Project description:Vast amounts of dyeing wastewater released from the textile industry can not only cause water pollution but also have negative effects on the human body, such as skin irritation and respiratory diseases. Dye adsorption technology is necessary for the treatment of wastewater discharged from the dyeing industry and for environmental improvement. However, to remove dyeing wastewater, more energy and solvents are used to fabricate adsorbents, or excessive energy is used to filter dyeing wastewater out, resulting in more environmental pollution. Therefore, it is necessary to develop a method of filtering dyeing wastewater in a more environmentally friendly manner by minimizing the use of solvents and energy. In this study, we modified the surface of a textile substrate through UV irradiation to create a monomer capable of facilely bonding with dyes. Employing the UV photografting method, we were able to produce a dye adsorption filter in a more environmentally friendly manner, minimizing solvent usage and heat energy consumption required for absorbent synthesis. At a monomer concentration of 10%, the fabricated filter exhibited a dye removal efficiency of 97.34% after 24 h, all without the need for a pressure treatment or temperature increase. Moreover, it displayed an adsorption capacity of approximately 77.88 mg per 1 g of filter material.

Project description:This study proposes the treatment and valorization of denim textile effluents through a fermentative hydrogen production process. Also, the study presents the decolorizing capabilities of bacterial and fungal isolates obtained from the fermented textile effluents. The maximum hydrogen production rate was 0.23 L H2/L-d, achieving at the same time color removal. A total of thirty-five bacteria and one fungal isolate were obtained from the fermented effluents and screened for their abilities to decolorize indigo dye, used as a model molecule. From them, isolates identified as Bacillus BT5, Bacillus BT9, Lactobacillus BT20, Lysinibacillus BT32, and Aspergillus H1T showed notable decolorizing capacities. Lactobacillus BT20 reached 90% of decolorization using glucose as co-substrate after 11 days of incubation producing colorless metabolites. Bacillus BT9 was able to utilize the indigo dye as the sole carbon source achieving a maximum decolorization of 60% after 9 days of incubation and producing a red-colored metabolite. In contrast, Bacillus BT5 and Lysinibacillus BT32 exhibited the lowest percentages of decolorization, barely 33% after 16 and 11 days of incubation, respectively. When Aspergillus H1T was grown in indigo dye supplemented with glucose, 96% of decolorization was reached after 2 days. This study demonstrates the valorization of denim textile effluents for the production of hydrogen via dark fermentation with concomitant color removal.

Project description:Glycosylphosphatidylinositols (GPIs) are lipid anchors allowing the exposure of proteins at the outer layer of the plasma membrane. In fungi, a number of GPI-anchored proteins (GPI-APs) are involved in the remodeling of the cell wall polymers. GPIs follow a specific biosynthetic pathway in the endoplasmic reticulum. After the transfer of the protein onto the GPI-anchor, a lipid remodeling occurs to substitute the diacylglycerol moiety by a ceramide. In addition to GPI-APs, A. fumigatus produces a GPI-anchored polysaccharide, the galactomannan (GM), that remains unique in the fungal kingdom. To investigate the role of the GPI pathway in the biosynthesis of the GM and cell wall organization, the deletion of PER1-coding for a phospholipase required for the first step of the GPI lipid remodeling-was undertaken. Biochemical characterization of the GPI-anchor isolated from GPI-APs showed that the PER1 deficient mutant produced a lipid anchor with a diacylglycerol. The absence of a ceramide on GPI-anchors in the Δper1 mutant led to a mislocation of GPI-APs and to an alteration of the composition of the cell wall alkali-insoluble fraction. On the other hand, the GM isolated from the Δper1 mutant membranes possesses a ceramide moiety as the parental strain, showing that GPI anchor of the GM follow a distinct unknown biosynthetic pathway.

