Project description:The growth of watermelon is often threatened by Fusarium oxysporum f. sp. niveum (Fon) in successively monocultured soil, which results in economic loss. The objective of this study was to investigate the effect of D123 wheat as a companion crop on soil enzyme activities, microbial biomass and microbial communities in the rhizosphere of watermelon and to explore the relationship between the effect and the incidence of wilt caused by Fon. The results showed that the activities of soil polyphenol oxidase, urease and invertase were increased, the microbial biomass nitrogen (MBN) and microbial biomass phosphorus (MBP) were significantly increased, and the ratio of MBC/MBN was decreased (P < 0.05). Real-time PCR analysis showed that the Fon population declined significantly in the watermelon/wheat companion system compared with the monoculture system (P < 0.05). The analysis of microbial communities showed that the relative abundance of microbial communities was changed in the rhizosphere of watermelon. Compared with the monoculture system, the relative abundances of Alphaproteobacteria, Actinobacteria, Gemmatimonadetes and Sordariomycetes were increased, and the relative abundances of Gammaproteobacteria, Sphingobacteria, Cytophagia, Pezizomycetes, and Eurotiomycetes were decreased in the rhizosphere of watermelon in the watermelon/wheat companion system; importantly, the incidence of Fusarium wilt was also decreased in the watermelon/wheat companion system. In conclusion, this study indicated that D123 wheat as a companion crop increased soil enzyme activities and microbial biomass, decreased the Fon population, and changed the relative abundance of microbial communities in the rhizosphere of watermelon, which may be related to the reduction of Fusarium wilt in the watermelon/wheat companion system.

Project description:The production of American ginseng (Panax quinquefolius L.) is severely limited by the replant disorders in China. Crop rotation with maize might reduce the replant problems, but little information is available on the effect of maize rotation on soil cultivated with ginseng. In this study, we analyzed nutrients, phenolic acids, and microbial communities in soils from the fields with continuous maize, mono-culture ginseng, and 1-, 3-, and 5-year maize rotation after ginseng. Pot experiments were also conducted to evaluate the performance of replanting ginseng in these soils. The results showed that Mn, Cu, and 5 phenolic acids in ginseng-cultivated soil were significantly decreased by maize rotation. A 5-year maize rotation significantly increased the relative abundance of beneficial soil bacteria, such as Arthrobacter, rather than decreasing the abundances of potential pathogenic genera. Clustering analysis revealed that the physicochemical properties and microbial communities of 3- and 5-year maize rotation soil were more similar to CM than to G soil. The biomass of replanted ginseng root was improved, and root disease was reduced over 3 years of maize rotation. Overall, the results showed that at least a 3-year maize rotation is needed to overcome the replant failure of American ginseng.

Project description:Irrigation and nitrogen (N) fertilization rates are widely used to increase crop growth and yield and promote the sustainable production of the maize crop. However, our understanding of irrigation and N fertilization in the soil microenvironment is still evolving, and further research on soil bacterial communities under maize crop with irrigation and N management in subtropical regions of China is needed. Therefore, we evaluated the responses of two irrigation levels (low and high irrigation water with 60 and 80% field capacity, respectively) and five N fertilization rates [i.e., control (N0), N200 (200 kg N ha-1), N250 (250 kg N ha-1), N300 (300 kg N ha-1), and N350 (350 kg N ha-1)] on soil bacterial communities, richness, and diversity. We found that both irrigation and N fertilization significantly affected bacterial richness, diversity index, and number of sequences. Low irrigation with N300 treatment has significantly higher soil enzymes activities, soil nutrient content, and bacterial alpha and beta diversity than high irrigation. In addition, the Proteobacteria, Actinobacteriota, Chloroflexi, and Firmicutes were the dominant bacterial phyla under both irrigation regimes. The acidic phosphates, acidic invertase, β-glucosidase, catalase, cellulase, and urease were positively correlated with the Shannon index under both low and high irrigation. Therefore, low irrigation improves soil nutrient utilization by boosting soil enzyme activity, directly affecting soil bacterial communities. It was concluded that greater soil nutrients, enzyme activities with higher bacterial diversity are the main indicators of soil reactivity to low irrigation water and N300 for maintaining soil fertility and soil microbial community balance.

