Project description:A new efficient and practical fluorescent probe 6-(benzo[d]thiazol-2-yl)naphthalen-2-yl-thiophene-2-carboxylate (probe 1) was synthesized to detect hydrogen sulfide (H2S). The addition of H2S caused the solution of probe 1 to change from colorless to yellow, and the solution of probe 1 changes to different colors with respect to different concentrations of H2S. Importantly, probe 1 could help detect H2S efficiently by a distinct color response as a visible detection agent. Probe 1 reacted with various concentrations of H2S (0-200 μM), and the detection limit for H2S was 0.10 μM. Particularly, probe 1 can be applied as a sensor to detect H2S accurately in wine samples.

A Cell Trappable Methyl Rhodol-Based Fluorescent Probe for Hydrogen Sulfide Detection.

Project description:Hydrogen sulfide is a biologically important molecule and developing chemical tools that enable further investigations into the functions of H2 S is essential. Fluorescent turn-on H2 S probes have been developed for use in cellulo and in vivo, but the membrane permeability of these probes can lead to probe leakage and signal attenuation over time. Here we report a cell trappable fluorescent probe for H2 S, CT-MeRhoAz, which is based on a methylrhodolazide scaffold derivatized with an acetoxymethyl ester group. Prior to ester cleavage, the CT-MeRhoAz probe generates a 2500-fold turn-on response to H2 S, which is enhanced to a 3000-fold response for the carboxylic acid form of the probe. Additionally, the probe is highly selective for H2 S over other biologically relevant sulfur, oxygen, and nitrogen-based analytes. Live cell imaging experiments confirmed the biocompatibility of CT-MeRhoAz and also that it is cell trappable, unlike the parent MeRhoAz scaffold.

A Single Fluorescent Probe to Visualize Hydrogen Sulfide and Hydrogen Polysulfides with Different Fluorescence Signals.

Project description:Hydrogen sulfide (H2 S) and hydrogen polysulfides (H2 Sn , n>1) are endogenous regulators of many physiological processes. In order to better understand the symbiotic relationship and cellular cross-talk between H2 S and H2 Sn , it is highly desirable to develop single fluorescent probes which enable dual-channel discrimination between H2 S and H2 Sn . Herein, we report the rational design, synthesis, and evaluation of the first dual-detection fluorescent probe DDP-1 that can visualize H2 S and H2 Sn with different fluorescence signals. The probe showed high selectivity and sensitivity to H2 S and H2 Sn in aqueous media and in cells.

A New Fluorescent Probe for Hydrogen Sulfide Detection in Solution and Living Cells.

Project description:Since Hydrogen Sulfide (H2S) was recognized as a gas transmitter, its detection and quantification have become a hot research topic among chemists and biologists. In this area, fluorescent probes have shown great advantages: fast and strong response, low detection limit and easy manipulation. Here we developed a new fluorescent probe that detected H2S selectively among various bioactive and inorganic salts. This probe was based on the core structure of fluorescein and reacted with H2S through azide-reduction. Great linearity was achieved correlating fluorescence intensity and H2S concentrations in solution. The detection of H2S in cancer cells was also achieved.

A turn-on fluorescent probe with a dansyl fluorophore for hydrogen sulfide sensing.

Project description:Hydrogen sulfide (H2S) is a biologically relevant molecule that has been newly identified as a gasotransmitter and is also a toxic gaseous pollutant. In this study, we report on a metal complex fluorescent probe to achieve the sensitive detection of H2S in a fluorescent "turn-on" mode. The probe bears a dansyl fluorophore with multidentate ligands for coordination with copper ions. The fluorescent "turn-on" mode is facilitated by the strong bonding between H2S and the Cu(ii) ions to form insoluble copper sulfide, which leads to the release of a strongly fluorescent product. The H2S limit of detection (LOD) for the proposed probe is estimated to be 11 nM in the aqueous solution, and the utilization of the probe is demonstrated for detecting H2S in actual lake and mineral water samples with good reproducibility. Furthermore, we designed detector vials and presented their successful application for the visual detection of gaseous H2S.

A biotin-guided hydrogen sulfide fluorescent probe and its application in living cell imaging.

Project description:Hydrogen sulfide (H2S), a well-known signaling molecule, exerts significant regulatory effects on the cardiovascular and nervous systems. Therefore, monitoring the metabolism of H2S offers a potential mechanism to detect various diseases. In addition, biotin is significantly used as a targeting group to detect cancer cells exclusively. In this work, a biotin-guided benzoxadizole-based fluorescent probe, NP-biotin, was developed for H2S detection and evaluated in normal liver cell (LO2) and liver cancer cell (HepG2) lines. Results reveal that NP-biotin can detect cellular H2S with high sensitivity and selectivity. Moreover, NP-biotin has been confirmed to possess the ability to target cancer cells under the guidance of the biotin group.