Project description:BACKGROUND:Fungal infections affect various parts of the body and can be difficult to treat. Aspergillus infection causes a spectrum of diverse diseases particularly in lung according to host immunity. The two major entities are invasive pulmonary aspergillosis and chronic pulmonary aspergillosis. Candida infections can be superficial or invasive. Superficial infections often affect the skin or mucous membranes. However, invasive fungal infections are often life-threatening. Advances in nanotechnology have opened new horizons in nanomedicine, allowing the synthesis of nanoparticles that can be assembled into complex architectures. Novel studies and technologies are devoted to understanding the mechanisms of disease for the design of new drugs. OBJECTIVES:In the present study, the antifungal activity of biogenic selenium nanoparticles (Se NPs) against Aspergillus fumigatus and Candida albicans was investigated. MATERIALS AND METHODS:Se-reducing bacteria previously identified as Bacillus sp. MSh-1 were used for the intracellular biosynthesis of elemental Se NPs. The shape, size, and purity of the extracted NPs were determined with various instrumental techniques. The nanoparticles antifungal characterization mainly derives from the following pathways: (i) to generate sustained flux of nano-ions from the compounds that deposited on special substrates or imbedded in colloidal or semisolid matrices. (ii) To transport active those ions to sensitive targets on plasma membrane of fungi. RESULTS:The results of energy-dispersive X-ray demonstrated that the purified NPs consisted of only Se. In addition, transmission electron micrographs showed that 120- to 140-nm spherical Se NPs were the most common. An antifungal assay was performed with a standard Clinical and Laboratory Standards Institute broth microdilution method. Minimum inhibitory concentration (MIC) measurements of the antifungal activity of the Se NPs against C. albicans (70 μg/mL) and A. fumigatus (100 μg/mL) showed that yeast cells were more sensitive than mold cells. CONCLUSIONS:The MICs against A. fumigatus (100 μg/mL) and C. albicans (70 μg/mL) showed that biogenic Se NPs are useful antifungal agents.

Project description:A bacterial strain RMLRT03 with ability to decolorize textile dye Acid Orange dye was isolated from textile effluent contaminated soil of Tanda, Ambedkar Nagar, Uttar Pradesh (India). The decolorization studies were performed in Bushnell and Haas medium (BHM) amended with Acid Orange dye. The bacterial strain was identified as Staphylococcus hominis on the basis of 16S rDNA sequence. The bacterial strain exhibited good decolorization ability with glucose and yeast extract supplementation as cosubstrate in static conditions. The optimal condition for the decolorization of Acid Orange dye by Staphylococcus hominis RMLRT03 strain were at pH 7.0 and 35°C in 60 h of incubation. The bacterial strain could tolerate high concentrations of Acid Orange dye up to 600 mg l(-1). The high decolorizing activity under natural environmental conditions indicates that the bacterial strain has practical application in the treatment of dye containing wastewaters.

Project description:Currently, bionanotechnologies are attracting great interest due to their promising results and potential benefits on many aspects of life. In this study, the objectives was to biosynthesis CuO-NPs using cell-free extract(s) of copper-resistant fungi and use them in bioremediation of textile industry wastewater. Out of 18 copper-resistant fungal isolates, the novel fungus strain Fusarium oxysporum OSF18 was selected for this purpose. This strain showed a high efficiency in extracellular reducing copper ions to their nano-form. The myco-synthesized CuO-NPs were characterized using UV-Vis spectroscopy, HRTEM, FTIR, and XRD and were found to be spherical nanocrystals with the size range of 21-47 nm. The bio-synthesized CuO-NPs showed promising antimicrobial activity as well as high efficiency in removing heavy metals and textile dye from industrial wastewater. The myco-synthesized CuO-NPs immobilized in alginate beads exhibited superior microbial disinfection (99.995%), heavy metals removal (93, 55, and 30 % for Pb, Cr, and Ni, respectively), and dye decolorization (90%). Such results represent a promising step to produce an eco-friendly, cost-effective, and easy-to handle tool for the bioremediation of textile industry wastewater.

Project description:The release of different hazardous substances into the water bodies during the industrial and textile processing stages is a serious problem in recent decades. This study focuses on the potentiality of Fe3O4-NPs-based polymer in sustainable bioremediation of toxic substances from contaminated water. The biosynthesis of Fe3O4-NPs by A. tamarii was performed for the first time. The effect of different independent variables on the Fe3O4-NPs production were optimized using Plackett-Burman design and central composite design (CCD) of Response Surface Methodology. The optimum Fe3O4-NPs production was determined using incubation period (24 h), temperature (30 °C), pH (12), stirring speed (100 rpm) and stirring time (1 h). The incorporation of Fe3O4-NPs into chitosan beads was successfully performed using sol-gel method. The modified nanocomposite exhibited remarkable removal capability with improved stability and regeneration, compared to control beads. The optimal decolorization was 94.7% at 1.5 g/l after 90 min of treatment process. The reusability of biosorbent beads displayed 75.35% decolorization after the 7th cycle. The results showed a highly significant reduction of physico-chemical parameters (pH, TDS, TSS, COD, EC, and PO4) of contaminated wastewater. The sorption trials marked Fe3O4-NPs-based biopolymer as efficient and sustainable biosorbent for the elimination of hazardous toxic pollutants of wastewater in a high-speed rate.

