Catalytic Asymmetric Synthesis of α-Arylpyrrolidines and Benzo-fused Nitrogen Heterocycles.

Project description:Herein, we report a practical two-step synthetic route to α-arylpyrrolidines through Suzuki-Miyaura cross-coupling and enantioselective copper-catalyzed intramolecular hydroamination reactions. The excellent stereoselectivity and broad scope for the transformation of substrates with pharmaceutically relevant heteroarenes render this method a practical and versatile approach for pyrrolidine synthesis. Additionally, this intramolecular hydroamination strategy facilitates the asymmetric synthesis of tetrahydroisoquinolines and medium-ring dibenzo-fused nitrogen heterocycles.

An unusual formal migrative cycloaddition of aurone-derived azadienes: synthesis of benzofuran-fused nitrogen heterocycles.

Project description:Aurone-derived azadienes are well-known four-atom synthons for direct [4 + n] cycloadditions owing to their s-cis conformation as well as the thermodynamically favored aromatization nature of these processes. However, distinct from this common reactivity, herein we report an unusual formal migrative annulation with siloxy alkynes initiated by [2 + 2] cycloaddition. Unexpectedly, this process generates benzofuran-fused nitrogen heterocyclic products with formal substituent migration. This observation is rationalized by less common [2 + 2] cycloaddition followed by 4π and 6π electrocyclic events. DFT calculations provided support to the proposed mechanism.

Rhodium(III)-catalyzed C-H functionalization of C-alkenyl azoles with sulfoxonium ylides for the synthesis of bridgehead <i>N</i>-fused [5,6]-bicyclic heterocycles.

Project description:The synthesis of bridgehead N-fused [5,6]-bicyclic heterocycles via rhodium(III)-catalyzed C-H functionalization of C-alkenyl azoles with sulfoxonium ylides is disclosed. Reactions proceeded in good to high yields for a range of aryl, heteroaryl and alkyl sulfoxonium ylides. In addition, 2-alkenyl imidazoles with different substitution patterns as well as C-alkenyl triazoles were effective inputs. The reaction could also be performed under straightforward bench top conditions.

Diversity-Oriented Synthesis of [2.2]Paracyclophane-derived Fused Imidazo[1,2-a]heterocycles by Groebke-Blackburn-Bienayme Reaction: Accessing Cyclophanyl Imidazole Ligands Library.

Project description:This report describes the synthesis of a [2.2]paracyclophane-derived annulated 3-amino-imidazole ligand library through a Groebke-Blackburn-Bienaymé three-component reaction (GBB-3CR) approach employing formyl-cyclophanes in combination with diverse aliphatic and aromatic isocyanides and heteroaromatic amidines. The GBB-3CR process gives access to skeletally-diverse cyclophanyl imidazole ligands, namely 3-amino-imidazo[1,2-a]pyridines and imidazo[1,2-a]pyrazines. Additionally, a one-pot protocol for the GBB-3CR by an in situ generation of cyclophanyl isocyanide is demonstrated. The products were analyzed by detailed spectroscopic techniques, and the cyclophanyl imidazo[1,2-a]pyridine was confirmed unambiguously by single-crystal X-Ray crystallography. The cyclophanyl imidazole ligands can be readily transformed to showcase their useful utility in preparing N,C-palladacycles through regioselective ortho-palladation.

Synthesis of Tunable Fluorescent Imidazole-Fused Heterocycle Dimers.

Project description:A short, concise, and one-pot synthesis of imidazo-fused heterocycle dimers with tunable fluorescent properties has been developed. By the first time use of glyoxal dimethyl acetal in the Groebke-Blackburn-Bienaymé (GBB) three-component reaction (3CR), the innovation features a new series of fluorescence-tunable imidazo-fused heterocycle dimers exhibiting a broad substrate scope with good yields. Luminescence studies demonstrate that these GBB-dimers possess color-tunable properties, and their emission colors can be successively changed from blue to green and yellow by easy substituent control.

A multicomponent reaction for modular assembly of indole-fused heterocycles.

Project description:Indoles are privileged chemical entities in natural products and drug discovery. Indole-fused heterocycles, particularly seven-membered ones, have received increasing attention due to their distinctive chemical characteristics and wide spectrum of bioactivities. However, the synthetic access to these compounds is highly limited. Herein, we report a unique multicomponent reaction (MCR) for modular assembly of indole-fused seven-membered heterocycles. In this process, indole, formaldehyde and amino hydrochloride could assemble rapidly to yield indole-fused oxadiazepines, and another addition of sodium thiosulphate would furnish indole-fused thiadiazepines. The biological evaluation disclosed the promising anticancer activity of these compounds. Furthermore, this MCR could be applicable in the late-stage and selective modifications of peptides. Therefore, this work provides a powerful strategy for indole functionalization and valuable tool for construction of seven-membered heterocycles.

Synthesis, structure and aromaticity of carborane-fused carbo- and heterocycles.

Project description:Conjugation between a 3-D icosahedral carborane and a fused 2-D π-ring system is ambiguous. To address this issue, we prepared several carborane-fused carbo- and heterocycles. Detailed studies on their molecular structures, NMR data, and NICS (nucleus-independent chemical shift) and ISE (isomerization stabilization energy) values as well as molecular orbital analyses clearly suggest the presence of (1) considerable aromatic character in the exo five-membered ring of carborane-fused carbo- and heterocycles and (2) considerable conjugation between a 3-D carborane and a fused 2-D π-ring system. These results will shed some light on the design of new carborane-based materials.

NEXAFS Spectra Simulations of Nitrogen-Bearing Heterocycles.

Project description:Five-membered heterocyclic compounds containing nitrogen atoms are important biomolecule building blocks. In addition to their fundamental biological importance, these molecular structures are used in several technological applications. Consequently, it is essential to develop techniques that allow the characterization of these fundamental systems. We address this issue by performing simulations of K-edge NEXAFS spectra by applying a time-dependent density functional theory (TDDFT) and an inner-shell multiconfigurational self-consistent field (IS-MCSCF) of selected molecules. Also, vibronic coupling simulations were considered for the TDDFT computations. Surprisingly, molecular orbital binding energies do not reproduce the order of the transition energies obtained by IS-MCSCF, indicating a possible breakdown of the orbital picture concerning the NEXAFS spectrum. In general, the TDDFT and IS-MCSCF results are compatible and are in close agreement with experimental data. Moreover, vibronic coupling and vertical transition results were very similar. Finally, it is important to mention that, to the best of our knowledge, this is the first time that the IS-MCSCF method has been applied to molecular systems of this size.
