Persistence of foliar applied and pre-storage seed-treated insecticides in rice and its processed products.

ABSTRACT: A field study was conducted to investigate the persistence of foliar-applied thiamethoxam 25% WG at a rate of 25 g ai ha^-1 and chlorantraniliprole 18.5% SC at 30 g ai ha^-1 in various parts of rice plants, including whole grain rice, brown rice, bran, husk, straw, and cooked rice. Liquid Chromatography-Mass spectrometry/Mass spectrometry was used for sample analysis. Chlorantraniliprole residues were found to persist in whole grains, bran, husk, and straw at the time of harvest, while thiamethoxam residue was not detected in harvested grains, processed products, or straw. The study concluded that foliar-applied chlorantraniliprole and thiamethoxam did not pose any dietary risk in cooked rice. In a pre-storage seed treatment study, thiamethoxam 30% FS at 3 mL kg^-1 was evaluated against Angoumois grain moth infestation during storage. The seeds remained unharmed for nine months and exhibited significantly less moth damage (2.0%) even after twelve months of storage. Thiamethoxam residues persisted for more than one year in whole rice grain, brown rice, bran, and husk with seed treatment, with higher residue levels observed in bran and husk. Parboiling and cooking led to the degradation of thiamethoxam residues.

Project description:For the treatment of foliar diseases of cereals, fungicides may be applied as foliar sprays or systemic seed treatments which are translocated to leaves. Little research has been done to assess the resistance risks associated with foliar-acting systemic seed treatments when used alone or in combination with foliar sprays, even though both types of treatment may share the same mode of action. It is therefore unknown to what extent adding a systemic seed treatment to a foliar spray programme poses an additional resistance risk and whether in the presence of a seed treatment additional resistance management strategies (such as limiting the total number of treatments) are necessary to limit the evolution of fungicide-resistance. A mathematical model was developed to simulate an epidemic and the resistance evolution of Zymoseptoria tritici on winter wheat, which was used to compare different combinations of seed and foliar treatments by calculating the fungicide effective life, i.e. the number of years before effective disease control is lost to resistance. A range of parameterizations for the seed treatment fungicide and different fungicide uptake models were compared. Despite the different parameterizations, the model consistently predicted the same trends in that i) similar levels of efficacy delivered either by a foliar-acting seed treatment, or a foliar application, resulted in broadly similar resistance selection, ii) adding a foliar-acting seed treatment to a foliar spray programme increased resistance selection and usually decreased effective life, and iii) splitting a given total dose-by adding a seed treatment to foliar treatments, but decreasing dose per treatment-gave effective lives that were the same as, or shorter than those given by the spray programme alone. For our chosen plant-pathogen-fungicide system, the model results suggest that to effectively manage selection for fungicide-resistance, foliar acting systemic seed treatments should be included as one of the maximum number of permitted fungicide applications.

Project description:Plant hormones influence various physiological processes during the growth and development of plants, but their critical roles in influencing yield and antioxidant activities in dry-seeded rice (DSR) have not been adequately explored. This study aims to analyze the performance and antioxidant activity of contrasting genotypes of DSR in response to soil moisture regimes and foliar-applied hormones. The study comprised sixteen treatments that were evaluated under field conditions as per split-plot design in three replications. Treatments comprised combinations of two soil moisture tension regimes (10 kPa and 20 kPa) and two genotypes (PR-111, non-stay-green type and PR-123, stay-green type) applied to the main plots and foliar application of three hormones (gibberellic acid (GA3) 40 mg kg-1, abscisic acid (ABA) 20 mg kg-1, and cytokinin (CK) 40 mg kg-1)) and a control (unsprayed) to subplots. The non-stay-green genotype (PR-111) resulted in 34.6% more grain yield (6.48 t ha-1) than the stay-green genotype (PR-123) at the lower soil moisture tension regime (SMTR) (10 kPa) due to the increased number of filled grains per panicle and improvement in harvest index (HI). At the higher SMTR (20 kPa), the stay-green genotype (PR-123) produced 26.4% more grain yield (5.21 t ha-1) than non-stay green genotype (4.12 t ha-1) and showed enhanced superoxide dismutase (SOD) and peroxide dismutase (POD) activity that may have contributed in maintaining sink size through improved chlorophyll content. Grain yield (6.35 t ha-1) with foliar-applied GA3 (40 mg kg-1) at SMTR of 10 kPa was higher by 12.2% and 24.0% than with foliar-applied ABA (20 mg kg-1) and unsprayed treatments, respectively. Irrigation application at SMTR of 20 kPa and foliar application of ABA gave 24.1% higher grain yield (5.15 t ha-1) than the unsprayed treatment, but it was similar to foliar-applied GA3 and CK. This study implied that the stay-green genotype (PR-123) was more suitable under moisture stress conditions (20 kPa) in DSR, as it maintained sink size even under moisture stress conditions by improving dry matter translocation and enhancing SOD and POD activity. The study suggests the need to find out the endogenous level of these plant hormones in rice genotypes under a range of water regimes to develop high yielding and water use efficient genotypes of DSR.

