Effect of Shear History on Solid-Liquid Transition of Particulate Gel Fuels.

ABSTRACT: Investigating the structural evolution of particulate gels is a very challenging task due to their vulnerability and true flow characteristics. In this work, deeper insight into the rheological properties of gel fuels filled with fumed silica (FS) and aluminum microparticles (Al MPs) was gained by changing shear procedures. Firstly, the flow curves were found to no longer follow the monotonic power law and exhibited subtle thixotropic responses. As the shear rate increased, the gel structure underwent a transition from local shear to bulk shear in the nonlinear region after yielding. This finding reveals the prevalence of nonideal local shear in industry. Secondly, the time-dependent rheological responses demonstrated that the strength spectrum of gel fuels depends on the applied shear rate, with stress relaxation more easily observed at lower shear rates. Those results involved the structural disruption, recovery, and equilibrium of particulate gels from two scales of shear rate and shear time.

Biomolecular condensates undergo a generic shear-mediated liquid-to-solid transition.

Project description:Membrane-less organelles resulting from liquid-liquid phase separation of biopolymers into intracellular condensates control essential biological functions, including messenger RNA processing, cell signalling and embryogenesis1-4. It has recently been discovered that several such protein condensates can undergo a further irreversible phase transition, forming solid nanoscale aggregates associated with neurodegenerative disease5-7. While the irreversible gelation of protein condensates is generally related to malfunction and disease, one case where the liquid-to-solid transition of protein condensates is functional, however, is that of silk spinning8,9. The formation of silk fibrils is largely driven by shear, yet it is not known what factors control the pathological gelation of functional condensates. Here we demonstrate that four proteins and one peptide system, with no function associated with fibre formation, have a strong propensity to undergo a liquid-to-solid transition when exposed to even low levels of mechanical shear once present in their liquid-liquid phase separated form. Using microfluidics to control the application of shear, we generated fibres from single-protein condensates and characterized their structural and material properties as a function of shear stress. Our results reveal generic backbone-backbone hydrogen bonding constraints as a determining factor in governing this transition. These observations suggest that shear can play an important role in the irreversible liquid-to-solid transition of protein condensates, shed light on the role of physical factors in driving this transition in protein aggregation-related diseases and open a new route towards artificial shear responsive biomaterials.

Liquid-liquid-solid transition in viscoelastic liquids.

Project description:Liquid-liquid-solid transitions (LLST) are known to occur in confined liquids, exist in supercooled liquids and emerge in liquids driven from equilibrium. Molecular dynamics (MD) simulations claim many successes in forecasting the phenomena. The transitions are also studied in the framework of thermodynamics based methods and minimalistic models. In here, the proposed approach is derived in the framework of continuum and includes spatial and temporal dynamic heterogeneities; the approach is meant to capture the material behavior at small scales. We conjecture that the liquid-like and solid-like behaviors are dissimilar enough for the two to be governed by different constitutive relations. In this way, we gain additional degree of freedom, which is found essential when predicting the transitional phenomena. As a result, we derive the LLST criteria for liquids in equilibrium, during steady flow and at transient conditions. Lastly, we forecast short-lived LLSTs in human blood during cardiac cycle.

Colossal and reversible barocaloric effect in liquid-solid-transition materials n-alkanes.

Project description:Emerging caloric cooling technology provides a green alternative to conventional vapor-compression technology which brings about serious environmental problems. However, the reported caloric materials are much inferior to their traditional counterparts in cooling capability. Here we report the barocaloric (BC) effect associated with the liquid-solid-transition (L-S-T) in n-alkanes. A low-pressure of ~50 MPa reversibly triggers an entropy change of ~700 J kg-1 K-1, comparable to those of the commercial refrigerants in vapor-based compression systems. The Raman study and theoretical calculations reveal that applying pressure to the liquid state suppresses the twisting and random thermal motions of molecular chains, resulting in a lower configurational entropy. When the pressure is strong enough to drive the L-S-T, the configurational entropy will be fully suppressed and induce the colossal BC effect. This work could open a new avenue for exploring the colossal BC effect by evoking L-S-T materials.

