Hierarchical computation of 3D motion across macaque areas MT and FST.

ABSTRACT: Computing behaviorally relevant representations of three-dimensional (3D) motion from two-dimensional (2D) retinal signals is critical for survival. To ascertain where and how the primate visual system performs this computation, we recorded from the macaque middle temporal (MT) area and its downstream target, the fundus of the superior temporal sulcus (area FST). Area MT is a key site of 2D motion processing, but its role in 3D motion processing is controversial. The functions of FST remain highly underexplored. To distinguish representations of 3D motion from those of 2D retinal motion, we contrast responses to multiple motion cues during a motion discrimination task. The results reveal a hierarchical transformation whereby many FST but not MT neurons are selective for 3D motion. Modeling results further show how generalized, cue-invariant representations of 3D motion in FST may be created by selectively integrating the output of 2D motion selective MT neurons.

Hierarchical and distinct biological motion processing in macaque visual cortical areas MT and MST.

Project description:It is widely accepted that biological motion (BM) perception involves the posterior superior temporal sulcus (pSTS). Yet, how individual neurons and neural circuits in pSTS encode BM remains unclear. Here we combined electrophysiological recordings with neural network modeling to elucidate BM computations in two subregions of pSTS. We recorded single-cell activity from the middle temporal area (MT) and the medial superior temporal area (MST) of three macaque monkeys when they viewed point-light displays portraying BM walking in different directions (left vs. right), orientations (upright vs. inverted), and forms (intact vs. scrambled). We found that, while individual neurons in both MT and MST showed selectivity for these features, neural populations in MST but not MT exhibit BM-specific encoding, i.e., preferential representation of intact upright BM-the defining characteristic of BM recognition. A neural network model trained to replicate these neurophysiological findings implicated that, BM-specific encoding in MST may arise from feedforward connectivity patterns, i.e., MT subpopulations selective for linear translational motion and nonlinear optic flow projected preferentially to distinct MST cells. Taken together, our findings highlight hierarchical and distinct BM processing in MT and MST, advancing our understanding of BM computations in pSTS at the single-cell and neural circuit levels in the primate brain.

Motion-selective areas V5/MT and MST appear resistant to deterioration in choroideremia.

Project description:Choroideremia (CHM) is an X-linked recessive form of hereditary retinal degeneration, which preserves only small islands of central retinal tissue. Previously, we demonstrated the relationship between central vision and structure and population receptive fields (pRF) using functional magnetic resonance imaging (fMRI) in untreated CHM subjects. Here, we replicate and extend this work, providing a more in-depth analysis of the visual responses in a cohort of CHM subjects who participated in a retinal gene therapy clinical trial. fMRI was conducted in six CHM subjects and six age-matched healthy controls (HC's) while they viewed drifting contrast pattern stimuli monocularly. A single ∼3-minute fMRI run was collected for each eye. Participants also underwent ophthalmic evaluations of visual acuity and static automatic perimetry (SAP). Consistent with our previous report, a single ∼ 3 min fMRI run accurately characterized ophthalmic evaluations of visual function in most CHM subjects. In-depth analyses of the cortical distribution of pRF responses revealed that the motion-selective regions V5/MT and MST appear resistant to progressive retinal degenerations in CHM subjects. This effect was restricted to V5/MT and MST and was not present in either primary visual cortex (V1), motion-selective V3A or regions within the ventral visual pathway. Motion-selective areas V5/MT and MST appear to be resistant to the continuous detrimental impact of CHM. Such resilience appears selective to these areas and may be mediated by independent retina-V5/MT anatomical connections that bypass V1. We did not observe any significant impact of gene therapy.

Functional localization of visual motion area FST in humans.