Project description:Soil salinity is one of the important abiotic stress factors that affect rice productivity and quality. Research with several dicotyledonous plants indicated that the detrimental effects associated with salinity stress can (partly) be overcome by the external application of antioxidative substances. For instance, sodium selenate (Na2SeO4) significantly improved the growth and productivity of several crops under various abiotic stress conditions. At present there is no report describing the impact of Na2SeO4 on salinity stressed cereals such as rice. Rice cultivation is threatened by increasing salinity stress, and in future this problem will further be aggravated by global warming and sea level rise, impacting coastal areas. The current study reports on the effect of Na2SeO4 in alleviating salinity stress in rice plants. The optimal concentration of Na2SeO4 and the most efficient mode of selenium application were investigated. Selenium, sodium, and potassium contents in leaves were determined. Antioxidant enzyme activities as well as proline, hydrogen peroxide (H2O2), and malondialdehyde (MDA) concentrations were analyzed. In addition, the transcript levels for OsNHX1, an important Na+/H+ antiporter, were quantified. Treatment of 2-week-old rice plants under 150 mM NaCl stress with 6 mg l-1 Na2SeO4 improved the total biomass. A significantly higher biomass was observed for the plants that received Na2SeO4 by a combination of seed priming and foliar spray compared to the individual treatments. The Na2SeO4 application enhanced the activity of antioxidant enzymes (SOD, APX, CAT, and GSH-Px), increased the proline content, and reduced H2O2 and MDA concentrations in plants under NaCl stress. These biochemical changes were accompanied by increased transcript levels for OsNHX1 resulting in a higher K+/Na+ ratio in the rice plants under NaCl stress. The results suggest that Na2SeO4 treatment alleviates the adverse effect of salinity on rice plant growth through enhancing the antioxidant defense system and increase of OsNHX1 transcript levels.

Project description:Soil salinity is one of the most challenging environmental factors affecting rice productivity, particularly in regions with high saline soils such as Egypt. The ability of rice to maintain high yield and quality under saline stress is often limited, leading to significant reductions in productivity. With the increasing salinization of agricultural lands, finding effective agronomic practices and treatments to mitigate salt-induced damage in rice crops is critical for ensuring food security. This study investigates the potential of exogenous glycine betaine (GB) and proline (Pro) applications to mitigate the adverse effects of salt stress on rice (cv. Sakha 108) over two consecutive growing seasons (2021-2022). Treatments of 30 mM GB and 30 mM Pro significantly enhanced dry weight (162.2 and 169.7 g in 2021 and 2022, respectively), plant height (88.94 and 99.00 cm), tiller number (10.58 and 10.33), and grain yield (4.22 and 4.30 t/ha) compared to control groups. Combined treatments of 30 mM GB and 30 mM Pro exhibited the greatest improvements across both years, with maximum dry weight (193.44 and 186.56 g), plant height (112.00 and 112.33 cm), tiller number (15.33 and 16.28), spikelet number per meter (264.00 and 264.05), thousand-kernel weight (70.00 and 73.2 g), and grain yield (6.17 and 6.64 t/ha). Additionally, the combined treatments resulted in the highest harvest index (53.22% in 2021 and 48.94% in 2022), amylose content (24.24% and 20.09%), and protein content (12.33% and 12.00%). Correlation analysis highlighted strong positive relationships among traits, such as plant height with grain yield (r = 0.94), biomass yield (r = 0.92), and harvest index (r = 0.90). Path analysis further demonstrated that thousand-kernel weight and biomass yield had the most significant direct effects on grain yield, with values of 0.43 and 0.42, respectively. Heatmap clustering and principal component analysis (PCA) confirmed the synergistic effects of combined GB and Pro treatments, with the 30P_30GB treatment consistently clustering with high-yield traits, enhancing nitrogen use efficiency and stress resilience. In conclusion, the combined application of glycine betaine and proline significantly enhances the agronomic and chemical traits of rice under salt stress. This study demonstrates that these osmoprotectants improve vegetative growth, grain yield, and quality, with synergistic effects observed at optimal concentrations. The findings highlight the potential of glycine betaine and proline as effective tools for improving salt tolerance in rice, offering practical solutions to address challenges in saline-affected agricultural regions.