Liquid to solid transition of elastin condensates.

Project description:Liquid-liquid phase separation of tropoelastin has long been considered to be an important early step in the complex process of elastin fiber assembly in the body and has inspired the development of elastin-like peptides with a wide range of industrial and biomedical applications. Despite decades of study, the material state of the condensed liquid phase of elastin and its subsequent maturation remain poorly understood. Here, using a model minielastin that mimics the alternating domain structure of full-length tropoelastin, we examine the elastin liquid phase. We combine differential interference contrast (DIC), fluorescence, and scanning electron microscopy with particle-tracking microrheology to resolve the material transition occurring within elastin liquids over time in the absence of exogenous cross-linking. We find that this transition is accompanied by an intermediate stage marked by the coexistence of insoluble solid and dynamic liquid phases giving rise to significant spatial heterogeneities in material properties. We further demonstrate that varying the length of the terminal hydrophobic domains of minielastins can tune the maturation process. This work not only resolves an important step in the hierarchical assembly process of elastogenesis but further contributes mechanistic insight into the diverse repertoire of protein condensate maturation pathways with emerging importance across biology.

Liquid-Liquid Phase-Separated Systems from Reversible Gel-Sol Transition of Protein Microgels.

Project description:Liquid-liquid phase-separated biomolecular systems are increasingly recognized as key components in the intracellular milieu where they provide spatial organization to the cytoplasm and the nucleoplasm. The widespread use of phase-separated systems by nature has given rise to the inspiration of engineering such functional systems in the laboratory. In particular, reversible gelation of liquid-liquid phase-separated systems could confer functional advantages to the generation of new soft materials. Such gelation processes of biomolecular condensates have been extensively studied due to their links with disease. However, the inverse process, the gel-sol transition, has been less explored. Here, a thermoresponsive gel-sol transition of an extracellular protein in microgel form is explored, resulting in an all-aqueous liquid-liquid phase-separated system with high homogeneity. During this gel-sol transition, elongated gelatin microgels are demonstrated to be converted to a spherical geometry due to interfacial tension becoming the dominant energetic contribution as elasticity diminishes. The phase-separated system is further explored with respect to the diffusion of small particles for drug-release scenarios. Together, this all-aqueous system opens up a route toward size-tunable and monodisperse synthetic biomolecular condensates and controlled liquid-liquid interfaces, offering possibilities for applications in bioengineering and biomedicine.

Maximum Entropy-Mediated Liquid-to-Solid Nucleation and Transition.

Project description:Molecular dynamics (MD) simulations are a powerful tool for studying matter at the atomic scale. However, to simulate solids, an initial atomic structure is crucial for the successful execution of MD simulations but can be difficult to prepare due to insufficient atomistic information. At the same time, wide-angle X-ray scattering (WAXS) measurements can determine the radial distribution function (RDF) of atomic structures. However, the interpretation of RDFs is often challenging. Here, we present an algorithm that can bias MD simulations with RDFs by combining the information on the MD atomic interaction potential and the RDF under the principle of maximum relative entropy. We show that this algorithm can be used to adjust the RDF of one liquid model, e.g., the TIP3P water model, to reproduce the RDF and improve the angular distribution function (ADF) of another model, such as the TIP4P/2005 water model. In addition, we demonstrate that the algorithm can initiate crystallization in liquid systems, leading to both stable and metastable crystalline states defined by the RDF, e.g., crystallization of water to ice and liquid TiO2 to rutile or anatase. Finally, we discuss how this method can be useful for improving interaction models, studying crystallization processes, interpreting measured RDFs, or training machine-learned potentials.

Liquid-to-gel transition for visual and tactile detection of biological analytes.

Project description:So far all visual and instrument-free methods have been based on a color change. However, colorimetric assays cannot be used by blind or color-blind people. Here we introduce a liquid-to-gel transition as a general output platform. The signal output (a piece of gel) can be unambiguously distinguished from liquid both visually and by touch. This approach promises to contribute to the development of an accessible environment for visually impaired persons.

Shear-mediated sol-gel transition of regenerated silk allows the formation of Janus-like microgels.

