Analyzing global utilization and missed opportunities in debt-for-nature swaps with generative AI.

ABSTRACT: We deploy a prompt-augmented GPT-4 model to distill comprehensive datasets on the global application of debt-for-nature swaps (DNS), a pivotal financial tool for environmental conservation. Our analysis includes 195 nations and identifies 21 countries that have not yet used DNS before as prime candidates for DNS. A significant proportion demonstrates consistent commitments to conservation finance (0.86 accuracy as compared to historical swaps records). Conversely, 35 countries previously active in DNS before 2010 have since been identified as unsuitable. Notably, Argentina, grappling with soaring inflation and a substantial sovereign debt crisis, and Poland, which has achieved economic stability and gained access to alternative EU conservation funds, exemplify the shifting suitability landscape. The study's outcomes illuminate the fragility of DNS as a conservation strategy amid economic and political volatility.

Are debt-for-nature swaps scalable: Which nature, how much debt, and who pays?

Project description:With the ongoing sovereign debt and biodiversity crises in many emerging economies, applications of debt-for-nature swaps as a dual solution for sovereign debt and nature conservation have been re-emerging. We analyze how debt-for-nature swaps (DNS) can be scaled to protect biodiversity priority areas and reduce debt burden. We build a dataset for biodiversity conservation and debt restructuring in 67 countries at risk of sovereign debt distress and show that they hold over 22% of global biodiversity priority areas, 82.96% of which are unprotected. Furthermore, we show that for 35 of the 67 countries, using conservative cost estimates, 100% of unprotected biodiversity priority areas could be protected for a fraction of debt; for the remaining countries, applying DNS would allow the protection of 11-13% of currently unprotected biodiversity priority areas. By applying interdisciplinary research combining fundamental biodiversity and economic data and methods merging, the research contributes methodologically and practically to the understanding of debt-for-nature swaps for emerging economies.

Opportunities and challenges of diffusion models for generative AI.

Project description:Diffusion models, a powerful and universal generative artificial intelligence technology, have achieved tremendous success and opened up new possibilities in diverse applications. In these applications, diffusion models provide flexible high-dimensional data modeling, and act as a sampler for generating new samples under active control towards task-desired properties. Despite the significant empirical success, theoretical underpinnings of diffusion models are very limited, potentially slowing down principled methodological innovations for further harnessing and improving diffusion models. In this paper, we review emerging applications of diffusion models to highlight their sample generation capabilities under various control goals. At the same time, we dive into the unique working flow of diffusion models through the lens of stochastic processes. We identify theoretical challenges in analyzing diffusion models, owing to their complicated training procedure and interaction with the underlying data distribution. To address these challenges, we overview several promising advances, demonstrating diffusion models as an efficient distribution learner and a sampler. Furthermore, we introduce a new avenue in high-dimensional structured optimization through diffusion models, where searching for solutions is reformulated as a conditional sampling problem and solved by diffusion models. Lastly, we discuss future directions about diffusion models. The purpose of this paper is to provide a well-rounded exposure for stimulating forward-looking theories and methods of diffusion models.

Deep generative AI models analyzing circulating orphan non-coding RNAs enable detection of early-stage lung cancer.

Project description:Liquid biopsies have the potential to revolutionize cancer care through non-invasive early detection of tumors. Developing a robust liquid biopsy test requires collecting high-dimensional data from a large number of blood samples across heterogeneous groups of patients. We propose that the generative capability of variational auto-encoders enables learning a robust and generalizable signature of blood-based biomarkers. In this study, we analyze orphan non-coding RNAs (oncRNAs) from serum samples of 1050 individuals diagnosed with non-small cell lung cancer (NSCLC) at various stages, as well as sex-, age-, and BMI-matched controls. We demonstrate that our multi-task generative AI model, Orion, surpasses commonly used methods in both overall performance and generalizability to held-out datasets. Orion achieves an overall sensitivity of 94% (95% CI: 87%-98%) at 87% (95% CI: 81%-93%) specificity for cancer detection across all stages, outperforming the sensitivity of other methods on held-out validation datasets by more than ~ 30%.

