Hydrogenation of Alkynes and Olefins Catalyzed by Quaternary Ammonium Salts.

ABSTRACT: Catalytic hydrogenation of unsaturated hydrocarbons to alkenes and alkanes using molecular hydrogen is one of the most fundamental transformations in organic synthesis. While methodologies involving transition metals as catalysts in homogeneous and heterogeneous processes have been well developed, metal-free catalytic hydrogenation offers an ideal approach for future chemistry. Herein, the common and inexpensive quaternary ammonium salts are first introduced as catalysts in the catalytic hydrogenation system for the transformations from alkynes or olefins into the corresponding olefins or alkanes. Interestingly, the hydrogenation process of alkynes can be controlled to selectively produce alkenes or alkanes under different conditions. Moreover, the possible mechanism is discussed in new insights into the catalytic behavior of quaternary ammonium salts.

Iridium-catalyzed enantioconvergent hydrogenation of trisubstituted olefins.

Project description:Asymmetric hydrogenation of olefins constitutes a practical and efficient method to introduce chirality into prochiral substrates. However, the absolute majority of the developed methodologies is enantiodivergent and thus require isomerically pure olefins which is a considerable drawback since most olefination strategies produce (E/Z)-mixtures. Although some advances have been reported, a general enantioconvergent hydrogenation featuring a broad functional group tolerance remains elusive. Here, we report the development of a general iridium-catalyzed enantioconvergent hydrogenation of a broad range of functionalized trisubstituted olefins. The substitution pattern around the olefin is critical; whereas α-prochiral olefins can undergo an enantioconvergent hydrogenation, β-prochiral olefins react in an enantiodivergent manner. The presented methodology hydrogenates α-prochiral substrates with excellent control of enantioselection and high isolated yields. Most importantly, both isomerically pure alkenes as well as isomeric mixtures can be hydrogenated to yield the same major enantiomer in excellent enantiomeric excesses which is unusual in transition-metal catalyzed asymmetric hydrogenations.

Enantioselective nucleophilic difluoromethylation of aromatic aldehydes with Me3SiCF2SO2Ph and PhSO2CF2H reagents catalyzed by chiral quaternary ammonium salts.

Project description:BackgroundAlthough the nucleophilic difluoromethylation of aldehydes, ketones, and imines has been realized with PhSO(2)CF(2)H and related reagents, there are still no reports on the enantioselective nucleophilic reactions.ResultsWith a chiral quaternary ammonium salt as the catalyst and KOH as the base, we describe the first enantioselective difluoromethylation of aromatic aldehydes with PhSO(2)CF(2)H or Me(3)SiCF(2)SO(2)Ph. The enantioselectivity is substrate-dependent and for 2-chlorinated benzaldehyde an ee up to 64% was obtained.ConclusionThese results provide some insights into the enantioselective nucleophilic difluoromethylation chemistry, which will stimulate further progress in this field.

Sustainable triazine-derived quaternary ammonium salts as antimicrobial agents.

Project description:The first examples of highly efficient antimicrobial triazine-derived bis imidazolium quaternary ammonium salts (TQAS) are reported. TQAS have been prepared with an easy, atom efficient, economically sustainable strategy and tested as antimicrobial agents, reaching MIC values below 10 mg L-1. Distinctively, TQAS have low MIC and low cytotoxicity.

Oxoammonium salts are catalysing efficient and selective halogenation of olefins, alkynes and aromatics.

Project description:Electrophilic halogenation reactions have been a reliable approach to accessing organohalides. During the past decades, various catalytic systems have been developed for the activation of haleniums. However, there is still a short of effective catalysts, which could cover various halogenation reactions and broad scope of unsaturated compounds. Herein, TEMPO (2,2,6,6-tetramethylpiperidine nitroxide) and its derivatives are disclosed as active catalysts for electrophilic halogenation of olefins, alkynes, and aromatics. These catalysts are stable, readily available, and reactive enough to activate haleniums including Br+, I+ and even Cl+ reagents. This catalytic system is applicable to various halogenations including haloarylation of olefins or dibromination of alkynes, which were rarely realized in previous Lewis base catalysis or Lewis acid catalysis. The high catalytic ability is attributed to a synergistic activation model of electrophilic halogenating reagents, where the carbonyl group and the halogen atom are both activated by present TEMPO catalysis.

Synthesis and Antimicrobial Evaluation of Bis-morpholine Triazine Quaternary Ammonium Salts.

Project description:Efficient, environmentally and economically sustainable, and nontoxic antibacterial products are of global relevance in the fight against microorganism contamination. In this work, an easy and straightforward method for the synthesis of bis-morpholino triazine quaternary ammonium salts (bis-mTQAS) is reported, starting from 2,4,6-trichloro-1,3,5-triazine or 2,4-dichloro-6-methoxy-1,3,5-triazine and various N-alkylmorpholines. Bis-mTQAS were tested as antimicrobials against Gram-negative and Gram-positive bacterial strains. The best-performing bis-mTQAS were found to achieve total disinfection against Staphylococcus aureus ATCC 25923 and Escherichia coli ATCC 25922 at 50 and 400 μg/mL, respectively. Distinctively, bis-mTQAS with the highest antimicrobial efficiency had lowest cytotoxicity.

Synthesis, physiochemical property and antimicrobial activity of novel quaternary ammonium salts.

