Uranium Biogeochemistry in the Rhizosphere of a Contaminated Wetland.

Project description:The objective of this study was to determine if U sediment concentrations in a U-contaminated wetland located within the Savannah River Site, South Carolina, were greater in the rhizosphere than in the nonrhizosphere. U concentrations were as much as 1100% greater in the rhizosphere than in the nonrhizosphere fractions; however and importantly, not all paired samples followed this trend. Iron (but not C, N, or S) concentrations were significantly enriched in the rhizosphere. XAS analyses showed that in both sediment fractions, U existed as UO22+ coordinated with iron(III)-oxides and organic matter. A key difference between the two sediment fractions was that a larger proportion of U was adsorbed to Fe(III)-oxides, not organic matter, in the rhizosphere, where significantly greater total Fe concentrations and greater proportions of ferrihydrite and goethite existed. Based on 16S rRNA analyses, most bacterial sequences in both paired samples were heterotrophs, and population differences were consistent with the generally more oxidizing conditions in the rhizosphere. Finally, U was very strongly bound to the whole (unfractionated) sediments, with an average desorption Kd value (Usediment/Uaqueous) of 3972 ± 1370 (mg-U/kg)/(mg-U/L). Together, these results indicate that the rhizosphere can greatly enrich U especially in wetland areas, where roots promote the formation of reactive Fe(III)-oxides.

Collembola growth in heavy metal-contaminated soils.

Project description:Collembola play a key role in soil ecosystems by decomposing organic matter. Most of them inhabit the upper layers of the soil and are susceptible to contamination present in the pore water. These arthropods serve as model organisms in ecotoxicology for short and long-term exposure. This study aimed to assess the toxicity of three heavy metals (lead [Pb], cadmium [Cd], and copper [Cu]) using the springtail species Folsomia candida as the test organism, with mortality and growth inhibition as the measure of toxicity. We hypothesised that increasing metal concentrations in the soil would correspond to a growth reduction of Collembola. Each heavy metal was tested at a minimum of eight increasing concentrations in six replications. Twenty 10-12-day-old individuals were introduced into each test container filled with contaminated or control soil and incubated for 14 days. The test endpoints included growth inhibition determined by comparing F. candida growth rates in contaminated soil with those in control soil, as well as mortality rates. The EC50 values (mg/kg) for heavy metals were as follows: Cd = 66.89, Cu = 791.01, Pb = 10075.48. Our findings suggest that growth inhibition is a reliable indicator of Collembola toxicity to heavy metals.

Biodiversity of Actinomycetes from Heavy Metal Contaminated Technosols.

Project description:Technosols are artificial soils generated by diverse human activities and frequently contain toxic substances resulting from industrial processes. Due to lack of nutrients and extreme physico-chemical properties, they represent environments with limited bacterial colonization. Bacterial populations of technosols are dominated usually by Actinobacteria, including streptomycetes, known as a tremendous source of biotechnologically important molecules. In this study, the biodiversity of streptomycete-like isolates from several technosols, mainly mine soils and wastes (landfills and sludge) in Slovakia, was investigated. The combination of basic morphological and biochemical characterisations, including heavy metal resistance determination, and molecular approaches based on 16S rRNA gene analysis were used for the identification of the bacterial strains. From nine isolates of Actinobacteria collected from different habitats, one was found to represent a new species within the Crossiella genus. Eight other isolates were assigned to the genus Streptomyces, of which at least one could represent a new bacterial species. Some isolates showed high resistance to Pb, Zn, Cu or Ni. The most tolerated metal was Pb. The results obtained in this study indicate that technosols are a prospective source of new actinomycete species resistant to heavy metals what underlines their bioremediation potential.

Abundance and diversity of Archaea in heavy-metal-contaminated soils.

Project description:The impact of heavy-metal contamination on archaean communities was studied in soils amended with sewage sludge contaminated with heavy metals to varying extents. Fluorescent in situ hybridization showed a decrease in the percentage of Archaea from 1.3% +/- 0.3% of 4', 6-diamidino-2-phenylindole-stained cells in untreated soil to below the detection limit in soils amended with heavy metals. A comparison of the archaean communities of the different plots by denaturing gradient gel electrophoresis revealed differences in the structure of the archaean communities in soils with increasing heavy-metal contamination. Analysis of cloned 16S ribosomal DNA showed close similarities to a unique and globally distributed lineage of the kingdom Crenarchaeota that is phylogenetically distinct from currently characterized crenarchaeotal species.