Project description:BackgroundCrop rotation is an important agricultural practice that often affects the metabolic processes of soil microorganisms through the composition and combination of crops, thereby altering nutrient cycling and supply to the soil. Although the benefits of crop rotation have been extensively discussed, the effects and mechanisms of different crop combinations on the soil microbial community structure in specific environments still need to be analyzed in detail.Materials and methodsIn this study, six crop rotation systems were selected, for which the spring crops were mainly tobacco or gramineous crops: AT (asparagus lettuce and tobacco rotation), BT (broad bean and tobacco rotation), OT (oilseed rape and tobacco rotation), AM (asparagus lettuce and maize rotation), BM (broad bean and maize rotation), and OR (oilseed rape and rice rotation). All crops had been cultivated for > 10 years. Soil samples were collected when the rotation was completed in spring, after which the soil properties, composition, and functions of bacterial and fungal communities were analyzed.ResultsThe results indicate that spring cultivated crops play a more dominant role in the crop rotation systems than do autumn cultivated crops. Crop rotation systems with the same spring crops have similar soil properties and microbial community compositions. pH and AK are the most important factors driving microbial community changes, and bacteria are more sensitive to environmental responses than fungi. Rotation using tobacco systems led to soil acidification and a decrease in microbial diversity, while the number of biomarkers and taxonomic indicator species differed between rotation patterns. Symbiotic network analysis revealed that the network complexity of OT and BM was the highest, and that the network density of tobacco systems was lower than that of gramineous systems.ConclusionsDifferent crop rotation combinations influence both soil microbial communities and soil nutrient conditions. The spring crops in the crop rotation systems had stronger dominating effects, and the soil bacteria were more sensitive than the fungi were to environmental changes. The tobacco rotation system can cause soil acidification and thereby affect soil sustainability, while the complexity of soil microbial networks is lower than that of gramineous systems. These results provide a reference for future sustainable applications of rotation crop systems.

Project description:The effect of crop rotation on soil-borne diseases is a representative case of plant-soil feedback in the sense that plant disease resistance is influenced by soils with different cultivation histories. This study examined the microbial mechanisms inducing the differences in the clubroot (caused by Plasmodiophora brassicae pathogen) damage of Chinese cabbage (Brassica rapa subsp. pekinensis) after the cultivation of different preceding crops. It addresses two key questions in crop rotation: changes in the soil bacterial community induced by the cultivation of different plants and the microbial mechanisms responsible for the disease-suppressive capacity of Chinese cabbage. Twenty preceding crops from different plant families showed significant differences in the disease damage, pathogen density, and bacterial community composition of the host plant. Structural equation modelling revealed that the relative abundance of four key bacterial orders in Chinese cabbage roots can explain 85% and 70% of the total variation in pathogen density and disease damage, respectively. Notably, the relative dominance of Bacillales and Rhizobiales, which have a trade-off relationship, exhibited predominant effects on pathogen density and disease damage. The disease-suppressive soil legacy effects of preceding crops are reflected in compositional changes in key bacterial orders, which are intensified by the bacterial community network.

