ABSTRACT: Global nocturnal temperatures are rising more rapidly than daytime temperatures and have a large effect on crop productivity. In particular, stomatal conductance at night (g_sn ) is surprisingly poorly understood and has not been investigated despite constituting a significant proportion of overall canopy water loss. Here, we present the results of 3 yr of field data using 12 spring Triticum aestivum genotypes which were grown in NW Mexico and subjected to an artificial increase in night-time temperatures of 2°C. Under nocturnal heating, grain yields decreased (1.9% per 1°C) without significant changes in daytime leaf-level physiological responses. Under warmer nights, there were significant differences in the magnitude and decrease in g_sn , values of which were between 9 and 33% of daytime rates while respiration appeared to acclimate to higher temperatures. Decreases in grain yield were genotype-specific; genotypes categorised as heat tolerant demonstrated some of the greatest declines in yield in response to warmer nights. We conclude the essential components of nocturnal heat tolerance in wheat are uncoupled from resilience to daytime temperatures, raising fundamental questions for physiological breeding. Furthermore, this study discusses key physiological traits such as pollen viability, root depth and irrigation type may also play a role in genotype-specific nocturnal heat tolerance.

Project description:Quinoa high nutritive value increases interest worldwide, especially as a crop that could potentially feature in different cropping systems, however, climate change, particularly rising temperatures, challenges this and other crop species. Currently, only limited knowledge exists regarding the grain yield and other key traits response to higher temperatures of this crop, especially to increased night temperatures. In this context, the main objective of this study was to evaluate the effect of increased night temperature on quinoa yield, grain number, individual grain weight and processes involved in crop growth under the environmental conditions (control treatment) and night thermal increase at two phases: flowering (T1) and grain filling (T2) in southern Chile. A commercial genotype, Regalona, and a quinoa accession (Cod. BO5, N°191, grain bank from Semillas Baer, hereby referred to as Accession) were used, due to their adaptability to Southern Chilean conditions and contrasting grain yield potential, grain weight and size of plants. Temperature was increased ≈4°C above the ambient from 8 pm until 9 am the next morning. Control treatments reached a high grain yield (600 and 397 g m-2, i.e., Regalona and Accession). Temperature increase reduced grain yield by 31% under T1 treatment and 12% when under T2 in Regalona and 23 and 26% in Accession, respectively. Aboveground biomass was negatively affected by the thermal treatments and a positive linear association was found between grain yield and aboveground biomass across treatments. By contrast, the harvest index was unaffected either by genotype, or by thermal treatments. Grain number was significantly affected between treatments and this key trait was linearly associated with grain yield. On the other hand, grain weight showed a narrow range of variation across treatments. Additionally, leaf area index was not affected, but significant differences were found in SPAD values at the end of T1 treatment, compared to control. Little change was found in the harvest index, individual grain weight, grain protein content or water soluble carbohydrates in response to the increased night temperature in this crop.

Project description:The diurnal and seasonal temperature rising patterns "asymmetric warming," plays an important role in crop distribution and productivity. Asymmetric warming during the early growth periods of winter wheat (Triticum aestivum L.) profoundly affects vegetative growth and post-anthesis grain productivity, but the underlying physiological mechanism is still unclear. We conducted field experiments from 2012 to 2014 using two wheat cultivars, namely, Yangmai-13 (vernal type) and Yannong-19 (semi-winter type), to investigate the influences of night-warming during the winter (warming by 1.56-1.67°C from tillering to jointing) or during the spring (warming by 1.78-1.92°C from jointing to booting) on post-anthesis physiological activities and grain-filling processes. Both night-warming treatments enhanced the source activity by increasing flag leaf area, chlorophyll content, and photosynthetic capability in both cultivars compared with those of the control. The night-warming treatments caused an increase in the antioxidant activities of superoxide dismutase (SOD), peroxidase, and catalase (CAT) in the flag leaves of both cultivars, while ROS contents such as superoxide anion radical ([Formula: see text]) and hydrogen peroxide (H2O2) decreased. Moreover, the expression levels of Rubisco activase B (RcaB), major chlorophyll a/b-binding protein (Cab), chloroplast Cu/Zn superoxide dismutase (Cu/Zn-SOD), mitochondrial manganese superoxide dismutase (Mn-SOD), and CAT genes were upregulated at anthesis and were associated with higher photosynthetic capacity and antioxidant activities. Furthermore, night-warming improved sink activities by increasing the concentrations of grain indole-3-acetic acid and cytokinins as well as the sucrose synthase activity for both cultivars. Winter night-warming showed greater potential for improving source strength and grain filling, with consistent performance in both cultivars compared with that of spring night-warming. We concluded form these results that night-warming can improve source and sink capacities in winter wheat, and winter night-warming has greater advantages in this respect than does spring warming.

Project description:MicroRNAs (miRNAs) mediate the degradation of target mRNA and inhibit mRNA translation to regulate gene expression at the transcriptional and post-transcriptional levels in response to environmental stress in plants. We characterized the post-transcriptional mechanism by deep sequencing small RNA (sRNA) to examine how miRNAs were involved in low night temperature (LNT) stress in tomato and whether the molecular mechanism depended on the abscisic acid (ABA) signaling pathway. We annotated conserved miRNAs and novel miRNAs with four sRNA libraries composed of wild-type (WT) tomato plants and ABA-deficient mutant (sit) plants under normal growth and LNT stress conditions. Reverse genetics analysis suggested that miR162 participated in LNT resistance and the ABA-dependent signaling pathway in tomato. miR162-overexpressing (pRI-miR162) and miR162-silenced (pRNAi-miR162) transgenic tomato plants were generated to evaluate miR162 functions in response to LNT stress. miR162 deficiency exhibited high photosynthetic capacity and regulated stomatal opening, suggesting negative regulation of miR162 in the ABA-dependent signaling pathway in response to LNT stress. As feedback regulation, miR162 positively regulated ABA to maintain homeostasis of tomato under diverse abiotic stresses. The mRNA of DICER-LIKE1 (DCL1) was targeted by miR162, and miR162 inhibited DCL1 cleavage in LNT response, including the regulation of miRNA160/164/171a and their targets. The DCL1-deficient mutants (dcl1) with CRISPR/Cas9 prevented stomatal opening to influence photosynthesis in the ABA signaling pathway under LNT stress. Finally, we established the regulatory mechanism of ABA-miR162-DCL1, which systematically mediated cold tolerance in tomato. This study suggests that post-transcriptional modulators acted as systemic signal responders via the stress hormone signaling pathway, and the model at the post-transcriptional level presents a new direction for research in plant abiotic stress resistance.
