Project description:In this work, the rheological behavior of stable poly(lactic acid) (PLA) dispersions in water, intended for coating applications, was investigated. The newly prepared dispersion consists of PLA particles with an average diameter of 222 ± 2 nm based on dynamic light scattering (DLS) and scanning electron microscopy (SEM) analyses, at concentrations varying in the 5-22 wt % range. Xanthan gum (XG), a bacterial polysaccharide, was used as a thickening agent to modulate the viscosity of the formulations. The rheological properties of the PLA dispersions with different XG and PLA contents were studied in steady shear, amplitude sweep, and frequency sweep experiments. Under steady shear conditions, the viscosity of all the formulations showed a shear-thinning behavior similar to XG solutions in the whole investigated 1-1000 s-1 range, with values dependent on both PLA particles and XG concentrations. Amplitude and frequency sweep data revealed a weak-gel behavior except in the case of the most diluted sample, with moduli dependent on both PLA and XG contents. A unified scaling parameter was identified in the volume fraction (ϕ) of the PLA particles, calculated by considering the dependence of the continuous phase density on the XG concentration. Accordingly, a master curve at different volume fractions was built using the time-concentration-superposition approach. The master curve describes the rheological response of the system over a wider frequency window than the experimentally accessible one and reveals the presence of a superimposed β relaxation process in the high-frequency region.

Project description:This paper presents a dataset collected in laboratory experiments on the settling of solid spheres and disks in shear-thinning and viscoelastic aqueous solutions of xanthan gum with sodium chloride addition. Two types of spheres with density of 1.41 g/cm3 varying in diameter (3.00 mm and 1.59 mm) and four types of disks with density of 1.43 g/cm3 and thickness of 0.3 mm varying in diameter (1.5 mm, 2.0 mm, 2.5 mm, and 3.0 mm) were considered. A single particle was settling in a column filled with a test solution which varied in salt content (from 0 M to 0.9 M), while xanthan gum content was constant (1 g/L). The total of elven solutions were tested. For each experimental set, a sequence of images with a falling particle was captured using a camera with macro lenses. Dataset includes position of particle in time and enables the evaluation of settling velocity. Rheological measurements were carried out for each test solution to assess flow properties and viscoelasticity. The following measurements were performed: shear dependent viscosity, shear stress amplitude sweeps, frequency sweeps, the dependence of the first normal stresses difference on shear strain at constant frequency (1 Hz). Datasets may be useful in various areas on fluid mechanics and rheology, e.g., in research on the impact of salinity on rheological properties of exopolymer solutions, to develop numerical models on solid particles settling in non-Newtonian fluids, and in studies on the impact of exopolymers and electrolytes dissolved in water on settling dynamics of solid particles.

Project description:Oral cancer is one of the leading causes of death worldwide. Oral precancerous lesions (OPL) are the precursors of oral cancer, with varying degrees of progression. Tetrahydrocurcumin (THC) is a major metabolite of curcumin with superior anticancer properties against various types of cancer. However, THC's clinical outcome is limited by its poor aqueous solubility. Herein, we developed novel mucoadhesive biopolymer-based composite sponges for buccal delivery of THC, exploiting nanotechnology and mucoadhesion for efficient prevention and treatment of oral cancer. Firstly, THC-nanocrystals (THC-NC) were formulated and characterized for subsequent loading into mucoadhesive composite sponges. The anticancer activity of THC-NC was assessed on a human tongue squamous carcinoma cell line (SCC-4). Finally, the chemopreventive activity of THC-NC loaded sponges (THC-NC-S) was examined in DMBA-induced hamster OPL. The selected THC-NC exhibited a particle size of 532.68 ± 13.20 nm and a zeta potential of -46.08 ± 1.12 mV. Moreover, THC-NC enhanced the anticancer effect against SCC-4 with an IC50 value of 80 µg/mL. THC-NC-S exhibited good mucoadhesion properties (0.24 ± 0.02 N) with sustained drug release, where 90% of THC was released over 4 days. Furthermore, THC-NC-S had a magnificent potential for maintaining high chemopreventive activity, as demonstrated by significant regression in the dysplasia degree and a decline in cyclin D1 (control: 40.4 ± 12.5, THC-NC-S: 12.07 ± 5.2), culminating in significant amelioration after 25 days of treatment. Conclusively, novel THC-NC-S represent a promising platform for local therapy of OPL, preventing their malignant transformation into cancer.

