Project description:Abstract The Botryosphaeriales represents an ecologically diverse group of fungi, comprising endophytes, saprobes, and plant pathogens. In this study, taxonomic analyses were conducted based on morphological characteristics and phylogenetic analyses of multi-gene sequence data from four loci (ITS, LSU, tef1-α, and tub2). Thirteen isolates obtained from Beijing and Yunnan Province were identified as seven species of Botryosphaeriales, including Aplosporellajaveedii, Dothiorellaalpina, Phaeobotryonaplosporum and Ph.rhois, and three previously undescribed species, namely Aplosporellayanqingensis, Dothiorellabaihuashanensis, and Phaeobotryonplatycladi. Additionally, the new records of Dothiorellaalpina from the host species Populusszechuanica, Phaeobotryonaplosporum from Juglansmandshurica, and Phaeobotryonrhois from Populusalbavar.pyramidalis are included.

Project description:Cytospora (Cytosporaceae, Diaporthales) is a fungal genus that usually inhabits plants as endophytes, saprobes, as well as pathogens. Species of this genus are characterized by possessing allantoid hyaline conidia and ascospores. Samples with typical Cytospora canker symptoms on Prunus davidiana, P. padus and Salix sp. were collected in Tibet and Xinjiang, China. Species were identified using both morphological and molecular approaches of combined loci of internal transcribed spacer region rDNA (ITS), the partial actin (act) region, RNA polymerase II second largest subunit (rpb2), the translation elongation factor 1-alpha (tef1) gene and the partial be-ta-tubulin (tub2) gene. Six isolates in the present study formed three distinct clades from previously known species. Cytospora hejingensis sp. nov. from Salix sp., C. jilongensis sp. nov. from P. davidiana and C. kunsensis from P. padus were proposed herein. The current study improves the understanding of species concept in Cytospora.

Project description:Ophiocordyceps species have a wide range of insect hosts, from solitary beetle larva to social insects. However, among the species of Ophiocordyceps, only a few attack cicada nymphs. These species are mainly clustered in the Ophiocordycepssobolifera clade in Ophiocordyceps. A new entomopathogenic fungus parasitic on cicada nymphs, and another fungus parasitic on the larva of Coleoptera, are described in this study. The two new species viz. Ophiocordycepshydrangea and Ophiocordycepsbidoupensis were introduced based on morphology and multigene phylogenetic evidence. The phylogenetic framework of Ophiocordyceps was reconstructed using a multigene (nrSSU, nr LSU, tef-1α, rpb1, and rpb2) dataset. The phylogenetic analyses results showed that O.hydrangea and O.bidoupensis were statistically well-supported in the O.sobolifera clade, forming two separate subclades from other species of Ophiocordyceps. The distinctiveness of these two new species was strongly supported by both molecular phylogeny and morphology.

Project description:This study presents the identification of four novel Xylaria species, discovered in the karst region of China. The discovery was facilitated by a rigorous analysis that encompassed both morpho-anatomical features and multi-locus phylogenetics utilizing sequences from the ITS, rpb2, and TUB2 loci. The newly identified species are designated as Xylariajichuanii sp. nov., X.nanningensis sp. nov., X.orientalis sp. nov., and X.taiyangheensis sp. nov. The distinction of these species from their known counterparts was verified through comparison of morphological features and phylogenetic analysis. The study further provides detailed morphological descriptions, illustrative representations, and a phylogenetic tree, all of which contribute to the taxonomic positioning of these novel species.

Project description:The genus Samsoniella was erected based on orange cylindrical to clavate stromata, superficial perithecia and conidiophores with Isaria-like phialides and to segregate them from the Akanthomyces group. In this study, based on morphological features and multigene (SSU, LSU, TEF, RPB1 and RPB2) phylogenetic analysis six Samsoniella species parasitizing spiders were collected in China. Three of them belong to known species S.alpina, S.erucae and S.hepiali. Three new species S.anhuiensissp. nov., S.araneasp. nov. and S.fusiformisporasp. nov. are illustrated and described. They are clearly distinct from other species in Samsoniella occurring in independent subclades. Furthermore, among the four insect-pathogenic fungi specimens collected from similar sites, three of them were identified as the new species described below. Our study significantly broadens the host range of Samsoniella from Insecta to Arachnida, marking a noteworthy expansion in understanding the ecological associations of these fungi. Additionally, the identification of both mononematous and synnematous conidiophores in our study not only expands the knowledge of Samsoniella species but also provides a basis for future research by comparing the ecological significance between these conidiophore types. In conclusion, our study enhances the understanding of Samsoniella diversity, presenting a refined phylogenetic framework and shedding light on the ecological roles of these fungi in spider parasitism.

