Effect of Butyl Paraben on Oxidative Stress in the Liver of Mauremys sinensis.

ABSTRACT: Butyl paraben (BuP) has been widely used as a preservative in the cosmetics, food, and medicine industries. Recently, it has become a new pollutant and has attracted much attention. In order to evaluate the toxic effect of BuP on aquatic animals, Chinese striped-neck turtles (Mauremys sinensis) were exposed to BuP solutions with different concentrations of 0, 5, 50, 500, and 5000 µg/L for 20 weeks. The results showed that with an increase in BuP concentration, the activity of antioxidant enzymes (SOD, CAT and GSH-PX) in liver decreased. The expression of key genes in the Nrf2-Keap1 signal pathways first increased and then decreased, while the expression of the HSP70 and HSP90 genes increased. In addition, the liver had an inflammatory reaction. The expression of the BAFF and IL-6 genes increased and then decreased with an increase in BuP concentration, while the expression of P50 and P65 increased significantly. Oxidative stress induced apoptosis, and the expression of pro-apoptosis genes (BAX, cytc, Caspase3 and Caspase9) increased, while the expression of the anti-apoptosis gene Bcl2 decreased. The results provide an important reference for the comprehensive ecological and health risk assessment of environmental BuP.

Project description:Among neonicotinoid insecticides, thiacloprid (THI) is extensively utilized in agricultural practices, which poses a potential toxicity risk to aquatic fauna. Turtles, integral to aquatic ecosystems, have not yet been comprehensively assessed for their vulnerability to THI exposure. In this study, we aimed to evaluate the effects of THI on oxidative stress, endoplasmic reticulum stress (ERS), and apoptosis in aquatic turtles. We categorized Mauremys reevesii into three groups: a control group and two experimental groups exposed to environmentally relevant (4.5 μg/mL) and high (15 mg/mL) concentrations of THI, respectively. Transcriptome analysis revealed that genes significantly associated with the elimination of superoxide radicals, organelle inner membrane functions, peroxiredoxin activity, and apoptotic pathways were abundantly expressed in the high-concentration THI group. Notably, exposure to high concentrations of THI led to a marked increase in glutathione peroxidase (GPX) and superoxide dismutase (SOD) activities, whereas catalase (CAT) activity declined and malondialdehyde (MDA) levels rose, indicating the presence of oxidative stress. Moreover, THI upregulated the expression of the ER stress marker GRP78. Simultaneously, the mRNA levels of pivotal unfolded protein response genes, including AFT6, AFT4, IRE1α, CHOP, XBP1, and eIF2α, were significantly elevated in response to THI exposure. Furthermore, high concentrations of THI significantly activated the activities of caspase-3, caspase-8, and caspase-9 enzymes in the liver tissue. The expression of anti-apoptotic gene Bcl-2 was downregulated, whereas the pro-apoptotic genes Bax and caspase-3 were upregulated, leading to an increase in hepatic apoptotic cells following THI exposure. Collectively, our study indicates that THI can induce hepatic damage in turtles through the promotion of oxidative stress, ERS, and apoptosis. These findings gain a deeper understanding of the toxic effects of THI on keystone species in aquatic ecosystems, thereby improving our overall understanding of their environmental impacts.

Project description:Phenols and parabens are used in a multitude of consumer products resulting in ubiquitous human exposure. Animal and in vitro studies suggest that exposure to these compounds may be related to a number of adverse health outcomes, as well as potential mediators such as oxidative stress and inflammation. We examined urinary phenol (bisphenol A (BPA), triclosan (TCS), benzophenone-3 (BP-3), 2,4-dichlorophenol (24-DCP), 2,5-dichlorophenol (25-DCP)) and paraben (butyl paraben (B-PB), methyl paraben (M-PB), propyl paraben (P-PB)) concentrations measured three times during pregnancy in relation to markers of oxidative stress and inflammation among participants in the Puerto Rico Testsite for Exploring Contamination Threats (PROTECT) project. Serum markers of inflammation (c-reactive protein (CRP), IL-1β, IL-6, IL-10, and tumor necrosis factor-α (TNF-α)) were measured twice during pregnancy (n=105 subjects, 187 measurements) and urinary markers of oxidative stress (8-hydroxydeoxyguanosine (OHdG) and isoprostane) were measured three times during pregnancy (n=54 subjects, 146 measurements). We used linear mixed models to assess relationships between natural log-transformed exposure and outcome biomarkers while accounting for within individual correlation across study visits. After adjustment for urinary specific gravity, study visit, maternal pre-pregnancy BMI, and maternal education, an interquartile range (IQR) increase in urinary BPA was associated with 21% higher OHdG (p=0.001) and 29% higher isoprostane (p=0.0002), indicating increased oxidative stress. The adjusted increase in isoprostane per IQR increase in marker of exposure was 17% for BP-3, 27% for B-PB, and 20% for P-PB (all p<0.05). An IQR increase in triclosan (TCS) was associated with 31% higher serum concentrations of IL-6 (p=0.007), a pro-inflammatory cytokine. In contrast, IQR increases in BP-3 and B-PB were significantly associated with 16% and 18% lower CRP, a measure of systemic inflammation. Our findings suggest that exposure to BPA, select parabens, and TCS during pregnancy may be related to oxidative stress and inflammation, potential mechanisms by which exposure to these compounds may influence birth outcomes and other adverse health effects, but additional research is needed.
