Fluid ejections in nature.

ABSTRACT: From microscopic fungi to colossal whales, fluidic ejections are a universal and intricate phenomenon in biology, serving vital functions such as animal excretion, venom spraying, prey hunting, spore dispersal, and plant guttation. This review delves into the complex fluid physics of ejections across various scales, exploring both muscle-powered active systems and passive mechanisms driven by gravity or osmosis. We introduce a framework using dimensionless numbers to delineate transitions from dripping to jetting and elucidate the governing forces. Highlighting the understudied area of complex fluid ejections, this work not only rationalizes the biophysics involved but also uncovers potential engineering applications in soft robotics, additive manufacturing, and drug delivery. By bridging biomechanics, the physics of living systems, and fluid dynamics, this review offers valuable insights into the diverse world of fluid ejections and paves the way for future bioinspired research across the spectrum of life.

Turn on the super-elastic collision nature of coronal mass ejections through low approaching speed.

Project description:It has been proved from the observations and numerical simulations that the collision between solar coronal mass ejections (CMEs), the largest plasmoids in the heliosphere, could be super-elastic. This finding suggests that the CMEs' magnetic energy and thermal energy could be converted into kinetic energy through a more efficient way. However CME collisions are not always super-elastic, which means that this distinct property of plasmoids is probably excited conditionally. As the first attempt, we carry out a series of three-dimensional numerical experiments, and establish a diagram showing the dependence of the collision nature on the CME speed and k-number, the ratio of the CME's kinetic energy to the CME's total energy. It is found that the super-elastic nature of CMEs appears at the relatively low approaching speed, and most of the previous case studies are in agreement with this diagram. Our study firmly advances the understanding of the super-elastic property of plasmoids, and does give us new clues to deeply understand why and how the magnetic energy and/or thermal energy of the colliding plasmoids can be converted into kinetic energy in such an efficient way.

The source and engine of coronal mass ejections.

Project description:Coronal mass ejections (CMEs) are large-scale expulsions of coronal plasma and magnetic field propagating through the heliosphere. Because CMEs are observed by white-light coronagraphs which, by design, occult the solar disc, supporting disc observations (e.g. in EUV, soft X-rays, Halpha and radio) must be employed for the study of their source regions and early development phases. We review the key properties of CME sources and highlight a certain causal sequence of effects that may occur whenever a strong (flux-massive and sheared) magnetic polarity inversion line develops in the coronal base of eruptive active regions (ARs). Storing non-potential magnetic energy and helicity in a much more efficient way than ARs lacking strong polarity inversion lines, eruptive regions engage in an irreversible course, making eruptions inevitable and triggered when certain thresholds of free energy and helicity are crossed. This evolution favours the formation of pre-eruption magnetic flux ropes. We describe the steps of this plausible path to sketch a picture of the pre-eruptive phase of CMEs that may apply to most events, particularly the ones populating the high end of the energy/helicity distribution, that also tend to have the strongest space-weather implications. Ion-dependent dynamics of DNA ejections for bacteriophage lambda.

Project description:We studied the control parameters that govern the dynamics of in vitro DNA ejection in bacteriophage lambda. Previous work demonstrated that bacteriophage DNA is highly pressurized, and this pressure has been hypothesized to help drive DNA ejection. Ions influence this process by screening charges on DNA; however, a systematic variation of salt concentrations to explore these effects has not been undertaken. To study the nature of the forces driving DNA ejection, we performed in vitro measurements of DNA ejection in bulk and at the single-phage level. We present measurements on the dynamics of ejection and on the self-repulsion force driving ejection. We examine the role of ion concentration and identity in both measurements, and show that the charge of counterions is an important control parameter. These measurements show that the mobility of ejecting DNA is independent of ionic concentrations for a given amount of DNA in the capsid. We also present evidence that phage DNA forms loops during ejection, and confirm that this effect occurs using optical tweezers.

Swarm Hunting and Cluster Ejections in Chemically Communicating Active Mixtures.

Project description:A large variety of microorganisms produce molecules to communicate via complex signaling mechanisms such as quorum sensing and chemotaxis. The biological diversity is enormous, but synthetic inanimate colloidal microswimmers mimic microbiological communication (synthetic chemotaxis) and may be used to explore collective behaviour beyond the one-species limit in simpler setups. In this work we combine particle based and continuum simulations as well as linear stability analyses, and study a physical minimal model of two chemotactic species. We observed a rich phase diagram comprising a "hunting swarm phase", where both species self-segregate and form swarms, pursuing, or hunting each other, and a "core-shell-cluster phase", where one species forms a dense cluster, which is surrounded by a (fluctuating) corona of particles from the other species. Once formed, these clusters can dynamically eject their core such that the clusters almost turn inside out. These results exemplify a physical route to collective behaviours in microorganisms and active colloids, which are so-far known to occur only for comparatively large and complex animals like insects or crustaceans.

Bioinspired Hemostatic Strategy via Pulse Ejections for Severe Bleeding Wounds.

Project description:Efficient hemostasis during emergency trauma with massive bleeding remains a critical challenge in prehospital settings. Thus, multiple hemostatic strategies are critical for treating large bleeding wounds. In this study, inspired by bombardier beetles to eject toxic spray for defense, a shape-memory aerogel with an aligned microchannel structure was proposed, employing thrombin-carrying microparticles loaded as a built-in engine to generate pulse ejections for enhanced drug permeation. Bioinspired aerogels, after contact with blood, can rapidly expand inside the wound, offering robust physical barrier blocking, sealing the bleeding wound, and generating a spontaneous local chemical reaction causing an explosive-like generation of CO2 microbubbles, which provide propulsion thrust to accelerate burst ejection from arrays of microchannels for deeper and faster drug diffusion. The ejection behavior, drug release kinetics, and permeation capacity were evaluated using a theoretical model and experimentally demonstrated. This novel aerogel showed remarkable hemostatic performance in severely bleeding wounds in a swine model and demonstrated good degradability and biocompatibility, displaying great potential for clinical application in humans.

