Project description:Aeromonas veronii (A. veronii) is a ubiquitous bacterium in terrestrial and aquatic environments. It has a significant impact on animal and human health, with it becoming an emerging crucial pathogen worldwide. However, there have been no reports of mute swan infections. In the present study, after an observation of pathological changes, one bacterial strain isolated from a dead migratory mute swan was identified as A. veronii HNZZ-1/2022 based on its morphology, matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS), and sequence analysis of the 16S rRNA and gyrB genes. To explore its pathogenicity, virulence gene detection and a gosling infection experiment were subsequently carried out, respectively. Six virulence genes for cytotonic enterotoxins (alt), lateral elastase (ela), lipase (lip), cytotoxic enterotoxin (act), aerolysin (aerA), and polar flagellin (fla) were present in the template DNA of A. veronii HNZZ-1/2022. Experimentally infected goslings exhibited hemorrhages of various different degrees in multiple organs. The half-maximal lethal dose (LD50) value of A. veronii strain HNZZ-1/2022 was estimated to be 3.48 × 108 colony forming units (CFUs) per mL for goslings. An antimicrobial susceptibility test showed that the A. veronii HNZZ-1/2022 strain was resistant to meropenem, ampicillin, and enrofloxacin. To date, this is the first report of A. veronii in migratory mute swans, thus expanding the currently known host spectrum. These results suggest that the migratory mute swan is a new host for A. veronii and demonstrate the need for extensive surveillance and research of A. veronii to minimize its transmission between animals, the environment, and humans.

Project description:Genetic variance in characters under natural selection in natural populations determines the way those populations respond to that selection. Whether populations show temporal and/or spatial constancy in patterns of genetic variance and covariance is regularly considered, as this will determine whether selection responses are constant over space and time. Much less often considered is whether characters show differing amounts of genetic variance over the life-history of individuals. Such age-specific variation, if present, has important potential consequences for the force of natural selection and for understanding the causes of variation in quantitative characters. Using data from a long-term study of the mute swan Cygnus olor, we report the partitioning of phenotypic variance in timing of breeding (subject to strong natural selection) into component parts over 12 different age classes. We show that the additive genetic variance and heritability of this trait are strongly age-dependent, with higher additive genetic variance present in young and, particularly, old birds, but little evidence of any genetic variance for birds of intermediate ages. These results demonstrate that age can have a very important influence on the components of variation of characters in natural populations, and consequently that separate age classes cannot be assumed to be equivalent, either with respect to their evolutionary potential or response.

Project description:ObjectiveDuck virus enteritis is a severe viral disease that kills ducks and swans worldwide. The clinical manifestations, gross pathology, molecular detection, and characterization of the duck virus enteritis virus (DVEV) in Australian black swans at a safari park in Bangladesh were described in this case report.Materials and methodsOn a safari park in Bangladesh, an Australian black swan flock exhibited clinical signs of anorexia, greenish watery diarrhea, increased thirst, partial paralysis, and death. Postmortem examinations of deceased swans revealed extensive pathological abnormalities in the trachea, liver, and spleen. To isolate DVEV, a viral inoculum produced from the liver and spleen of dead swans was implanted into 9-13-day-old embryonated duck eggs via the chorioallantoic membrane (CAM) route. DVEV was confirmed using a polymerase chain reaction (PCR) assay. Phylogenetic analysis was used to determine the genetic relationship between the DVEV isolates from Australian black swans, and 16 DVEV isolates previously described in the GenBank.ResultsHemorrhage was noted in the annular ring of the trachea, as well as an enlarged and hemorrhagic liver and spleen. The PCR assay amplified a 446-bp fragment of the DVEV DNA polymerase gene in the liver, spleen, and CAM homogenates. The phylogenetic analysis found that the DVEV isolates from swans were comparable to those from Bangladesh, India, Vietnam, China, Germany, the USA, and Egypt.ConclusionAccording to the findings of this study, the DVEV was the cause of illness and mortality in an Australian black swan flock.

Project description:Identification and analysis of the avian reovirus from black swan. Isolation of the strain through the chorioallantoic membrane route of duck embryos, identified through transmission electron microscopy and RT-PCR based on the ARV S2 gene. The complete genome of the ARV strain was obtained using next-generation sequencing technology. The isolated strain of ARV was named CD200801 and was identified through transmission electron microscopy and RT-PCR based on the ARV S2 gene. Experimental infection with CD200801 resulted in the death of ducklings with serious spleen and liver focal necrosis. BLAST analysis of CD200801 sequences showed a 35.5 to 98.6% similarity to a novel duck reovirus that was isolated in recent years. Phylogenetic analysis revealed that CD200801 was closely related to ARV isolates YL, GX-Y7, and XT-18. We report the first avian reovirus infection in the black swan. This study provides important new insights into the evolutionary relationships among different ARV strains and highlights the need for continued surveillance and monitoring of these viruses in both domestic and wild bird flocks. These findings have significant implications for the development of effective strategies for disease prevention and control in the poultry industry.

