Project description:The study of urban spatial interaction is closely linked to that of economic geography, urban planning, regional development, and so on. Currently, this topic is generating a great deal of interest among researchers who are striving to find accurate ways to measure urban spatial interaction. Classical spatial interaction models lack theoretical guidance and require complicated parameter-adjusting processes. The radiation model, however, as proposed by Simini et al. with rigorous formula derivation, can simulate directional urban spatial interaction. We applied the radiation model in China to simulate the directional migration number among 337 nationwide research units, comprising 4 municipalities and 333 prefecture-level cities. We then analyzed the overall situation in Chinese cities, the interaction intensity hierarchy, and the prime urban agglomerations from the perspective of migration. This was done to ascertain China's urban spatial interaction and regional development from 2000 to 2010 to reveal ground realities.

Project description:Urban shrinkage is currently spreading at global level. At the same time, the scale of urban shrinkage is not limited to urban agglomerations, metropolitan areas, or big cities, but begins to expand to a vast number of small towns. Over the years, the characteristics, models, and mechanisms of shrinkage in large cities have attracted the attention of scholars; however, the problem of shrinkage in small towns has not been fully discussed. In China, small towns are located at the margins of cities and are the first part of the countryside; hence, they are the link and the bridge between urban and rural areas, and a space carrier to solve the diseases of big cities and for rural rejuvenation as a whole. However, in the process of rapid urbanization, some small towns have experienced urban shrinkage, mainly through a decline in township enterprises and the deterioration of the living environment, which has restricted their role in coordinating the spatial support of urban and rural development. Therefore, a correct understanding of the shrinkage of small towns has considerable theoretical and practical guiding significance. We focused on the towns of the Jilin Province as the research unit, and combined township population, economy, land use, and employment indices to establish an urban shrinkage index, identifying the status, spatial distribution, and influencing factors of small towns shrinkage in the Jilin Province. Moreover, we analyzed the similarities and differences of small towns shrinkage between the Jilin Province and developed countries. The results show that small towns shrinkage in the Jilin Province shares similar characteristics with developed countries, as well as important differences in aspects such as population migration, low-level industrial structure, and administrative division adjustments.

Project description:Atmospheric particulate matter (PM) is a recognized risk factor for the global burden of disease in human populations. We are presenting here the application of toxicogenomics in the evaluation of the toxic effects of organic content of atmospheric particle matter (PM), from urban and rural environments (city of Barcelona and village of La Pobla, NE Spain), using the developing zebrafish embryo. The main goal is to identify the metabolic pathways involved in the adverse effects observed in zebrafish embryos exposed to PM organic content from urban and rural environments, also allowing the selection of genes of interest that are differentially expressed. The relevance of particle size to the PM toxicity is also addressed. Indeed, the zebrafish embryos were exposed to PM of aerodynamic diameter larger than 7.2 μm and smaller than 0.5 μm (PM10 and PM0.5, respectively). PM0.5 concentrated biological and toxic activities linked to organic substances. Transcriptomic analyses showed strong induction of the AhR signalling pathway (a.k.a. dioxin-like activity) for embryos exposed to both rural and urban extracts, correlating with the concentrations of PAHs. Urban extracts, with strong contribution of traffic emissions, specifically de-regulated oxidative stress-related genes, as well as pancreatic and eye-lens specific genes, two organs known to be affected by air pollution in humans. Exposure to rural extracts, with high contribution of wood burning emissions, affected genes implicated in basic cellular functions, in agreement with their strong embryotoxicity. Extracts from rural and urban samples elicited both common and specific transcriptome responses, suggesting different potentially adverse outcomes depending on PM source and composition. The PM filter samples used in the present study were PM10 and PM0.5 from the urban site - total of 4 samples from 2 sampling months; and PM0.5 from the rural site - total of 2 samples from 2 sampling months (PM10 samples from the rural site were not tested due to their very low concentration in organic compounds, and insignificant biological activity, previously measured). Sampling occurred during the cold periods of the year 2012/2013. The organic fraction of PM samples was extracted by sonication using a mixture of dichloromethane:methanol. Then, the extracts were filtered, evaporated and re-dissolved in Dimethyl sulfoxide (DMSO). Zebrafish fertilized eggs were exposed to PM organic extracts (0.2% DMSO) from the urban and rural sites from 24 to 120h post-fertilization. Total RNA was isolated from whole embryos (pools of 20 individuals), using Trizol reagent protocol (Invitrogen Life Technologies, Carlsberg, CA). The microarray study was performed using the Agilent Two-colour D. rerio Oligo Microarray v3 platform, following the Agilent Microarray â?? Based Gene Expression Analysis protocol. Three biological replicates were performed for each PM sample and controls. Biological replicates, a technical replicate and a self-to-self, were simultaneously amplified and labelled using Cyanine 3 (Cy3) and Cyanine 5 (Cy5) dye. After cRNA purification (RNeasy Kit, Quiagen GmbH, Hilden, Germany), the cRNA concentration and labelling were quantified in the NanoDrop spectrophotometer, obtaining incorporation rates in the range of 15-20pmol of cyanide dye/µl cRNA and yield values > 0.825ug, as recommended. cRNA was fragmented and hybridized in 4x44K arrays, for 17h at 65oC. After the hybridization the array slides were washed, and immediately scanned using Microarray Scanner Agilent G2505C system. Data was extracted using Agilent Feature Extraction Software v10.5.1.1, and the quality of microarray data was evaluated using the Quality Control report provided by Agilent Software. Based on the microarray data analysis, 22 primers were selected and designed using Primer Express 2.0 software (Applied Biosystems, Foster City, CA). Genes were quantified by real-time PCR to validate the microarray data, and a subset of 13 genes were further selected by their robust behaviour in the validation assays, allowing to differentiate the toxic potential of PM from urban and rural sources.