Project description:As essential approaches for conservation agricultural practices, straw residue retention and crop rotation have been widely used in the Mollisols of Northeast China. Soil organic carbon, root development and microbial community are important indicators representing soil, crop and microbiota, respectively, and these factors work together to influence soil fertility and crop productivity. Studying their changes and interactions under different conservation practices is crucial to provide a theoretical basis for developing rational agricultural practices. The experiment in this study was conducted using the conventional practice (continuous maize without straw retention, C) and three conservation practices, namely, continuous maize with straw mulching (CS), maize-peanut rotation (R), and maize-peanut rotation with straw mulching (RS). Straw mulching (CS) significantly increased soil total organic carbon (TOC), active organic carbon (AOC), and microbial biomass carbon (MBC), but did not promote maize yield. Maize-peanut rotation (R and RS) significantly increased dissolved organic carbon (DOC) in the rhizosphere by promoting root growth, and maize yield (increased by 10.2%). For the microbial community structure, PERMANOVA and PCoA indicated that the bacterial community differed significantly between rhizosphere soil and bulk soil, but the fungal community shifted more under different agricultural practices. The correlation analysis indicated that the rotation system promoted the association between the soil DOC and the microbial community (especially the bacterial community), and straw mulching enhanced the connection between the soil TOC and the fungal community. Some plant growth-promoting rhizobacteria (including Bacillus, Streptomyces, Rhizobium, and Pseudomonas) were enriched in the rhizosphere soil and were increased in the rotation system (R and RS), which might be due to an increase in the soil rhizosphere DOC level. These beneficial microbes had significantly negative correlations with several fungal groups (such as Mycosphaerella, Penicillium, Paraphoma and Torula) that were classified as plant pathotrophs by FUNGuild. These results indicated that ensuring plant root development and improving root-bacteria interactions are of great importance to guarantee crop yield when implementing conservation tillage practices.

Project description:Organic fertilizer is effective in improving soil quality, and promoting crop growth. Combined organic and inorganic fertilization has been proved as a more favorable way to tobacco yield and quality. However, the mechanisms underlying tobacco yield and quality under combinations of different organic and inorganic fertilizer remain unclear. We conducted a 12-year tobacco (Nicotiana tabacum L.)-maize crop rotation field experiment in Yanhe experimental station, China to examine the yields and qualities of tobacco, soil nutrients, and extracellular enzyme activities associated with carbon, nitrogen, and phosphorus cycles in response to different fertilization treatments. Five fertilization treatments (no fertilization; 75 kg N fertilizer ha-1; 450 kg oil cake ha-1 + 75 kg N fertilizer ha-1; 15,000 kg pig dung ha-1 + 60 kg N fertilizer ha-1; 3,000 kg straw ha-1 + 75 kg N fertilizer ha-1) were applied to tobacco while maize was fertilized with inorganic compound fertilizers. After 12 years of tobacco-maize rotation, the results showed that organic fertilizer additions elevate tobacco yield and quality, and the soil extracellular enzymes activities. Gram-negative bacteria, actinomycetes, and total soil microbial biomass were increased by organic fertilizer additions, both plant-based (oil cake and straw) and animal-based (pig dung) organics. The levels of soil organic matter, total organic carbon, total phosphorus and available phosphorus are higher in pig dung addition treatment than oil cake and straw additions. By variance analysis with respect to fertilization treatments, organic sources differentially affected the activities of diverse soil enzymes. The redundancy analysis gave that yield and quality of tobacco leaves (upper, middle, and lower leaves) positively related to soil extracellular enzyme activities. Based on analysis of yield and quality of tobacco leaves with extracellular enzyme activities and soil nutrients, it is suggested animal-based organic fertilizer, thus pig dung, should be used in combining with chemical fertilizers to improve the quality of tobacco and soil nutrients.

Project description:Nitrifying microorganisms play an important role in nitrogen (N) cycling in agricultural soils as nitrification leads to accumulation of nitrate (NO3-) that is readily lost through leaching and denitrification, particularly in high rainfall regions. Legume crop rotation in sugarcane farming systems can suppress soil pathogens and improve soil health, but its effects on soil nitrifying microorganisms are not well understood. Using shotgun metagenomic sequencing, we investigated the impact of two legume break crops, peanut (Arachis hypogaea) and soybean (Glycine max), on the nitrifying communities in a sugarcane cropping soil. Cropping with either legume substantially increased abundances of soil bacteria and archaea and altered the microbial community composition, but did not significantly alter species richness and evenness relative to a bare fallow treatment. The ammonia oxidisers were mostly archaeal rather than bacterial, and were 24-44% less abundant in the legume cropping soils compared to the bare fallow. Furthermore, abundances of the archaeal amoA gene encoding ammonia monooxygenase in the soybean and peanut cropping soils were only 30-35% of that in the bare fallow. These results warrant further investigation into the mechanisms driving responses of ammonia oxidising communities and their nitrification capacity in soil during legume cropping.