Prioritizing debt conversion opportunities for marine conservation.

Project description:Incentivized debt conversion is a financing mechanism that can assist countries with a heavy debt burden to bolster their long-term domestic investment in nature conservation. The Nature Conservancy, an international conservation-based nongovernmental organization, is adapting debt conversions to support marine conservation efforts by small island developing states and coastal countries. Prioritizing debt conversion opportunities according to their potential return on investment can increase the impact and effectiveness of this finance mechanism. We developed guidance on how to do so with a decision-support approach that relies on a novel threat-based adaptation of cost-effectiveness analysis. We constructed scenarios by varying parameters of the approach, including enabling conditions, expected benefits, and threat classifications. Incorporating both abatable and unabatable threats affected priorities across planning scenarios. Similarly, differences in scenario construction resulted in unique solution sets for top priorities. We show how environmental organizations, private entities, and investment banks can adopt structured prioritization frameworks for making decisions about conservation finance investments, such as debt conversions. Our guidance can accommodate a suite of social, ecological, and economic considerations, making the approach broadly applicable to other conservation finance mechanisms or investment strategies that seek to establish a transparent process for return-on-investment decision-making.

Inconsistent Provider Testing Practices for Congenital Cytomegalovirus: Missed Diagnoses and Missed Opportunities.

Project description:Newborn congenital cytomegalovirus (cCMV) screening programs have been found to increase the rates of early diagnosis and treatment. In North America, newborn cCMV screening programs have not been widely implemented, leaving healthcare providers to rely on clinical suspicion alone to prompt testing. This study sought to examine healthcare providers' cCMV testing practices at a quaternary children's hospital. A retrospective review of the electronic health record was completed for eligible infants over a six-year period. Bivariate calculations and analyses were performed. Between 2014 and 2019, a total of 40,091 infants were cared for at the study institution, of which 178 were tested for cCMV and 10 infants were diagnosed with cCMV. Isolated small-for-gestational age was the most common indication (53/178) to prompt testing. Overall, the cCMV testing rate was 4.5 tests per 1000 infants, with a resulting diagnostic prevalence of 0.2 cases per 1000 infants, which is 15-fold lower than the expected prevalence. Providers relying on clinical suspicion alone are infrequently testing infants for cCMV, resulting in missed diagnoses and missed opportunities for treatment. Systematic cCMV screening practices may improve diagnosis, treatment, and childhood outcomes.

Empowering personalized pharmacogenomics with generative AI solutions.

Project description:ObjectiveThis study evaluates an AI assistant developed using OpenAI's GPT-4 for interpreting pharmacogenomic (PGx) testing results, aiming to improve decision-making and knowledge sharing in clinical genetics and to enhance patient care with equitable access.Materials and methodsThe AI assistant employs retrieval-augmented generation (RAG), which combines retrieval and generative techniques, by harnessing a knowledge base (KB) that comprises data from the Clinical Pharmacogenetics Implementation Consortium (CPIC). It uses context-aware GPT-4 to generate tailored responses to user queries from this KB, further refined through prompt engineering and guardrails.ResultsEvaluated against a specialized PGx question catalog, the AI assistant showed high efficacy in addressing user queries. Compared with OpenAI's ChatGPT 3.5, it demonstrated better performance, especially in provider-specific queries requiring specialized data and citations. Key areas for improvement include enhancing accuracy, relevancy, and representative language in responses.DiscussionThe integration of context-aware GPT-4 with RAG significantly enhanced the AI assistant's utility. RAG's ability to incorporate domain-specific CPIC data, including recent literature, proved beneficial. Challenges persist, such as the need for specialized genetic/PGx models to improve accuracy and relevancy and addressing ethical, regulatory, and safety concerns.ConclusionThis study underscores generative AI's potential for transforming healthcare provider support and patient accessibility to complex pharmacogenomic information. While careful implementation of large language models like GPT-4 is necessary, it is clear that they can substantially improve understanding of pharmacogenomic data. With further development, these tools could augment healthcare expertise, provider productivity, and the delivery of equitable, patient-centered healthcare services.