Project description:The molecular basis of evolutionary change is assumed to be genetic variation. However, growing evidence suggests that epigenetic mechanisms, such as DNA methylation, may also be involved in evolutionary change. An important first step in evaluating this hypothesis is to test for the presence of epigenetic variation between natural populations living under different environmental conditions. In the current study we explored variation between populations of Darwin’s finches living in adjacent “urban” and “rural” environments on Santa Cruz Island in the Galápagos. We tested for morphological, genetic, and epigenetic differences between the urban and rural populations of each of two species of ground finches, Geospiza fortis and G. fuliginosa. Using data collected from more than 1000 birds, we found significant morphological differences between populations of G. fortis, but not G. fuliginosa. We did not find genetic differences between populations of either species, based on comparisons of copy number variation (CNV). In contrast, we did find epigenetic differences between the urban and rural populations of both species, based on DNA methylation analysis. We explored genomic features and gene associations of the differentially methylated regions (DMR), as well as their possible functional significance. In summary, our study documents local population epigenetic variation within species of Darwin’s finches.

Project description:Continuous access to adequate quantities of safe water is essential for human health and socioeconomic development. Piped water systems (PWSs) are an increasingly common type of water supply in rural African small towns. We assessed temporal and spatial patterns in water consumption from public standpipes of four PWSs in Ghana in order to assess clean water demand relative to other available water sources. Low water consumption was evident in all study towns, which manifested temporally and spatially. Temporal variability in water consumption that is negatively correlated with rainfall is an indicator of rainwater preference when it is available. Furthermore, our findings show that standpipes in close proximity to alternative water sources such as streams and hand-dug wells suffer further reductions in water consumption. Qualitative data suggest that consumer demand in the study towns appears to be driven more by water quantity, accessibility, and perceived aesthetic water quality, as compared to microbiological water quality or price. In settings with chronic under-utilization of improved water sources, increasing water demand through household connections, improving water quality with respect to taste and appropriateness for laundry, and educating residents about health benefits of using piped water should be prioritized. Continued consumer demand and sufficient revenue generation are important attributes of a water service that ensure its function over time. Our findings suggest that analyzing water consumption of existing metered PWSs in combination with qualitative approaches may enable more efficient planning of community-based water supplies and support sustainable development.