Project description:The fundus of the superior temporal sulcus (FST) in macaques is implicated in the processing of complex motion signals, yet a human homolog remains elusive. Here we considered potential localizers and evaluated their effectiveness in delineating putative FST (pFST), from hMT and MST, two nearby motion-sensitive areas in humans. Nine healthy participants underwent scanning sessions with 2D and 3D motion localizers, as well as population receptive field (pRF) mapping. We observed consistent anterior and inferior activation relative to hMT and MST in response to stimuli that contained coherent 3D, but not 2D, motion. Motion opponency and myelination measures further validated the functional and structural distinction between pFST and hMT/MST. At the same time, standard pRF mapping techniques that reveal the visual field organization of hMT/MST proved suboptimal for delineating pFST. Our findings provide a robust framework for localizing pFST in humans, and underscore its distinct functional role in motion processing.

Anatomy and Physiology of Macaque Visual Cortical Areas V1, V2, and V5/MT: Bases for Biologically Realistic Models.

Project description:The cerebral cortex of primates encompasses multiple anatomically and physiologically distinct areas processing visual information. Areas V1, V2, and V5/MT are conserved across mammals and are central for visual behavior. To facilitate the generation of biologically accurate computational models of primate early visual processing, here we provide an overview of over 350 published studies of these three areas in the genus Macaca, whose visual system provides the closest model for human vision. The literature reports 14 anatomical connection types from the lateral geniculate nucleus of the thalamus to V1 having distinct layers of origin or termination, and 194 connection types between V1, V2, and V5, forming multiple parallel and interacting visual processing streams. Moreover, within V1, there are reports of 286 and 120 types of intrinsic excitatory and inhibitory connections, respectively. Physiologically, tuning of neuronal responses to 11 types of visual stimulus parameters has been consistently reported. Overall, the optimal spatial frequency (SF) of constituent neurons decreases with cortical hierarchy. Moreover, V5 neurons are distinct from neurons in other areas for their higher direction selectivity, higher contrast sensitivity, higher temporal frequency tuning, and wider SF bandwidth. We also discuss currently unavailable data that could be useful for biologically accurate models.

A hierarchical 3D-motion learning framework for animal spontaneous behavior mapping.

Project description:Animal behavior usually has a hierarchical structure and dynamics. Therefore, to understand how the neural system coordinates with behaviors, neuroscientists need a quantitative description of the hierarchical dynamics of different behaviors. However, the recent end-to-end machine-learning-based methods for behavior analysis mostly focus on recognizing behavioral identities on a static timescale or based on limited observations. These approaches usually lose rich dynamic information on cross-scale behaviors. Here, inspired by the natural structure of animal behaviors, we address this challenge by proposing a parallel and multi-layered framework to learn the hierarchical dynamics and generate an objective metric to map the behavior into the feature space. In addition, we characterize the animal 3D kinematics with our low-cost and efficient multi-view 3D animal motion-capture system. Finally, we demonstrate that this framework can monitor spontaneous behavior and automatically identify the behavioral phenotypes of the transgenic animal disease model. The extensive experiment results suggest that our framework has a wide range of applications, including animal disease model phenotyping and the relationships modeling between the neural circuits and behavior.

Dynamics of macaque MT cell responses to grating triplets.

Project description:Neurons in area MT are sensitive to the direction of motion of gratings and of plaids made by summing 2 gratings moving in different directions. MT component direction-selective (CDS) neurons respond to the individual gratings of a plaid. Pattern direction-selective (PDS) neurons on the other hand, combine component information and respond selectively to the resulting pattern motion. Adding a third grating creates a "triplaid," which contains 3 grating and 3 plaid motions and is perceptually multistable. To examine how direction-selective mechanisms parse the motion signals in triplaids, we recorded MT responses of anesthetized and awake macaques to stimuli in which 3 identical moving gratings whose directions were separated by 120° were introduced in 3 successive epochs, going from grating to plaid to triplaid. CDS and PDS neurons-selected based on their responses to gratings and plaids-had strikingly different tuning properties in the triplaid epoch. CDS neurons were strongly tuned for the direction of motion of individual gratings, but PDS neurons nearly lost their selectivity for either the gratings or the plaids in the stimulus. We explain this reduced motion selectivity with a model that relates pattern selectivity of PDS neurons to a broad pooling of V1 afferents with a near-cosine weighting profile. Because PDS neurons signal both component and pattern motion in gratings and plaids, their reduced selectivity for motion in triplaids may be what makes these stimuli perceptually multistable.

