Project description:Substantial evidence indicates that China's afforestation statistically contributed to the ecosystem services (ES) improvement. However, we found the potential challenges behind this improvement, especially in water-limited areas. We propose an attribution analysis method, which can assess the specific contribution of natural, human and cognition degree drivers to ES dynamics. The results found that the ratio of natural and human drivers in the area north of China's 400 mm precipitation isopleth is 2:7. This means local vegetation capacity has already exceeded water limitation, implying a conflict between nature and humans. However, the natural contribution in the area between 400 and 800 mm precipitation isopleth is negative, whereas the human contribution is 91%. This means this area has fragile natural conditions and needs more flexible policies. The ratio of natural and human drivers in the region south of 800 mm precipitation isopleth is 6:3, suggesting the ecological policies here can be maintained.

Project description:Finding new etiological components is of great interest in disease epidemiology. We consider time series version of invariant coordinate selection (tICS) as an exploratory tool in the search of hidden structures in the analysis of population-based registry data. Increasing cancer burden inspired us to consider a case study of age-stratified cervical cancer incidence in Finland between the years 1953 and 2014. The latent components, which we uncover using tICS, show that the etiology of cervical cancer is age dependent. This is in line with recent findings related to the epidemiology of cervical cancer. Furthermore, we are able to explain most of the variation of cervical cancer incidence in different age groups by using only two latent tICS components. The second tICS component, in particular, is interesting since it separates the age groups into three distinct clusters. The factor that separates the three clusters is the median age of menopause occurrence.

Project description:Exploration for superconductivity is one of the research frontiers in condensed matter physics. In strongly correlated electron systems, the emergence of superconductivity is often inhibited by the formation of a thermodynamically more stable magnetic/charge order. Thus, to develop the superconductivity as the thermodynamically most stable state, the free-energy balance between the superconductivity and the competing order has been controlled mainly by changing thermodynamic parameters, such as the physical/chemical pressure and carrier density. However, such a thermodynamic approach may not be the only way to materialize the superconductivity. We present a new kinetic approach to avoiding the competing order and thereby inducing persistent superconductivity. In the transition-metal dichalcogenide IrTe2 as an example, by using current pulse-based rapid cooling of up to ~107 K s-1, we successfully kinetically avoid a first-order phase transition to a competing charge order and uncover metastable superconductivity hidden behind. Because the electronic states at low temperatures depend on the history of thermal quenching, electric pulse applications enable nonvolatile and reversible switching of the metastable superconductivity, a unique advantage of the kinetic approach. Thus, our findings provide a new approach to developing and manipulating superconductivity beyond the framework of thermodynamics.

Project description:Recent studies indicate that Generative Pre-trained Transformer 4 with Vision (GPT-4V) outperforms human physicians in medical challenge tasks. However, these evaluations primarily focused on the accuracy of multi-choice questions alone. Our study extends the current scope by conducting a comprehensive analysis of GPT-4V's rationales of image comprehension, recall of medical knowledge, and step-by-step multimodal reasoning when solving New England Journal of Medicine (NEJM) Image Challenges - an imaging quiz designed to test the knowledge and diagnostic capabilities of medical professionals. Evaluation results confirmed that GPT-4V outperforms human physicians regarding multi-choice accuracy (88.0% vs. 77.0%, p=0.034). GPT-4V also performs well in cases where physicians incorrectly answer, with over 80% accuracy. However, we discovered that GPT-4V frequently presents flawed rationales in cases where it makes the correct final choices (27.3%), most prominent in image comprehension (21.6%). Regardless of GPT-4V's high accuracy in multi-choice questions, our findings emphasize the necessity for further in-depth evaluations of its rationales before integrating such models into clinical workflows.

Project description:We propose metabolic models for the haptophyte microalgae Tisochrysis lutea with different possible organic carbon excretion mechanisms. These models-based on the DRUM (Dynamic Reduction of Unbalanced Metabolism) methodology-are calibrated with an experiment of nitrogen starvation under day/night cycles, and then validated with nitrogen-limited chemostat culture under continuous light. We show that models including exopolysaccharide excretion offer a better prediction capability. It also gives an alternative mechanistic interpretation to the Droop model for nitrogen limitation, which can be understood as an accumulation of carbon storage during nitrogen stress, rather than the common belief of a nitrogen pool driving growth. Excretion of organic carbon limits its accumulation, which leads to a maximal C/N ratio (corresponding to the minimum Droop N/C quota). Although others phenomena-including metabolic regulations and dissipation of energy-are possibly at stake, excretion appears as a key component in our metabolic model, that we propose to include in the Droop model.

Project description:The patch-clamp technique is a powerful tool that allows for a long observation of transport protein activity in real time. Experimental traces of single-channel currents can be considered as a record of the channel's conformational switching related to its activation and gating. In this work, we present a mathematically simple method of patch-clamp data analysis that assesses the connectivity and occupancy of distinct conformational substates of the channel. The proposed approach appears to be a big step forward due to its possible applications in the determination of channel substates related to disease and in the analysis of drug-channel interactions on the level of repetitive sequences of channel conformations. This is especially important in cases when molecular dynamics docking is impossible and Markovian modeling requires ambiguous optimization tasks.