Project description:Upon infection of a host, the pathogenic fungus Aspergillus fumigatus is attacked by the reactive oxygen species produced by phagocytic cells. Detoxification of hydrogen peroxide by catalases was proposed as a way to overcome this host response. A. fumigatus produces three active catalases; one is produced by conidia, and two are produced by mycelia. The mycelial catalase Cat1p was studied previously. Here we characterized the two other catalases, their genes, and the phenotypes of gene-disrupted mutants. CatAp, a spore-specific monofunctional catalase, is resistant to heat, metal ions, and detergent. This enzyme is a dimeric protein with 84.5-kDa subunits. The 749-amino-acid polypeptide exhibits high levels of similarity to the Aspergillus nidulans CatA catalase and to bacterial catalase HPII of Escherichia coli. In spite of increased sensitivity to H(2)O(2), killing of DeltacatA conidia by alveolar macrophages and virulence in animals were similar to the killing of conidia by alveolar macrophages and virulence in animals observed for the wild type. In contrast to the Cat1p and CatAp catalases, the mycelial Cat2p enzyme is a bifunctional catalase-peroxidase and is sensitive to heat, metal ions, and detergent. This enzyme, an 82-kDa monomer, is homologous to catalase-peroxidases of several fungi and bacteria. Surprisingly, mycelium of the double Deltacat1Deltacat2 mutant with no catalase activity exhibited only slightly increased sensitivity to H(2)O(2) and was as sensitive to killing by polymorphonuclear neutrophils as mycelium of the wild-type strain. However, this mutant exhibited delayed infection in the rat model of aspergillosis compared to infection by the wild-type strain. These results indicate that conidial catalase is not a virulence factor and that mycelial catalases transiently protect the fungus from the host.

Project description:Galactofuranose-containing molecules have been repeatedly shown to be important antigens among human fungal pathogens, including Aspergillus fumigatus. Immunogenic galactofuran determinants have been poorly characterized chemically, however. We reported here the characterization of two glycoproteins of A. fumigatus with an N-glycan containing galactofuranose. These proteins are a phospholipase C and a phytase. Chemical characterization of the N-glycan indicates that it is a mixture of Hex(5-13)HexNAc(2) oligosaccharides, the major molecular species corresponding to Hex(6-8)HexNAc(2). The N-glycan contained one galactofuranose unit that was in a terminal nonreducing position attached to the 2 position of Man. This single terminal nonreducing galactofuranose is essential for the immunoreactivity of the N-glycans assessed either with a monoclonal antibody that recognizes a tetra-beta-1,5-galactofuran chain of galactomannan or with Aspergillus-infected patient sera.

Project description:In this work, the treatment of textile wastewater by a facile and high-efficiency technology using eco-friendly Aspergillus niger as a biosorbent was investigated. We measured physical changes (weight, size) during the formation and growth of fungus pellets and the pH values that influence the adsorption performance and biosorption mechanism. Three acid anionic dyes containing Acid Orange 56, Acid Blue 40 and Methyl Blue were chosen as model dyes to investigate batch adsorption efficiency. Two adsorption models (in vivo and in vitro) were adopted to decolorize the acid dyes. The results show that fungus pellets have excellent decoloration abilities with a high adsorption efficiency of 98% for 200 mg L-1 of acid dye. The pH value of the dye solution varied with the adsorption time and the dye removal efficiency greatly depended on the pH. The bioadsorption mechanism of nano-scale hyphae was revealed to be mainly due to electrostatic interactions caused by the pH change. Furthermore, the surface morphologies of the fungus after adsorption indicated that the dyes had been adsorbed on the surface of the fungus mycelia. Moreover, prepared 3D fungus/GO aerogels demonstrated superior dye removal abilities compared with fungus aerogels.