Project description:Dead wood-associated fungi play an important role in wood degradation and the recycling of organic matter in the forest ecological system. Xenasmataceae is a cosmopolitan group of wood-rotting fungi that grows on tropical, subtropical, temperate, and boreal vegetation. In this study, a new fungal order, Xenasmatales, is introduced based on both morphology and multigene phylogeny to accommodate Xenasmataceae. According to the internal transcribed spacer and nuclear large subunit (ITS+nLSU) and nLSU-only analyses of 13 orders, Xenasmatales formed a single lineage and then grouped with orders Atheliales, Boletales, and Hymenochaetales. The ITS dataset revealed that the new taxon Xenasmatella nigroidea clustered into Xenasmatella and was closely grouped with Xenasmatella vaga. In the present study, Xenasmatella nigroidea collected from Southern China is proposed as a new taxon, based on a combination of morphology and phylogeny. Additionally, a key to the Xenasmatella worldwide is provided.

Project description:The genus Cantharellus, commonly known as chanterelles, has recently been divided into six subgenera; however, wider sampling approaches are needed to clarify the relationships within and between these groups. A phylogenetic overview of Cantharellus subgenus Parvocantharellus in China was inferred based on the large subunit nuclear ribosomal RNA gene (nrLSU), the DNA-directed RNA polymerase II subunit 2 (rpb2), and the transcription elongation factor 1-alpha (tef1). A total of nine species from China were assigned to the subgenus, including seven novel species, namely Cantharellusaurantinus, C. austrosinensis, C. galbanus, C. luteolus, C. luteovirens, C. minioalbus, and C. sinominior, and two known species, namely C. albus and C. zangii. The detailed descriptions and illustrations were provided based on the newly obtained data, with the comparisons to closely related species. C. zangii was restudied based on the paratype specimens and multiple new collections from the type locality. Futhermore, the Indian species C. sikkimensis was identified as a synonym of C. zangii based on the morphological and molecular analyses. A key to the Chinese species belonging to the subg. Parvocantharellus is also provided.

Project description:Agaricus is a saprophytic mushroom genus widely distributed throughout the world. In this study, a survey of the Agaricus species carried out around Chiang Mai University in northern Thailand from 2018 to 2019 yielded 12 collections. Morphological characteristics and phylogenic analyses based on the internal transcribed spacers (ITS) and a fragment of the large subunit (LSU) of the nuclear ribosomal DNA (rDNA), and a fragment of the translation elongation factor 1-alpha (tef1) genes were investigated. The results revealed that these collections belong to six species including Agaricus erectosquamosus, Agaricus pallidobrunneus, Agaricus subrufescens, and three new species. Agaricus thailandensis sp. nov. was found to belong to Agaricus sect. Minores, which is placed in Agaricus subg. Minores. Aagricus pseudoerectosquamosus sp. nov. was placed in Agaricus sect. Brunneopicti within Agaricus subg. Pseudochitonia. Furthermore, Agaricus lannaensis remains an incertae sedis in Agaricus subg. Pseudochitonia. Additionally, this study was proposed that A. pallidobrunneus was discovered in Thailand for the first time. Full descriptions, color photographs, illustrations, and phylogenetic trees are provided.

Project description:Peatswamp forest is a unique habitat that supports high biodiversity, particularly fungal diversity. The current study collected submerged and dead plant parts from Eleiodoxaconferta, Eugeissonatristis and Licualapaludosa from a peatswamp forest in Narathiwat Province, Thailand. Morphological features coupled with multigene phylogenetic analyses of ITS, LSU, rpb2 and tef1-α sequence data identified our isolates as new Distoseptispora species (viz. D.arecacearumsp. nov., D.eleiodoxaesp. nov. and D.narathiwatensissp. nov.). Morphological descriptions, illustrations and notes are provided.

Project description:Calonectria pauciramosa is a pathogen of numerous plant hosts worldwide. Recent studies have indicated that it included cryptic species, some of which are identified in this study. Isolates from various geographical origins were collected and compared based on morphology, DNA sequence data of the beta-tubulin, histone H3 and translation elongation factor-1alpha regions and mating compatibility. Comparisons of the DNA sequence data and mating compatibility revealed three new species. These included Ca. colombiana sp. nov. from Colombia, Ca. polizzii sp. nov. from Italy and Ca. zuluensis sp. nov. from South Africa, all of which had distinguishing morphological features. Based on DNA sequence data, Ca. brasiliensis is also elevated to species level.