Project description:BackgroundRecent advancements in artificial intelligence, such as GPT-3.5 Turbo (OpenAI) and GPT-4, have demonstrated significant potential by achieving good scores on text-only United States Medical Licensing Examination (USMLE) exams and effectively answering questions from physicians. However, the ability of these models to interpret medical images remains underexplored.ObjectiveThis study aimed to comprehensively evaluate the performance, interpretability, and limitations of GPT-3.5 Turbo, GPT-4, and its successor, GPT-4 Vision (GPT-4V), specifically focusing on GPT-4V's newly introduced image-understanding feature. By assessing the models on medical licensing examination questions that require image interpretation, we sought to highlight the strengths and weaknesses of GPT-4V in handling complex multimodal clinical information, thereby exposing hidden flaws and providing insights into its readiness for integration into clinical settings.MethodsThis cross-sectional study tested GPT-4V, GPT-4, and ChatGPT-3.5 Turbo on a total of 227 multiple-choice questions with images from USMLE Step 1 (n=19), Step 2 clinical knowledge (n=14), Step 3 (n=18), the Diagnostic Radiology Qualifying Core Exam (DRQCE) (n=26), and AMBOSS question banks (n=150). AMBOSS provided expert-written hints and question difficulty levels. GPT-4V's accuracy was compared with 2 state-of-the-art large language models, GPT-3.5 Turbo and GPT-4. The quality of the explanations was evaluated by choosing human preference between an explanation by GPT-4V (without hint), an explanation by an expert, or a tie, using 3 qualitative metrics: comprehensive explanation, question information, and image interpretation. To better understand GPT-4V's explanation ability, we modified a patient case report to resemble a typical "curbside consultation" between physicians.ResultsFor questions with images, GPT-4V achieved an accuracy of 84.2%, 85.7%, 88.9%, and 73.1% in Step 1, Step 2 clinical knowledge, Step 3 of USMLE, and DRQCE, respectively. It outperformed GPT-3.5 Turbo (42.1%, 50%, 50%, 19.2%) and GPT-4 (63.2%, 64.3%, 66.7%, 26.9%). When GPT-4V answered correctly, its explanations were nearly as good as those provided by domain experts from AMBOSS. However, incorrect answers often had poor explanation quality: 18.2% (10/55) contained inaccurate text, 45.5% (25/55) had inference errors, and 76.3% (42/55) demonstrated image misunderstandings. With human expert assistance, GPT-4V reduced errors by an average of 40% (22/55). GPT-4V accuracy improved with hints, maintaining stable performance across difficulty levels, while medical student performance declined as difficulty increased. In a simulated curbside consultation scenario, GPT-4V required multiple specific prompts to interpret complex case data accurately.ConclusionsGPT-4V achieved high accuracy on multiple-choice questions with images, highlighting its potential in medical assessments. However, significant shortcomings were observed in the quality of explanations when questions were answered incorrectly, particularly in the interpretation of images, which could not be efficiently resolved through expert interaction. These findings reveal hidden flaws in the image interpretation capabilities of GPT-4V, underscoring the need for more comprehensive evaluations beyond multiple-choice questions before integrating GPT-4V into clinical settings.

Project description:Nature finds a way to leverage nanotextures to achieve desired functions. Recent advances in nanotechnologies endow fascinating multi-functionalities to nanotextures by modulating the nanopixel's height. But nanoscale height control is a daunting task involving chemical and/or physical processes. As a facile, cost-effective, and potentially scalable remedy, the nanoscale capillary force lithography (CFL) receives notable attention. The key enabler is optical pre-modification of photopolymer's characteristics via ultraviolet (UV) exposure. Still, the underlying physics of the nanoscale CFL is not well understood, and unexplained phenomena such as the "forbidden gap" in the nano capillary rise (unreachable height) abound. Due to the lack of large data, small length scales, and the absence of first principles, direct adoptions of machine learning or analytical approaches have been difficult. This paper proposes a hybrid intelligence approach in which both artificial and human intelligence coherently work together to unravel the hidden rules with small data. Our results show promising performance in identifying transparent, physics-retained rules of air diffusivity, dynamic viscosity, and surface tension, which collectively appear to explain the forbidden gap in the nanoscale CFL. This paper promotes synergistic collaborations of humans and AI for advancing nanotechnology and beyond.

Project description:BackgroundAfrica is an important watershed in the genetic history of domestic cattle, as two lineages of modern cattle, Bos taurus and B. indicus, form distinct admixed cattle populations. Despite the predominant B. indicus nuclear ancestry of African admixed cattle, B. indicus mitochondria have not been found on the continent. This discrepancy between the mitochondrial and nuclear genomes has been previously hypothesized to be driven by male-biased introgression of Asian B. indicus into ancestral African B. taurus. Given that this hypothesis mandates extreme demographic assumptions relying on random genetic drift, we propose a novel hypothesis of selection induced by mitonuclear incompatibility and assess these hypotheses with regard to the current genomic status of African admixed cattle.ResultsBy analyzing 494 mitochondrial and 235 nuclear genome sequences, we first confirmed the genotype discrepancy between mitochondrial and nuclear genome in African admixed cattle: the absence of B. indicus mitochondria and the predominant B. indicus autosomal ancestry. We applied approximate Bayesian computation (ABC) to assess the posterior probabilities of two selection hypotheses given this observation. The results of ABC indicated that the model assuming both male-biased B. indicus introgression and selection induced by mitonuclear incompatibility explains the current genomic discrepancy most accurately. Subsequently, we identified selection signatures at autosomal loci interacting with mitochondria that are responsible for integrity of the cellular respiration system. By contrast with B. indicus-enriched genome ancestry of African admixed cattle, local ancestries at these selection signatures were enriched with B. taurus alleles, concurring with the key expectation of selection induced by mitonuclear incompatibility.ConclusionsOur findings support the current genome status of African admixed cattle as a potential outcome of male-biased B. indicus introgression, where mitonuclear incompatibility exerted selection pressure against B. indicus mitochondria. This study provides a novel perspective on African cattle demography and supports the role of mitonuclear incompatibility in the hybridization of mammalian species.