Project description:Epigenetic variation has the potential to control environmentally dependent development and contribute to phenotypic responses to local environments. Environmental epigenetic studies of sexual organisms confirm the responsiveness of epigenetic variation, which should be even more important when genetic variation is lacking. A previous study of an asexual snail, Potamopyrgus antipodarum, demonstrated that different populations derived from a single clonal lineage differed in both shell phenotype and methylation signature when comparing lake versus river populations. Here, we examine methylation variation among lakes that differ in environmental disturbance and pollution histories. The differential DNA methylation regions (DMRs) identified among the different lake comparisons suggested a higher number of DMRs and variation between rural Lake 1 and one urban Lake 2 and between the two urban Lakes 2 and 3, but limited variation between the rural Lake 1 and urban Lake 3. DMR genomic characteristics and gene associations were investigated. Observations suggest there is no effect of geographic distance or any consistent pattern of DMRs between urban and rural lakes. Environmental factors may influence epigenetic response.

Project description:With social and economic environment changes occurring in the world and deepening of the urbanization process, China's urban development exhibits a new phenomenon of growth and shrinkage fluctuations. The resource-based city shrinkage phenomenon is particularly prominent. Research on the commonalities and patterns of similar groups should be enhanced. We constructed an urban shrinkage evaluation index system from the three dimensions of population, economy and space. Accordingly, we explored the spatiotemporal evolution characteristics of 175 resource-based cities in China from a multidimensional perspective with the entropy method, shrinkage model and transfer matrix method. The results indicated that most resource-based cities in China occurred in the non-shrinking state, but their development speed gradually decreased or even presented stagflation. The shrinkage measure-related results in the different dimensions revealed that the number of shrinking cities is increasing. The population, economic and comprehensive shrinkage levels were mainly slight and remained stable. The number of cities experiencing moderate and severe shrinkage was relatively small and mostly encompassed short-term shrinkage. Spatial shrinkage demonstrated a clear administrative hierarchy difference. Moreover, the spatial distribution range of shrinking cities in each dimension expanded and exhibited obviously similar characteristics, i.e., shrinking cities were relatively concentrated in Northeast China, while they were more scattered in other regions. Furthermore, the geodetector technique was applied to reveal the influencing factors of resource-based city growth and shrinkage. Based on the results, the change in the secondary industry output value share at the start of the study was the primary factor. The impact of each employment structure indicator from 2014 to 2018 was particularly significant. Comprehensive exploration of the shrinkage characteristics of this particular group of cities and their development behavior from a multidimensional perspective can provide an important reference for the transformation and high-quality development of resource-based cities.

Project description:Understanding the spatial distribution of urban forest ecosystem services is essential for urban planners and managers to effectively manage cities and is an essential part of sustainable urban development. Mapping the spatial distribution of urban forest ecosystem services and improving the accuracy of its assessment scale will undoubtedly provide a more accurate reference basis for later management. In this study, we used the i-Tree Eco model and kriging interpolation to quantify and map urban forest ecosystem services and their spatial distribution in Zhengzhou, a city along the lower reaches of the Yellow River in China; analyzed the mapping errors and applicable conditions; and further explored the spatial differences using geographic probes. The i-Tree Eco model estimation results showed that the total carbon storage in the urban forest of Zhengzhou city was 75.7 tons, the annual carbon sequestration was 14.66 tons, the trees and shrubs in the urban area of Zhengzhou city could effectively avoid a total of 307.86 m3 of surface runoff per year, and trees and shrubs removed 411.8 kg/year of air pollution (O3, CO, NO2, PM2.5, PM10, and SO2). The spatial distribution of all urban forest ecosystem services showed significant heterogeneity, but the spatial evaluation precision of different factors varied. GDP and population data showed a negative correlation with ecosystem services, and ecosystem services were abundant in watershed and woodland areas. This study differs from traditional assessments based on regional data due to its improved spatial evaluation accuracy, and the results, discussion, and analysis not only help Zhengzhou's own urban development, but also provide a basis for the future construction and management of other cities, the Central Plains urban agglomeration, and the surrounding larger regions. This will contribute to the enhancement of ecosystem services and thus improve the ecological conditions of the region. This will also have a positive effect on the health of urban residents.
