Project description:Fluorescent semiconductor nanoparticles, or quantum dots, have become a promising platform for the engineering of biofunctional probes for a variety of biomedical applications, ranging from multicolor imaging to single-molecule tracking to traceable drug delivery. Advances in organometallic synthesis have enabled preparation of hydrophobic quantum dots with high quantum yields and narrow size distribution, offering bright optical materials with narrow size-tunable emission profiles. At the same time, polymer encapsulation procedures provided a simple and versatile methodology for transferring hydrophobic nanoparticles into physiologically-relevant aqueous buffers. Taken together, hydrophobic nanoparticle platforms and polymer encapsulation should offer great flexibility for implementation of novel probe designs. However, the success of the encapsulation and purification depends on many factors often overlooked in the scientific literature, such as close match between nanoparticle and polymer physicochemical properties and dimensions, slow dynamics of polymer arrangement on the nanoparticle surface, and the size and charge similarity of resultant polymer-coated quantum dots and empty byproduct polymer micelles. To make this general hydrophobic nanoparticle modification strategy accessible by a broad range of biomedical research groups, we focus on the important technical aspects of nanoparticle polymer encapsulation, purification, bioconjugation, and characterization.

Project description:This paper presents a simple and highly efficient method for dopamine detection using water-soluble carbon dot nanoparticles. The ZnO-associated carbon dots (CDZs) were synthesized using a green chemical strategy. An examination of the effects of biomolecules on the fluorescence of CDZs revealed selective dopamine-induced quenching. In a phosphate buffer (pH = 7.4) medium, a detection limit of 1.06 nM was obtained. This "turn off" phenomenon was attributed to the electronic interaction between CDZs and dopamine, during the oxidation of dopamine. At lower pH, however, the effects of dopamine on the fluorescence of CDZs were insignificant as the oxidation of dopamine was hindered when the proton concentration was increased. This method was found to be free from the interference of coexisting molecules, that is, ascorbic acid and uric acid. This sensing platform was applied successfully in biological fluids to confirm the practical significance of the as-designed sensor.

Project description:Allylamine-functionalized silicon quantum dots (ASQDs) of high photostability are synthesized by a robust inverse micelle method to use the material as a fluorescent probe for selective recognition of thiocyanate (a biomarker of a smoker and a nonsmoker). The synthesized ASQDs were characterized by absorption, emission, and Fourier transform infrared spectroscopy. Surface morphology is studied by transmission electron microscopy and dynamic light scattering. The synthesized material exhibits desirable fluorescence behavior with a high quantum yield. A selective and accurate (up to 10-10 M) method of sensing of thiocyanate anion is developed based on fluorescence amplification and quenching of ASQDs. The sensing mechanism is investigated and interpreted with a crystal clear mechanistic approach through the modified Stern-Volmer plot. The developed material and the method is applied to recognize the anion in the human blood sample for identification of the degree of smoking. The material deserves high potentiality in the field of bio-medical science.

Project description:Environmentally friendly polyethylene terephthalate-based carbon dots (PET-CDs) with ultrahigh fluorescence quantum yield were prepared with waste PET textiles as raw materials. First, oligomers were prepared from the reaction of waste PET textile and ethylene glycol by the microwave method. Then, the mixture without isolation and purification as well as pyromellitic acid and urea were adopted as precursors for the preparation of PET-CDs by the hydrothermal method. It was found that the as-prepared PET-CDs had a spherical structure with an average particle size of 2.8 nm. The carbon core of PET-CDs was a graphene-like structure doped with nitrogen atoms in the form of pyrrole nitrogen and the surface contained -NH2, which is convenient for modification and functionalization with various materials in the form of chemical bonds. The as-prepared PET-CDs exhibit excitation-independent emission properties in the range from 340 to 440 nm, and the best excitation and emission wavelengths of PET-CDs are 406 and 485 nm, respectively, while the fluorescence quantum yield is 97.3%. In terms of the application, the as-prepared PET-CDs could be adopted as a fluorescence probe for the detection of Fe3+, and the limit of detection is as low as 0.2 μmol/L. The mechanism of PET-CDs by Fe3+ was found to be the static quenching mechanism. In addition, PET-CDs can be used in LEDs and fluorescent anticounterfeiting.