Project description:Cover crops and compost are organic matter inputs that can impact soil health in tillage-intensive, high-input, organic vegetable production systems in the central coast region of California. Data are presented on soil microbial biomass (carbon and nitrogen) and soil enzymes (?-glucosidase, ?-glucosaminidase, alkaline phosphatase, aspartase and L-asparaginase and dehydrogenase) from a relatively long-term organic systems experiment in Salinas, California that was focused on lettuce and broccoli production and included eight different certified organic systems. These systems differed in compost inputs, cover cropping frequency, cover crop type, and cover cropping seeding rate. The compost was made from urban yard waste, and the cover crops included rye, a legume-rye mixture, and a mustard mixture planted at two seeding rates (standard rate 1× versus high rate 3×). There were three legume-rye 3× systems that differed in compost inputs (0 versus 15?Mg?ha-1 year-1 and cover cropping frequency (every winter versus every fourth winter). The data in this article support and augment information presented in the research articles "Cover cropping frequency is the main driver of soil microbial changes during six years of organic vegetable production" (Brennan and Acosta-Martinez, 2017) and "Cover crops and compost influence soil enzymes during 6 years of tillage-intensive, organic vegetable production" (Brennan and Acosta-Martinez, 2018).

Project description:Background and Aims Cotton-peanut rotation is a sustainable farming practice that enhances land utilization and promotes the sustainable development of agriculture. Crop rotation can reduce the occurrence of pests and diseases, as different crops have varying levels of resistance to such threats. Additionally, by alternating the types of crops grown, the soil environment is changed, which can lead to the elimination of favorable conditions for pathogens and pests, thereby alleviating the impact of these issues. Furthermore, cotton-peanut rotation can improve soil fertility.To investigate the effects of different crop rotation systems on crop yield, soil nutrients, and soil microbial communities.MethodsUsing high-throughput sequencing technology, investigate the soil microbial diversity in the root zone after cotton-peanut rotation.Various planting patterns, including cotton continuous cropping (MC), peanut continuous cropping (HC), peanut-cotton-peanut rotation (HR), and fallow (X), were established to assess variations in crop yield, soil nutrients, and soil microbial diversity.ResultsSignificant differences were observed in crop yield, soil nutrients, and soil microbial community structure among different planting patterns. The HR system significantly increased the output compared with the HC and MC systems. Additionally, HR exhibited significantly lower total nitrogen (N) and basic nitrogen (BN) contents than HC and MC, whereas MC showed lower total potassium (K) and available potassium (AK) contents. HR led to a decrease in soil bacterial diversity but an increase in fungal diversity, with Ascomycota and Mortierellomycota being dominant. Various bacteria (Chloroflexi, Bacteroidota, and Actinobacteriota) associated with organic matter degradation and nutrient cycling were found across different planting systems, enhancing material cycling efficiency. Furthermore, Planctomycetota bacteria related to crop nutrient synthesis and Glomeromycota bacteria aiding plant nutrient absorption were significantly higher in the MC system than in the HR or HC systems. Redundancy analysis indicated a significant negative correlation between crop rotation and soil fungal community, whereas Ascomycota exhibited a significant negative correlation with organic matter.ConclusionPeanut-cotton rotation can mitigate soil nutrient loss, enhance beneficial microorganism diversity, suppress harmful bacterial populations, stabilize ecosystems, and boost crop yield.
