Complete mitochondrial genome of Striatobalanus tenuis Hoek, 1883 (Balanomorpha: Balanidae) and a novel molecular phylogeny within Cirripedia.

ABSTRACT: Barnacles are crustaceans that are critical model organisms in intertidal ecology and biofouling research. In this study, we present the first mitochondrial genome of Striatobalanus tenuis which is a circular molecule of 15,067 bp in length. Consistent with most barnacles, the mitochondrial genome of S. tenuis encodes 37 genes, including 13 PCGs, 22 tRNAs and 2 rRNAs. A novel insight into the phylogenetic analysis based on the nucleotide data of 13 PCGs showed that the S. tenuis clusters with Striatobalanus amaryllis (bootstrap value = 100) of the same genus, then groups with other Balanoidea species, the Chelonibiidae, Austrobalanidae and Tetraclitidae cluster together forming superfamily Coronuloidea. The result can help us to understand the novel classification within Balanomorpha.

Project description:The triangle barnacle Balanus trigonus Darwin, 1854, a cosmopolitan inhabitant of tropical and warm temperate seas, is a member of robust system for the study of evolutionary processes in the intertidal zone. The first mitochondrial genome of B. trigonus is presented. The complete mitochondrial genome of B. trigonus is a circular molecule of 15,560 bp, which encodes 13 protein-coding genes (PCGs), 2 rRNA genes, and 22 tRNA genes. In comparison within Sessilia, the arrangement of the mitochondrial genome of B. trigonus is more similar to Megabalanus spp. than the congener Balanus balanus, which share a same inversion of a large gene block (P-nd4L-nd4-H-nd5-F). Phylogenetic analysis based on mitochondrial PCGs reveals that B. trigonus clusters with Acasta Sulcata (BP = 100), then grouped with Megabalanus volcano and Megabalanus ajax with high support (BP = 90). In further, more data and research are needed to reveal the phylogeny within Cirripedia.

Articulation and growth of skeletal elements in balanid barnacles (Balanidae, Balanomorpha, Cirripedia).

Project description:The morphology and ultrastructure of the shells of two balanid species have been examined, paying special attention to the three types of boundaries between plates: (i) radii-parietes, (ii) alae-sheaths, and (iii) parietes-basal plate. At the carinal surfaces of the radii and at the rostral surfaces of the alae, there are series of crenulations with dendritic edges. The crenulations of the radius margins interlock with less prominent features of the opposing paries margins, whereas the surfaces of the longitudinal abutments opposing the ala margins are particularly smooth. The primary septa of the parietes also develop dendritic edges, which abut the internal surfaces of the primary tubes of the base plates. In all cases, there are chitino-proteinaceous organic membranes between the abutting structures. Our observations indicate that the very edges of the crenulations and the primary septa are permanently in contact with the organic membranes. We conclude that, when a new growth increment is going to be produced, the edges of both the crenulations and the primary septa pull the viscoelastic organic membranes locally, with the consequent formation of viscous fingers. For the abutting edges to grow, calcium carbonate must diffuse across the organic membranes, but it is not clear how growth of the organic membranes themselves is accomplished, in the absence of any cellular tissue.

The first mitochondrial genome of Megabalanus tintinnabulum (Sessilia: Balanidae) from China: phylogeny within Cirripedia based on mitochondrial genes.

Project description:Here we present the complete mitochondrial genome of Megabalanus tintinnabulum. The genome is 15,107 bp in length with a 67.35% AT content. It contains 13 protein-coding genes (PCGs), 2 rRNAs genes, and 22 tRNAs. Both rRNAs are encoded on the light strand, as in the other crustacean and barnacle mitochondrial genomes. Besides five tRNAs are encoded on the light strand (nad1, trnV, trnL1, trnC, trnQ, and trnK). Only one PCG is encoded on the light strand (nad1), whereas the other 12 PCGs are located on the heavy strand, which is consistent with M. ajax. Phylogenetic analysis based on mitochondrial PCGs shows that M. tintinnabulum is clustered with M. ajax into a branch (BP = 100), and the group with M. volcano with high support. This study contributes to further phylogenetic analysis within Cirripedia.

Complete mitochondrial genome of the European common barnacle Perforatus perforatus Bruguiere, 1789 (balanomorpha: balanidae).

Project description:This study is the first to sequence the complete mitochondrial genome (mitogenome) of Perforatus perforatus Bruguière, 1789 (Balanomorpha: Balanidae). The 15,536-bp long P. perforatus mitogenome contained a typical set of animal mitochondrial genes, along with one control region. The P. perforatus mitogenome had an inverted gene block (trnP-ND4L-ND4-trnH-ND5-trnF) between trnS(gct) and trnT. This inverted gene block had been detected six species in three subfamilies of the Balanidae family (Balaninae, Acastinae and Megabalaninae), but our results show that it is also present in Concavinae, in which P. perforatus is included. The phylogenetic tree based on the concatenated sequences of the 13 protein-coding genes and two rRNA genes showed that P. perforatus is closely associated with Acasta sulcate and Balanus trigonus within Balanidae.

Complete mitochondrial genome of the catophragmid barnacle Catomerus polymerus (Cirripedia, Thoracica, Balanomorpha, Catophragmidae).