Decoding of visual attention from LFP signals of macaque MT.

Project description:The local field potential (LFP) has recently been widely used in brain computer interfaces (BCI). Here we used power of LFP recorded from area MT of a macaque monkey to decode where the animal covertly attended. Support vector machines (SVM) were used to learn the pattern of power at different frequencies for attention to two possible positions. We found that LFP power at both low (<9 Hz) and high (31-120 Hz) frequencies contains sufficient information to decode the focus of attention. Highest decoding performance was found for gamma frequencies (31-120 Hz) and reached 82%. In contrast low frequencies (<9 Hz) could help the classifier reach a higher decoding performance with a smaller amount of training data. Consequently, we suggest that low frequency LFP can provide fast but coarse information regarding the focus of attention, while higher frequencies of the LFP deliver more accurate but less timely information about the focus of attention.

State dependence of stimulus-induced variability tuning in macaque MT.

Project description:Behavioral states marked by varying levels of arousal and attention modulate some properties of cortical responses (e.g. average firing rates or pairwise correlations), yet it is not fully understood what drives these response changes and how they might affect downstream stimulus decoding. Here we show that changes in state modulate the tuning of response variance-to-mean ratios (Fano factors) in a fashion that is neither predicted by a Poisson spiking model nor changes in the mean firing rate, with a substantial effect on stimulus discriminability. We recorded motion-sensitive neurons in middle temporal cortex (MT) in two states: alert fixation and light, opioid anesthesia. Anesthesia tended to lower average spike counts, without decreasing trial-to-trial variability compared to the alert state. Under anesthesia, within-trial fluctuations in excitability were correlated over longer time scales compared to the alert state, creating supra-Poisson Fano factors. In contrast, alert-state MT neurons have higher mean firing rates and largely sub-Poisson variability that is stimulus-dependent and cannot be explained by firing rate differences alone. The absence of such stimulus-induced variability tuning in the anesthetized state suggests different sources of variability between states. A simple model explains state-dependent shifts in the distribution of observed Fano factors via a suppression in the variance of gain fluctuations in the alert state. A population model with stimulus-induced variability tuning and behaviorally constrained information-limiting correlations explores the potential enhancement in stimulus discriminability by the cortical population in the alert state.

Motion-dependent representation of space in area MT+.

Project description:How is visual space represented in cortical area MT+? At a relatively coarse scale, the organization of MT+ is debated; retinotopic, spatiotopic, or mixed representations have all been proposed. However, none of these representations entirely explain the perceptual localization of objects at a fine spatial scale--a scale relevant for tasks like navigating or manipulating objects. For example, perceived positions of objects are strongly modulated by visual motion; stationary flashes appear shifted in the direction of nearby motion. Does spatial coding in MT+ reflect these shifts in perceived position? We performed an fMRI experiment employing this "flash-drag" effect and found that flashes presented near motion produced patterns of activity similar to physically shifted flashes in the absence of motion. This reveals a motion-dependent change in the neural representation of object position in human MT+, a process that could help compensate for perceptual and motor delays in localizing objects in dynamic scenes.

Orientation anisotropies in macaque visual areas.

Project description:In mammals, a larger number of neurons in V1 are devoted to cardinal (horizontal and vertical) orientations than to oblique orientations. However, electrophysiological results from the macaque monkey visual cortex are controversial. Both isotropic and anisotropic orientation distributions have been reported. It is also unclear whether different visual areas along the visual hierarchy have different orientation anisotropies. We analyzed orientation maps in a large set of intrinsic signal optical imaging data and found that both V1 and V4 exhibited significant orientation anisotropies. However, their overrepresented orientations were very different: in V1, both cardinal and radial orientations were overrepresented, while in V4, only cardinal bias was presented. These findings suggest that different cortical areas have evolved to emphasize different features that are suitable for their functional purposes, a factor that needs to be considered when efforts are made to explain the relationships between the visual environment and the cortical representation and between the cortical representation and visual perception.