Project description:Polycyclic aromatic hydrocarbons (PAHs) are potentially harmful pollutants that are emitted into the environment from a range of sources largely due to incomplete combustion. The potential toxicity and carcinogenic effects of these compounds warrants the development of rapid and cost-effective methods for their detection. This work reports on the synthesis and use of graphene quantum dots (GQDs) as rapid fluorescence sensors for detecting PAHs in water. The GQDs were prepared from two sources, i.e. graphene oxide (GO) and citric acid (CA) - denoted GO-GQDs and CA-GQDs, respectively. Structural and optical properties of the GQDs were studied using TEM, Raman, and fluorescence and UV-vis spectroscopy. The GQDs were then applied for detection of pyrene in environmental water samples based on a "turn-off-on" mechanism where ferric ions were used for turn-off and pyrene for turn-on of fluorescence emission. The fluorescence intensity of both GQDs was switched on linearly within the 2-10 × 10-6 mol L-1 range and the limits of detection were found to be 0.325 × 10-6 mol L-1 and 0.242 × 10-6 mol L-1 for GO-GQDs and CA-GQDs, respectively. Finally, the potential application of the sensor for environmental water samples was investigated using lake water and satisfactory recoveries (97-107%) were obtained. The promising results from this work demonstrate the feasibility of pursuing cheaper and greener environmental monitoring techniques.

Project description:The incorporation of functionalised carbon nanodots within a novel low molecular weight salt hydrogel derived from 5-aminosalicylic acid is reported. The carbon dots result in markedly enhanced gelation properties, while inclusion within the hydrophobic gel results in a dramatic fluorescence enhancement for the carbon nanomaterials. The resulting hybrid CD gels exhibit a useful sensor response for heavy metal ions, particularly Pb2+.

Project description:Herein, for the first time the carbon dots (CDs) were synthesized by reflux method from sawmill waste material. We also represent a novel strategy based on fluorescent CDs for determination of ponceau 4R and allura red dyes in soft drinks. Interestingly, both the dyes were sensitive and showed effective fluorescence quenching of the CDs owing to the interaction between them. The analytical applicability of CDs were evaluated for detection of both the dyes with a good linear relationship between the concentration range of 0.0 to 3.0 µg mL-1 and having detection limit 0.45 and 0.47 µg mL-1 for allura red and ponceau 4R dyes respectively. Meanwhile, the potential application of this novel fluorescent probe for dyes determination in real samples was validated in different soft drink samples with good accuracy and precision. Thus, these findings provides new insights for the potential risk assessment of both the dyes. Moreover, CDs acted as an excellent fluorescent material in cellular imaging owing to their cellular uptake and localization.

Project description:Currently, the large-scale application of carbon quantum dots (CQDs) is usually limited by their low quantum yield and detection limit. Herein, the abandoned longan nucleus was used as a carbon source to synthesize cerium-nitrogen-codoped carbon quantum dots (Ce/N-CQDs) with strong luminescence intensity. In this work, the fluorescent properties and fluorescent quantum yield of CQDs may be improved by the single cerium-doped carbon quantum dots (Ce-CQDs) and the single nitrogen-doped carbon quantum dots (N-CQDs). Nevertheless, the Ce/N-CQDs exhibited intense fluorescence with a high quantum yield. Compared with CQDs, the quantum yield of Ce/N-CQDs was significantly increased from 5 to 32% and showed high photostability and good water solubility. The Ce/N-CQDs can be used for the direct detection of rifampicin (RFP) in human serum. The concentration demonstrated a good linear relationship in the range of 1.0 × 10-7-9.0 × 10-6 mol/L, with a detection limit of 9.6 × 10-8 mol/L.

Project description:Quantum dots (QDs) are nanocrystals with bright fluorescence and long-term photostability, attributes particularly beneficial for single-molecule imaging and molecular counting in the life sciences. The size of a QD nanocrystal determines its physicochemical and photophysical properties, both of which dictate the success of imaging applications. Larger nanocrystals typically have better optical properties, with higher brightness, red-shifted emission, reduced blinking, and greater stability. However, larger nanocrystals introduce molecular-labeling biases due to steric hindrance and nonspecific binding. Here, we systematically analyze the impact of nanocrystal size on receptor labeling in live and fixed cells. We designed three (core)shell QDs with red emission (600-700 nm) and crystalline sizes of 3.2, 5.5, and 8.3 nm. After coating with the same multidentate polymer, hydrodynamic sizes were 9.2 nm (QD9.2), 13.3 nm (QD13.3), and 17.4 nm (QD17.4), respectively. The QDs were conjugated to streptavidin and applied as probes for biotinylated neurotransmitter receptors. QD9.2 exhibited the highest labeling specificity for receptors in the narrow synaptic cleft (∼20-30 nm) in living neurons. However, for dense receptor labeling for molecular counting in live and fixed HeLa cells, QD13.3 yielded the highest counts. Nonspecific binding rose sharply for hydrodynamic sizes larger than 13.3 nm, with QD17.4 exhibiting particularly diminished specificity. Our comparisons further highlight needs to continue engineering the smallest QDs to increase single-molecule intensity, suppress blinking frequency, and inhibit nonspecific labeling in fixed and permeabilized cells. These results lay a foundation for designing QD probes with further reduced sizes to achieve unbiased labeling for quantitative and single-molecule imaging.