Project description:Twenty-four novel 5-phenyl-1,3,4-oxadiazole-2-thiol (POT) analogues, benzo[d]oxazole-2-thiol, benzo[d]thiazole-2-thiol and 5-methyl-1,3,4-thiadiazole-2-thiol-substituted N,N-bis(2-hydroxyethyl) quaternary ammonium salts (QAS) (5a-d, 6a-d, 7a-d, 10a-d, 13a-d, 16a-d) were prepared and characterised by FTIR, NMR and elemental analysis. Part of target compounds (5d, 6d, 7d, 10d, 13d, 16d) displayed potent antimicrobial effect against ten common pathogens (S. aureus, α-H-tococcus, β-H-tococcus, E. coli, P. aeruginosa, Proteus vulgaris, Canidia Albicans, Cytospora mandshurica, Physalospora piricola, Aspergillus niger) and had relatively low cytotoxity against two human cell lines (HaCat and LO2). TEM and SEM images of E. coli and S. aureus morphologies treated with 7d showed that the antibacterial mechanism might be the QAS fixing on cell wall surfaces and puncturing to result in the release of bacterial cytoplasm. This study provides new information of QAS, which could be used to design novel antimicrobial agents applied in clinic or agriculture.

Synthesis of Both Enantiomers of Chiral Phenylalanine Derivatives Catalyzed by Cinchona Alkaloid Quaternary Ammonium Salts as Asymmetric Phase Transfer Catalysts.

Project description:A practical synthesis of both enantiomers of unnatural phenylalanine derivatives by using two pseudoenantiomeric phase transfer catalysts is described. Through asymmetric α-alkylation of glycine Schiff base with substituted benzyl bromides and 1-(bromomethyl)naphthalene under the catalysis of O-allyl-N-(9-anthracenmethyl) cinchoninium bromide (1f) and O-allyl-N-(9-anthracenylmethyl)cinchonidium bromide (1i), respectively, a series of both (R)- and (S)-enantiomers of unnatural α-amino acid derivatives were obtained in excellent yields and enantioselectivity. The synthetic method is simple and scalable, and the stereochemistry of the products is fully predictable and controlled: the cinchonine-type phase transfer catalyst 1f resulted in (R)-α-amino acid derivatives, and the cinchonidine-type phase transfer catalyst 1i afforded (S)-α-amino acid derivatives.

Crystal structures of three new N-halo-methyl-ated quaternary ammonium salts.

Project description:In the crystals of the title N-halo-methyl-ated quaternary ammonium salts, C19H23IN(+)·I(-), (I) [systematic name: N-(4,4-di-phenyl-but-3-en-1-yl)-N-iodo-methyl-N,N-di-methyl-ammonium iodide], C20H25IN(+)·I(-), (II) [systematic name: N-(5,5-di-phenyl-pent-4-en-1-yl)-N-iodo-methyl-N,N-di-methyl-ammonium iodide], and C21H27IN(+)·I(-), (III) [systematic name: N-(6,6-di-phenyl-hex-5-en-1-yl)-N-iodo-methyl-N,N-di-methyl-ammonium iodide], there are short I⋯I(-) inter-actions of 3.564 (4), 3.506 (1) and 3.557 (1) Å for compounds (I), (II) and (III), respectively. Compound (I) crystallizes in the Sohncke group P21 as an 'enanti-opure' compound and is therefore a potential material for NLO properties. In the crystal of compound (I), mol-ecules are linked by C-H⋯I(-) and C-H⋯π inter-actions which, together with the I⋯I(-) inter-actions, lead to the formation of ribbons along [100]. In (II), there are only C-H⋯I(-) inter-actions which, together with the I⋯I(-) inter-actions, lead to the formation of helices along [010]. In (III), apart from the I⋯I(-) inter-actions, there are no significant inter-molecular inter-actions present.

Quaternary ammonium salts based on (-)-borneol as effective inhibitors of influenza virus.

Project description:A series of compounds containing a 1,7,7-trimethylbicyclo[2.2.1]heptane fragment were evaluated for their antiviral activity against influenza A virus strain A/Puerto Rico/8/34 (H1N1) in vitro. The most potent antiviral compound proved to be a quaternary ammonium salt based on (-)-borneol, 10a. In in vitro experiments, compound 10a inhibited influenza A viruses (H1, H1pdm09, and H3 subtypes), with an IC50 value of 2.4-16.8 µM (depending on the virus), and demonstrated low toxicity (CC50 = 1311 µM). Mechanism-of-action studies for compound 10a revealed it to be most effective when added at the early stages of the viral life cycle. In direct haemolysis inhibition tests, compound 10a was shown to decrease the membrane-disrupting activity of influenza A virus strain A/Puerto Rico/8/34. According to molecular modelling results, the lead compound 10a can bind to different sites in the stem region of the viral hemagglutinin.

Data-Driven Insights into the Transition-Metal-Catalyzed Asymmetric Hydrogenation of Olefins.

Project description:The transition-metal-catalyzed asymmetric hydrogenation of olefins is one of the key transformations with great utility in various industrial applications. The field has been dominated by the use of noble metal catalysts, such as iridium and rhodium. The reactions with the earth-abundant cobalt metal have increased only in recent years. In this work, we analyze the large amount of literature data available on iridium- and rhodium-catalyzed asymmetric hydrogenation. The limited data on reactions using Co catalysts are then examined in the context of Ir and Rh to obtain a better understanding of the reactivity pattern. A detailed data-driven study of the types of olefins, ligands, and reaction conditions such as solvent, temperature, and pressure is carried out. Our analysis provides an understanding of the literature trends and demonstrates that only a few olefin-ligand combinations or reaction conditions are frequently used. The knowledge of this bias in the literature data toward a certain group of substrates or reaction conditions can be useful for practitioners to design new reaction data sets that are suitable to obtain meaningful predictions from machine-learning models.