Project description:There is growing recognition of the threat posed to wildlife by pollutants. Waterbirds are robust bioindicators of ecosystem health, and metal toxicity is a threat to these species in waterways worldwide. Urban waterbirds are likely to be at the highest risk of heavy metal exposure, but this issue has not been widely explored in Australia. Our aim was to estimate contemporary heavy metal exposure in a sedentary urban waterbird population: black swans (Cygnus atratus) inhabiting an inner-city wetland in one of Australia's largest cities, Melbourne. To investigate the physiological implications of legacy heavy metal exposure in these birds, we quantified blood biochemistry profiles and examined their relationships with metal concentrations in feathers. We caught 15 swans in 2021 and took feather samples to measure the concentration of eight heavy metals (chromium (Cr), manganese (Mn), iron (Fe), nickel (Ni), copper (Cu), zinc (Zn), lead (Pb), and mercury (Hg)), and blood samples to measure the concentration of 13 plasma analytes. Multivariate regression analysis revealed few associations between heavy metals and biochemistry markers, and no differences between sexes or age classes. This study presents a baseline dataset of these contaminants and blood biochemical profiles of swans at this wetland that can be used for future monitoring and is an important step toward a better understanding of the threat posed by heavy metals to Australian urban waterbirds.

Project description:Bluetongue virus (BTV), transmitted by midges (Culicoides sp), is distributed worldwide and causes disease in ruminants. In particular, BT can be a debilitating disease in sheep causing serious trade and socio-economic consequences at both local and global levels. Across Australia, a sentinel cattle herd surveillance program monitors the BTV activity. Prior to 2014, BTV-1, -2, -3, -7, -9, -15, -16, -20, -21 and -23 had been isolated in Australia, but no bluetongue disease has occurred in a commercial Australian flock. We routinely use a combination of serology, virus isolation, RT-PCR and next generation and conventional nucleotide sequencing technologies to detect and phylogenetically characterize incursions of novel BTV strains into Australia. Screening of Northern Territory virus isolates in 2015 revealed BTV-5, a serotype new to Australia. We derived the complete genome of this isolate and determined its phylogenetic relationship with exotic BTV-5 isolates. Gene segments 2, 6, 7 and 10 exhibited a close relationship with the South African prototype isolate RSArrrr/5. This was the first Australian isolation of a Western topotype of segment 10. Serological surveillance data highlighted the antigenic cross-reactivity between BTV-5 and BTV-9. Phylogenetic investigation of segments 2 and 6 of these serotypes confirmed their unconventional relationships within the BTV serogroup. Our results further highlighted a need for a revision of the current serologically based system for BTV strain differentiation and importantly, implied a potential for genome segments of pathogenic Western BTV strains to rapidly enter Southeast Asia. This emphasized a need for continued high-level surveillance of vectors and viruses at strategic locations in the north of Australia The expansion of routine characterization and classification of BTV to a whole genome approach is recommended, to better monitor the presence and level of establishment of novel Western topotype segments within the Australian episystem.

Project description:The Yellow River National Wetland in Baotou, China is an important resting and energy replenishment place for many migratory birds, such as tundra swan (Cygnus columbianus). The energy supply of food available at stopover sites plays an important role in the life cycle of migratory birds. In order to understand diet composition and energy supply of tundra swans for further protection of them, in this study, fecal of tundra swans (C. columbianus) were collected and fecal microhistological analysis was conducted to analyze the feeding habits and the energy supply. Results showed that: (1) tundra swans (C. columbianus) mainly fed on twelve species of plants from five families, including corn (Zea mays), quinoa (Chenopodium album) and rice (Oryza sativa), this is related to local crops and abundant plants. (2) The energy provided by crops to tundra swans (C. columbianus) was significantly higher than other abundant plants in wetlands (P < 0.05), corn and rice were the most consumed food, and other abundant wetland plants play complementary roles. (3) The daily energy intake of tundra swans (C. columbianus) was much higher than their daily energy consumption, the daily net energy intake of tundra swans (C. columbianus) was 855.51 ± 182.88 kJ (mean ± standard deviations). This suggested that the wetland provides energy for continue migrating to the tundra swan (C. columbianus). For further protection of tundra swans (C. columbianus) and other migratory birds, the Baotou Yellow River National Wetland environment and the surrounding farmland habitat should be protected.

Project description:Wild birds and waterfowl serve as the natural reservoirs of avian influenza viruses (AIVs). When AIVs originating from wild birds cross species barriers to infect mammals or humans, they pose a significant threat to public health. The H12 subtype of AIVs primarily circulates in wild birds, with relatively few isolates reported worldwide, and the evolutionary and biological characteristics of H12 subtype AIVs remain largely unknown. In this study, we analyzed the spatiotemporal distribution of H12 subtype AIVs worldwide and conducted a comprehensive investigation into the evolutionary and biological characteristics of an H12N2 virus isolated from a whooper swan in Central China. Phylogenetic analysis revealed that the H12N2 isolate belongs to the Eurasian lineage, with its HA gene likely originating from a duck-derived H12N5 virus and its NA gene potentially derived from an H9N2 virus, indicating that it is a complex reassorted virus. Animal experiments in domestic ducks and chickens demonstrated that the virus replicates at low levels in the respiratory tract of poultry and exhibits moderate horizontal transmission in ducks. However, it is capable of efficient horizontal transmission in chickens. Mouse infection experiments revealed that the virus could be detected in the nasal turbinates and lungs of mice, indicating that the H12N2 virus can infect mice without prior adaptation. In vitro studies revealed that the virus replicates efficiently in MDCK cells, with significantly higher titers than those in DF1 cells. These findings, combined with the mouse infection results, suggest that the H12N2 virus poses a potential risk of mammalian infection. This study provides valuable insights regarding the characteristics of the H12N2 virus and highlights the importance of ongoing surveillance and risk assessment of AIVs originating from wild birds.