Wound Healing Fluid Reflects the Inflammatory Nature and Aggressiveness of Breast Tumors.

Project description:Wound healing fluid that originates from breast surgery increases the aggressiveness of cancer cells that remain after the surgery. We determined the effects of the extent of surgery and tumor-driven remodeling of the surrounding microenvironment on the ability of wound-healing to promote breast cancer progression. In our analysis of a panel of 34 cytokines, chemokines, and growth factors in wound healing fluid, obtained from 27 breast carcinoma patients after surgery, the levels of several small molecules were associated with the extent of cellular damage that was induced by surgery. In addition, the composition of the resulting wound healing fluid was associated with molecular features of the removed tumor. Specifically, IP-10, IL-6, G-CSF, osteopontin, MIP-1a, MIP-1b, and MCP1-MCAF were higher in more aggressive tumors. Altogether, our findings indicate that the release of factors that are induced by removal of the primary tumor and subsequent wound healing is influenced by the extent of damage due to surgery and the reactive stroma that is derived from the continuously evolving network of interactions between neoplastic cells and the microenvironment, based on the molecular characteristics of breast carcinoma cells.

Real-time observations of single bacteriophage lambda DNA ejections in vitro.

Project description:The physical, chemical, and structural features of bacteriophage genome release have been the subject of much recent attention. Many theoretical and experimental studies have centered on the internal forces driving the ejection process. Recently, Mangenot et al. [Mangenot S, Hochrein M, Rädler J, Letellier L (2005) Curr Biol 15:430-435.] reported fluorescence microscopy of phage T5 ejections, which proceeded stepwise between DNA nicks, reaching a translocation speed of 75 kbp/s or higher. It is still unknown how high the speed actually is. This paper reports real-time measurements of ejection from phage lambda, revealing how the speed depends on key physical parameters such as genome length and ionic state of the buffer. Except for a pause before DNA is finally released, the entire 48.5-kbp genome is translocated in approximately 1.5 s without interruption, reaching a speed of 60 kbp/s. The process gives insights particularly into the effects of two parameters: a shorter genome length results in lower speed but a shorter total time, and the presence of divalent magnesium ions (replacing sodium) reduces the pressure, increasing ejection time to 8-11 s. Pressure caused by DNA-DNA interactions within the head affects the initiation of ejection, but the close packing is also the dominant source of friction: more tightly packed phages initiate ejection earlier, but with a lower initial speed. The details of ejection revealed in this study are probably generic features of DNA translocation in bacteriophages and have implications for the dynamics of DNA in other biological systems.

Ex vivo biomechanical characterization of syringe-needle ejections for intracerebral cell delivery.

Project description:Intracerebral implantation of cell suspensions is finding its clinical translation with encouraging results in patients with stroke. However, the survival of cells in the brain remains poor. Although the biological potential of neural stem cells (NSCs) is widely documented, the biomechanical effects of delivering cells through a syringe-needle remain poorly understood. We here detailed the biomechanical forces (pressure, shear stress) that cells are exposed to during ejection through different sized needles (20G, 26G, 32G) and syringes (10, 50, 250 µL) at relevant flow rates (1, 5, 10 µL/min). A comparison of 3 vehicles, Phosphate Buffered Saline (PBS), Hypothermosol (HTS), and Pluronic, indicated that less viscous vehicles are favorable for suspension with a high cell volume fraction to minimize sedimentation. Higher suspension viscosity was associated with greater shear stress. Higher flow rates with viscous vehicle, such as HTS reduced viability by ~10% and also produced more apoptotic cells (28%). At 5 µL/min ejection using a 26G needle increased neuronal differentiation for PBS and HTS suspensions. These results reveal the biological impact of biomechanical forces in the cell delivery process. Appropriate engineering strategies can be considered to mitigate these effects to ensure the efficacious translation of this promising therapy.

Onset and nature of flow-induced vibrations in cerebral aneurysms via fluid-structure interaction simulations.

Project description:Clinical, experimental, and recent computational studies have demonstrated the presence of wall vibrations in cerebral aneurysms, thought to be induced by blood flow instability. These vibrations could induce irregular, high-rate deformation of the aneurysm wall, and potentially disrupt regular cell behavior and promote deleterious wall remodeling. In order to elucidate, for the first time, the onset and nature of such flow-induced vibrations, in this study we imposed a linearly increasing flow rate on high-fidelity fluid-structure interaction models of three anatomically realistic aneurysm geometries. Prominent narrow-band vibrations in the range of 100-500 Hz were found in two out of the three aneurysm geometries tested, while the case that did not exhibit flow instability did not vibrate. Aneurysm vibrations consisted mostly of fundamental modes of the entire aneurysm sac, with the vibrations exhibiting more frequency content at higher frequencies than the flow instabilities driving those vibrations. The largest vibrations occurred in the case which exhibited strongly banded fluid frequency content, and the vibration amplitude was highest when the strongest fluid frequency band was an integer multiple of one of the natural frequencies of the aneurysm sac. Lower levels of vibration occurred in the case which exhibited turbulent-like flow with no distinct frequency bands. The current study provides a plausible mechanistic explanation for the high-frequency sounds observed in cerebral aneurysms, and suggests that narrow-band (vortex-shedding type) flow might stimulate the wall more, or at least at lower flow rates, than broad-band, turbulent-like flow.