Project description:To further extend the use of κ-carrageenan (κ-C) in real food systems (such as beverages), the understanding of gelation properties of κ-C with the presence of food ingredients is critical. The effects of xylitol and maltitol (up to 30 wt %) on the rheological and structural properties of κ-C were inspected by means of rheometer and Fourier transform infrared (FTIR). With the addition of xylitol, the gelation temperature increased from 44.1 to 57.3 °C, while the gelation temperature increased from 44.1 to 61.4 °C in maltitol systems. With the increasing concentration of both xylitol and maltitol, the values of fractal dimension df and complex modulus G* of κ-C increased, while the relaxation exponent n decreased from 0.87 to 0.39 of xylitol and 0.87 to 0.78 of maltitol, respectively. These indicated that the gel networks of aqueous κ-C were improved by the addition of xylitol and maltitol. The FTIR results showed that the interaction between κ-C and these polyols contributed to the increase of hydrogen bonds. The effects of maltitol on κ-C were stronger than those of xylitol because of more equatorial-OH bonds in maltitol. These findings contribute to a better understanding of the gelation processes of κ-C/polyols systems.

Project description:Microplastic debris ending up at the sea surface has become a known major environmental issue. However, how microplastic particles move and when they sink in the ocean remains largely unknown. Here, we model microplastic subject to biofouling (algal growth on a substrate) to estimate sinking timescales and the time to reach the depth where particles stop sinking. We combine NEMO-MEDUSA 2.0 output, that represents hydrodynamic and biological properties of seawater, with a particle-tracking framework. Different sizes and densities of particles (for different types of plastic) are simulated, showing that the global distribution of sinking timescales is largely size-dependent as opposed to density-dependent. The smallest particles we simulate (0.1 μm) start sinking almost immediately around the globe and their trajectories take the longest time to reach their first sinking depth (relative to larger particles). In oligotrophic subtropical gyres with low algal concentrations, particles between 1 and 0.01 mm do not sink within the simulation time of 90 days. This suggests that in addition to the comparatively well-known physical processes, biological processes might also contribute to the accumulation of floating plastic (of 1-0.01 mm) in subtropical gyres. Particles of 1 μm in the gyres start sinking largely due to vertical advection, whereas in the equatorial Pacific they are more dependent on biofouling. The qualitative impacts of seasonality on sinking timescales are small, however, localized sooner sinking due to spring algal blooms is seen. This study maps processes that affect the sinking of virtual microplastic globally, which could ultimately impact the ocean plastic budget.

Project description:The use of 3D printing began to diffuse in the pharmaceutical field in recent times, since 2015, with the approval of the first 3D printed drug from the FDA. The most used materials in association with this technique in this specific scope are hydrogels, already used widely in tissue engineering to produce scaffolds used in the realization of synthetic tissues. The aim of this project was to study the influence of different pH conditions on the release of a therapeutic molecule from a Carrageenan-k combined with alginate or xanthan gum hydrogel with a high level of biocompatibility and enough mechanical resistance to be used as printing material for a 3D bioprinter. The addition of biologically active supplements without affecting the model of the 3D printing structure of the biocompatible polymers was achieved by adopting the extrusion at moderately low temperatures. This study aimed to produce the necessary percentage of hydrogels which is responsible for the release of active drugs which respect to the pH of the system. In this case, the chosen curcumin drug which exhibits active release in the pH of the small intestine is a pH value of 6.