A multimodal generative AI copilot for human pathology.

Project description:Computational pathology1,2 has witnessed considerable progress in the development of both task-specific predictive models and task-agnostic self-supervised vision encoders3,4. However, despite the explosive growth of generative artificial intelligence (AI), there have been few studies on building general-purpose multimodal AI assistants and copilots5 tailored to pathology. Here we present PathChat, a vision-language generalist AI assistant for human pathology. We built PathChat by adapting a foundational vision encoder for pathology, combining it with a pretrained large language model and fine-tuning the whole system on over 456,000 diverse visual-language instructions consisting of 999,202 question and answer turns. We compare PathChat with several multimodal vision-language AI assistants and GPT-4V, which powers the commercially available multimodal general-purpose AI assistant ChatGPT-4 (ref. 6). PathChat achieved state-of-the-art performance on multiple-choice diagnostic questions from cases with diverse tissue origins and disease models. Furthermore, using open-ended questions and human expert evaluation, we found that overall PathChat produced more accurate and pathologist-preferable responses to diverse queries related to pathology. As an interactive vision-language AI copilot that can flexibly handle both visual and natural language inputs, PathChat may potentially find impactful applications in pathology education, research and human-in-the-loop clinical decision-making.

A generative AI-driven interactive listening assessment task.

Project description:IntroductionAssessments of interactional competence have traditionally been limited in large-scale language assessments. The listening portion suffers from construct underrepresentation, whereas the speaking portion suffers from limited task formats such as in-person interviews or role plays. Human-delivered tasks are challenging to administer at large scales, while automated assessments are typically very narrow in their assessment of the construct because they have carried over the limitations of traditional paper-based tasks to digital formats. However, computer-based assessments do allow for more interactive, automatically administered tasks, but come with increased complexity in task creation. Large language models present new opportunities for enhanced automated item generation (AIG) processes that can create complex content types and tasks at scale that support richer assessments.MethodsThis paper describes the use of such methods to generate content at scale for an interactive listening measure of interactional competence for the Duolingo English Test (DET), a large-scale, high-stakes test of English proficiency. The Interactive Listening task assesses test takers' ability to participate in a full conversation, resulting in a more authentic assessment of interactive listening ability than prior automated assessments by positing comprehension and interaction as purposes of listening.Results and discussionThe results of a pilot of 713 tasks with hundreds of responses per task, along with the results of human review, demonstrate the feasibility of a human-in-the-loop, generative AI-driven approach for automatic creation of complex educational assessments at scale.

Reinvent 4: Modern AI-driven generative molecule design.

Project description:REINVENT 4 is a modern open-source generative AI framework for the design of small molecules. The software utilizes recurrent neural networks and transformer architectures to drive molecule generation. These generators are seamlessly embedded within the general machine learning optimization algorithms, transfer learning, reinforcement learning and curriculum learning. REINVENT 4 enables and facilitates de novo design, R-group replacement, library design, linker design, scaffold hopping and molecule optimization. This contribution gives an overview of the software and describes its design. Algorithms and their applications are discussed in detail. REINVENT 4 is a command line tool which reads a user configuration in either TOML or JSON format. The aim of this release is to provide reference implementations for some of the most common algorithms in AI based molecule generation. An additional goal with the release is to create a framework for education and future innovation in AI based molecular design. The software is available from https://github.com/MolecularAI/REINVENT4 and released under the permissive Apache 2.0 license. Scientific contribution. The software provides an open-source reference implementation for generative molecular design where the software is also being used in production to support in-house drug discovery projects. The publication of the most common machine learning algorithms in one code and full documentation thereof will increase transparency of AI and foster innovation, collaboration and education.
