Evolutionary Insight into the Association between New Jersey Polyomavirus and Humans.

ABSTRACT: Advances in viral discovery techniques have led to the identification of numerous novel viruses in human samples. However, the low prevalence of certain viruses in humans raises doubts about their association with our species. To ascertain the authenticity of a virus as a genuine human-infecting agent, it can be useful to investigate the diversification of its lineage within hominines, the group encompassing humans and African great apes. Building upon this rationale, we examined the case of the New Jersey polyomavirus (NJPyV; Alphapolyomavirus terdecihominis), which has only been detected in a single patient thus far. In this study, we obtained and analyzed sequences from closely related viruses infecting all African great ape species. We show that NJPyV nests within the diversity of these viruses and that its lineage placement is compatible with an ancient origin in humans, despite its apparent rarity in human populations.

Project description:Although data about COVID-19 cases and deaths in the United States are readily available at the county-level, datasets on smaller geographic areas are limited. County-level data have been used to identify geospatial patterns of COVID-19 spread and, in conjunction with sociodemographic variables, have helped identify population health disparities concerning COVID-19 in the US. Municipality-level data are essential for advancing more targeted and nuanced understanding of geographic-based risk and resilience associated with COVID-19. We created a dataset that tracks COVID-19 cases and deaths by municipalities in the state of New Jersey (NJ), US, from April 22, 2020 to December 31, 2020. Data were drawn primarily from official county and municipality websites. The dataset is a spreadsheet containing cumulative case counts and case rates in each municipaly on three target dates, representing the peak of the first wave, the summer trough after the first wave, and the outbreak of the second wave in NJ. This dataset is valuable for four main reasons. First, the dataset is unique, because New Jersey's Health Department does not release COVID-19 data for the 77% (433/565) of municipalities with populations smaller than 20,000 individuals. Second, especially when combined with other data sources, such as publicly available sociodemographic data, this dataset can be used to advance epidemiological research on geographic differences in COVID-19, as well as to inform decision-making concerning the allocation of resources in response to the pandemic (e.g., strategies for targeted vaccine outreach campaigns). Third, county-level data mask important variations across municipalities, so municipality-level data permit a more nuanced exploration of health disparities related to local demographics, socioeconomic conditions, and access to resources and services. New Jersey is a good state to explore these patterns, because it is the most densely-populated and racially/ethnically diverse state in the US. Fourth, New Jersey was one of the few locations in the US with a high prevalence of COVID-19 during the first wave of the pandemic in the US. Thus, this dataset permits exploration of whether sociodemographic variables predicted COVID-19 differently as time progressed. To summarize, this unique municipality-level dataset in a diverse state with high COVID-19 cases is valuable for scholars and policy analysts to explore social and environmental factors related to the prevalence and transmission of COVID-19 in the US.

Project description:Importance:Use of electronic cigarettes (e-cigarettes) is high among adolescents, but the extent to which the JUUL e-cigarette brand accounts for the high prevalence has not been explored using population-based surveys. Objective:To examine e-cigarette and JUUL use among adolescents in New Jersey. Design, Setting, and Participants:Survey study using data from the 2018 New Jersey Youth Tobacco Survey, a cross-sectional statewide representative survey of tobacco use. The survey was school based and sampled New Jersey students in grades 9 to 12. Exposures:Use of tobacco products; JUUL as first tobacco product tried; exposure to JUUL at school; number of friends perceived as JUUL users; liking or following a tobacco brand on social media; and buying or receiving tobacco-branded merchandise. Main Outcomes and Measures:Prevalence ratio (PR) for current and frequent e-cigarette use, inclusive of JUUL. Results:In this sample of 4183 adolescents, respondents were 49.6% female and 49.6% non-Hispanic white. Students were evenly distributed across grades 9 through 12. Overall, the estimate for current use of e-cigarettes inclusive of JUUL was higher (24.2%; 95% CI, 22.5%-25.9%) compared with current use assessed by use of e-cigarettes only (17.8%; 95% CI, 16.4%-19.4%) or JUUL use only (21.3%; 95% CI, 19.7%-23.0%). Divergence in e-cigarette use estimates was higher for certain subgroups, including female respondents and non-Hispanic black respondents. Also, 88.8% (95% CI, 86.6%-91.1%) of current e-cigarette users reported JUUL as a brand they used. Hispanic students (PR, 0.78; 95% CI, 0.69-0.89) and non-Hispanic students of other races (PR, 0.64; 95% CI, 0.51-0.81) were significantly less likely than non-Hispanic white students to be current e-cigarette users, and students in 12th grade were more likely than those in 9th grade to be current users (PR, 1.29; 95% CI, 1.11-1.48). Current e-cigarette use was positively associated with current use of other tobacco products (PR, 2.57; 95% CI, 2.24-2.95), endorsing a tobacco brand on social media (PR, 1.43; 95% CI, 1.19-1.72), having tobacco-branded merchandise (PR, 1.70; 95% CI, 1.46-1.97), having close friends who used JUUL (PR, 3.81; 95% CI, 3.17-4.58), and seeing JUUL used on school grounds (PR, 1.43; 95% CI, 1.24-1.65). Estimates of prevalence were greater when modeling frequent use. Conclusions and Relevance:This study found that prevalence of current and frequent e-cigarette use among adolescents was higher when inclusive of JUUL use, and JUUL was by far the most common e-cigarette brand used, providing support for inclusion of brand-specific questions when assessing e-cigarette use. The results also identify characteristics of adolescents who may be more likely to use e-cigarettes.