Project description:There is a growing need for natural ingredients that could be utilized for the production of food, pharmaceutical, and cosmetic emulsions. Soy protein acid hydrolysate (SPAH) is a plant-based additive used in the food industry mainly as a flavor enhancer. For the purpose of this work, however, it was mixed with a well-known natural polysaccharide, xanthan gum (XG), to produce stable 30% (w/w) sunflower oil-in-water emulsions using a rotor-stator homogenizer. In order to assess the emulsifying properties of the SPAH and its mixtures with XG, the surface tension properties of their water solutions, particle size, creaming stability, and rheological properties of the emulsions were investigated. Since the emulsions prepared using only SPAH, in various concentrations, were not stable, systems containing 5% of SPAH and 0.1, 0.2, 0.3, 0.4, or 0.5% of XG were then studied. The increase in concentration of the macromolecule led to an increase in creaming stability. The emulsions with 5% SPAH and 0.5% XG were stable for at least 14 days. The increase in XG concentration led to a decrease in d4,3, while consistency index and non-Newtonian behavior increased. The systems containing SPAH, in the absence of XG, showed shear-thinning flow behavior, which was changed to thixotropic with the addition of XG. Viscoelastic properties of emulsions containing over 0.2% of XG were confirmed by oscillatory rheological tests, demonstrating the dominance of elastic (G') over viscous (G") modulus.

Project description:The growing popularity of probiotics has led to the generation of substantial by-products. Among these, cell-free supernatant is recognized for containing beneficial postbiotics. Here, we upcycled Lactobacillus casei-free supernatant (LFS) into cheese analogues using inulin (INU), locust bean gum (LBG), and kappa-carrageenen (kCG). In this system, LBG/kCG established the primary structure, while interstitial spaces were progressively filled by INU. Despite the absence of milk proteins and fats, the cheese analogue with 35% w/w INU, 0.2% w/w LBG, and 0.8% kCG exhibited a texture and appearance resembling commercial processed cheese, as determined by texture profile analysis and dynamic small amplitude oscillatory rheometry technique. This can be attributed to the effective fat-replacing activity of INU regarding texture and rheology. Furthermore, the potassium-dominated salt composition of LFS proved advantageous for the LBG/kCG-derived structure-forming. These findings hold significant promise for upcycling probiotics wastewater into low-fat vegan cheese analogues, enriched with both prebiotics and postbiotics.

Project description:The dynamical properties of nanogels of xanthan gum (XG) with hydrodynamic radius controlled in a size range from 5 nm to 35 nm, were studied at the different XG concentrations in water/sodium bis-2-ethylhexyl-sulfosuccinate (AOT)/decane reverse micelles (RMs) vs. mass fraction of nano-droplet (MFD) at W = 40, using dynamic light scattering (DLS). The diffusion study of nanometer-sized droplets by DLS technique indicated that enhancing concentration of the XG polysaccharide resulted in exchanging the attractive interaction between nano-gels to repulsive interaction, as the mass fraction of nano-droplets increased. The reorientation time (τr ) of water nanodroplets decreased with MFD for water-in-oil AOT micro-emulsion comprising high concentration (0.0000625) of XG. On the other hand, decreasing concentration of biopolymer led to increasing the rotational correlation time of water nanodroplets with MFD. In conclusion, a single relaxation curve was observed for AOT inverse microemulsions containing different XG concentrations. Furthermore, the interaction between nanogels was changed from attractive to repulsive versus concentration of XG in the AOT RMs.

Project description:The present study examined the effect of polysaccharides gels made of xanthan gum and locust bean gum (gel culture system) on oocyte maturation and explored the molecules causing the beneficial effect of the gel culture system. Oocytes and cumulus cells complexes were collected from slaughterhouse-derived ovaries and cultured on a plastic plate or gel. The gel culture system improved the rate of development to the blastocyst stage. The oocytes that matured on the gel contained high lipid contents and F-actin formation, and the resultant 8-cell stage embryos had low DNA methylation levels compared to their plate counterparts. RNA sequencing of the oocytes and embryos revealed the differentially expressed genes between the gel and plate culture systems, and upstream regulator analysis revealed estradiol and TGFB1 as top activated upstream molecules. The medium of the gel culture system contained higher concentrations of estradiol and TGFB1 than that of the plate cultures system. Supplementation of the maturation medium with either estradiol or TGFB1 resulted in high lipid content in oocytes. In addition, TGFB1 improved the developmental ability of the oocytes and increased F-actin content while reducing DNA methylation levels in the 8-cell stage embryos. In conclusion, the gel culture system is useful for embryo production, potentially through the upregulation of TGFB1.