Project description:The family Catophragmidae is one of the lower balanomorphs from traditional and recent multiple mitochondrial and nuclear markers molecular analysis. Here, we characterized the first mitogenome of the catophragmid barnacle Catomerus polymerus, which was 15,446 bp in length with a 68.3% AT content. The mitogenome had the typical pancrustacean gene arrangement, which was identical to the mitogenome configurations of the chthamalid Octomeris sp. and pachylasmatoid Eochionelasmus ohtai. On the mitogenomic tree, the catophragmid Catomerus polymerus formed an independent branch that was basal to the members of the superfamilies Tetraclitoidea and Balanoidea, which was inconsistent with previous findings.

Complete mitochondrial genome of Tetraclita squamosa squamosa (Sessilia: Tetraclitidae) from China and phylogeny within Cirripedia.

Project description:Here we present the complete mitochondrial genome of Tetraclita squamosa squamosa, which is 15,191 bp in length with 67.20% AT content. It contains 13 protein-coding genes, 2 ribosomal-RNA genes and 22 transfer-RNA genes. All PCGs except nad4l in T. squamosa squamosa start with ATN, and terminated with a complete stop codon, except nad3. Phylogenetic analysis based on mitochondrial PCGs shows that T. squamosa squamosa is clustered with T. serrata into a branch (BP = 100). Our result is consistent with previous reports that genus Tetraclita and family Tetraclitidae are not monophyletic. This study contributes to further phylogenetic analysis within Cirripedia.

The mitochondrial genome of Chthamalus malayensis (Sessilia: Chthamalidae) and its molecular phylogeny within Cirripedia.

Project description:Cirripedia is a lower crustacean that has an invaluable place in several aspects of intertidal ecology and anti-fouling research. In this study, we present the first mitochondrial genome of Chthamalus malayensis. The complete mitochondrial genome of C. malayensis is a circular molecule of 15,230 bp. In comparison to the pancrustacean ground pattern, the mitochondrial genome of C. malayensis has a deletion of the trnC gene. Phylogenetic analysis based on mitochondrial protein-coding genes showed that C. malayensis clusters with C. antennatus (BP = 98) and is grouped with C. challengeri, Octomeris sp. BKKC-2014, and Notochthamalus scabrosus. Further studies are needed to reveal the specific phylogenic relationships within Cirripedia.

The first mitochondrial genome of Tetraclita kuroshioensis (Crustacea: Sessilia) from China: insight into the phylogeny within Cirripedia.

Project description:We determined the first mitochondrial genome of Tetraclita kuroshioensis from China. The mitochondrial genome of T. kuroshioensis was found to be 15,175 bp in length and consisted of 13 protein-coding genes (PCGs), 22 tRNAs, and 2 rRNAs. The longest non-coding region was 425 bp in length. Phylogenetic analysis showed that T. kuroshioensis clustered with T. serrata and then clustered with T. squamosa squamosa with high bootstrap value (BP = 100). In the future, sequencing of additional mitochondrial genomes should provide additional insights into the deep phylogeny of Cirripedia.

Molecular phylogeny of selected dorid nudibranchs based on complete mitochondrial genome.

Project description:Dorid nudibranchs are a large group of mollusks with approximately 2,000 recorded species. Although agreement exists on the monophyletic nature of the dorid nudibranch group, the interfamily relationships of the suborder are subject to debate. Despite efforts to elucidate this issue using short molecular markers, the conclusiveness of the findings has been hindered by branching polytomy. Mitogenomes are known to be effective markers for use in phylogenetic investigations. In this study, eight mitogenomes of dorid nudibranchs were decoded and analyzed. Gene content and structure showed little change among species, reflecting the conserved mitogenomes of dorid nudibranchs. For most genes, the direction was typical for nudibranchs; nevertheless, tRNACys had an inverse direction in Cadlinidae species. Phylogenetic trees based on nucleotide and amino acid datasets revealed a relatively consistent pattern of interfamily relationships with little difference for positions of Phyllidiidae and Cadlinidae. Species of Cadlinidae were clustered together and did not form a clade with Chromododidae. Additionally, Goniodorididae was sister to Aegiridae, whereas Discodoridae was sister to Dorididae. This finding was supported by tree topology test based on mitogenome data. The results of the present study indicate that complete mitogenomes are promising markers for investigating interfamily relationships among dorid nudibranchs.

Complete mitochondrial genome sequence of Bekko Tombo Libellula angelina Selys, 1883 (Odonata: Libellulidae).

Project description:The dragonfly Libellula angelina Selys, 1883 (Odonata: Libellulidae) has been listed as a critically endangered species by the International Union for Conservation of Nature (IUCN) and is also an endangered insect in South Korea. We sequenced the whole genome (15,233 bp) of L. angelina species, which included a set of typical genes and one major non-coding AT-rich region with an arrangement identical to that observed in most insect genomes. The A + T-rich region harbored one identical repeat composed of 65 bp and two tRNA-like structures (trnF and trnK-like sequences) with proper anticodon and clover-leaf structures. Phylogenetic reconstruction using the concatenated sequences of 13 protein-coding genes (PCGs) and two rRNAs of the representative odonate mitogenomes utilizing both Bayesian inference and maximum-likelihood methods revealed a strong support for the monophyletic Zygoptera and a moderate to high support for the monophyletic Anisoptera suborders. Unlike that in conventional phylogenetic analysis, a relatively strong sister relationship was revealed between the suborders of Anisozygoptera and Zygoptera.