Project description:Microcapsules and microgels consisting of macromolecular networks have received increasing attention due to their biomedical and pharmaceutical applications. Protein microgels and in particular silk-based microcapsules have desirable properties due to their biocompatibility and lack of toxicity. Typically such structures formed through emulsion templating are spherical in geometry due to interfacial tension. However, approaches to synthesis particles with more complex and non-spherical geometries are sought due to their packing properties and cargo release characteristics. Here, we describe a droplet-microfluidic strategy for generating asymmetric tubular-like microgels from reconstituted silk fibroin; a major component of native silk. It was determined using fluorescence microscopy, that the shear stress within the microchannel promotes surface protein aggregation, resulting in the asymmetric morphology of the microgels. Moreover, the structural transition that the protein undergoes was confirmed using FTIR. Crucially, the core of the microgels remains liquid, while the surface has fully aggregated into a fibrillar network. Additionally, we show that microgel morphology could be controlled by varying the dispersed to continuous phase flow rates, while it was determined that the radius of curvature of the asymmetric microgels is correlated to the wall shear stress. By comparing the surface fluorescence intensity of the microgels as a function of radius of curvature, the effect of the shear stress on the amount of aggregation could be quantified. Finally, the potential use of these asymmetric microgels as carriers of cargo molecules is showcased. As the core of the microgel remains liquid but the shell has gelled, this approach is highly suitable for the storage of bio-active cargo molecules such as antibodies, making such a delivery system attractive in the context of biomedical and pharmaceutical applications.

Tunable liquid-solid hybrid thermal metamaterials with a topology transition.

Project description:Thermal metamaterials provide rich control of heat transport which is becoming the foundation of cutting-edge applications ranging from chip cooling to biomedical. However, due to the fundamental laws of physics, the manipulation of heat is much more constrained in conventional thermal metamaterials where effective heat conduction with Onsager reciprocity dominates. Here, through the inclusion of thermal convection and breaking the Onsager reciprocity, we unveil a regime in thermal metamaterials and transformation thermotics that goes beyond effective heat conduction. By designing a liquid-solid hybrid thermal metamaterial, we demonstrate a continuous switch from thermal cloaking to thermal concentration in one device with external tuning. Underlying such a switch is a topology transition in the virtual space of the thermotic transformation which is achieved by tuning the liquid flow via external control. These findings illustrate the extraordinary heat transport in complex multicomponent thermal metamaterials and pave the way toward an unprecedented regime of heat manipulation.

Polycyclic Aromatic Hydrocarbons in Fine Particulate Matter Emitted from Burning Kerosene, Liquid Petroleum Gas, and Wood Fuels in Household Cookstoves.

Project description:This study measures polycyclic aromatic hydrocarbon (PAH) compositions in particulate matter emissions from residential cookstoves. A variety of fuel and cookstove combinations are investigated, including: (i) liquid petroleum gas (LPG), (ii) kerosene in a wick stove, (iii) wood (10 and 30% moisture content on a wet basis) in a forced-draft fan stove, and (iv) wood in a natural-draft rocket cookstove. The wood burning in the natural-draft stove had the highest PAH emissions followed by the wood combustion in the forced-draft stove and kerosene burning. LPG combustion has the highest thermal efficiency (∼57%) and the lowest PAH emissions per unit fuel energy, resulting in the lowest PAH emissions per useful energy delivered (in the unit of megajoule delivered, MJd). Compared with the wood combustion emissions, LPG burning also emits a lower fraction of higher molecular weight PAHs. In rural regions where LPG and kerosene are unavailable or unaffordable, the forced-draft fan stove is expected to be an alternative because its benzo[a]pyrene (B[a]P) emission factor (5.17-8.24 μg B[a]P/MJd) and emission rate (0.522-0.583 μg B[a]P/min) are similar to those of kerosene burning (5.36 μg B[a]P/MJd and 0.452 μg B[a]P/min). Relatively large PAH emission variability for LPG suggests a need for additional future tests to identify the major factors influencing these combustion emissions. These future tests should also account for different LPG fuel formulations and stove burner types.
