Revealing the Diversity and Complex Relationships of Croatian Olive Germplasm.

ABSTRACT: Olive growing in Croatia has a long tradition and is of great economic and social impact. The present study includes a set of 108 tree samples (88 samples corresponding to 60 presumed cultivars and 20 trees of unnamed ones) collected from 27 groves in the entire olive growing area, and is the most comprehensive survey to be conducted in Croatia. The genetic diversity, relationships, and structures of olive plants were studied using eight microsatellite loci. All loci were polymorphic and revealed a total of 90 alleles. A total of 74 different genotypes were detected that were subjected to further diversity and genetic relationship studies. The Fitch-Margoliash tree and Bayesian analysis of population structure revealed a complex relationship between the identified olive genotypes, which were clustered into three gene pools, indicating different origins of Croatian olive germplasms. Excluding the redundant germplasms, 44 different genotypes among the sampled trees of well-known cultivars and 16 new local germplasms were identified. In addition, we provide the etymology of 46 vernacular names, which confirms that the vast majority of traditional Croatian cultivars have common and widespread names. The results presented herein underline the importance of safeguarding local cultivars and conducting continuous surveys.

Project description:Landraces represent valuable genetic resources for breeding programmes to produce high-yielding varieties adapted to stressful environmental conditions. Although the common bean (Phaseolus vulgaris L.) is an economically important food legume for direct human consumption worldwide, common bean production in Croatia is based almost exclusively on landraces and there is no common bean breeding program. Information on phaseolin type and results of population structure and genetic diversity obtained by analysis of SSR and SNP markers, in combination with the morphological characterization of 174 accessions of 10 common bean landraces (morphotypes), enabled thorough classification of accessions. The accessions were classified into phaseolin type H1 ("S") of Mesoamerican origin and phaseolin types H2 ("H" or "C") and H3 ("T") of Andean origin. By applying distance- and model-based clustering methods to SSR markers, the accessions were classified into two clusters at K = 2 separating the accessions according to the centres of origin, while at K = 3, the accessions of Andean origin were further classified into two clusters of accessions that differed in phaseolin type (H2 and H3). Using SNP markers, model-based analysis of population structure was performed, the results of which were consistent with those of SSR markers. In addition, 122 accessions were assigned to 14 newly formed true-type morphogenetic groups derived from three different domestication events: (1) Mesoamerican (H1A) ("Biser", "Kukuruzar", "Tetovac", "Trešnjevac"), (2) Andean-indeterminate type (H2B1) ("Dan noć", "Sivi", "Puter", "Sivi prošarani", "Trešnjevac") and (3) Andean-determinate type (H3B2) ("Bijeli", "Dan noć", "Puter", "Trešnjevac", "Zelenčec"). The rest of the accessions could represent putative hybrids between morphogenetic groups. The differences between the true-type groups of accessions were further analysed based on nine quantitative traits, and the subsets of traits that best distinguish among centres of origin (A: Mesoamerican, B: Andean) and genetic groups (H1A, H2B1, H3B2) were proposed.

Project description:Olive (Olea europaea L.) is a major fruit crop in the Mediterranean Basin. Ex-situ olive management is essential to ensure optimal use of genetic resources in breeding programs. The Worldwide Olive Germplasm Bank of Córdoba (WOGBC), Spain, and Marrakech (WOGBM), Morocco, are currently the largest existing olive germplasm collections. Characterization, identification, comparison and authentication of all accessions in both collections could thus provide useful information for managing olive germplasm for its preservation, exchange within the scientific community and use in breeding programs. Here we applied 20 microsatellite markers (SSR) and 11 endocarp morphological traits to discriminate and authenticate 1091 olive accessions belonging to WOGBM and WOGBC (554 and 537, respectively). Of all the analyzed accessions, 672 distinct SSR profiles considered as unique genotypes were identified, but only 130 were present in both collections. Combining SSR markers and endocarp traits led to the identification of 535 cultivars (126 in common) and 120 authenticated cultivars. No significant differences were observed between collections regarding the allelic richness and diversity index. We concluded that the genetic diversity level was stable despite marked contrasts in varietal composition between collections, which could be explained by their different collection establishment conditions. This highlights the extent of cultivar variability within WOGBs. Moreover, we detected 192 mislabeling errors, 72 of which were found in WOGBM. A total of 228 genotypes as molecular variants of 74 cultivars, 79 synonyms and 39 homonyms as new cases were identified. Both collections were combined to define the nested core collections of 55, 121 and 150 sample sizes proposed for further studies. This study was a preliminary step towards managing and mining the genetic diversity in both collections while developing collaborations between olive research teams to conduct association mapping studies by exchanging and phenotyping accessions in contrasted environmental sites.

Project description:An extensive macrophyte field survey of running and standing waters was conducted from 2016 to 2021 at 786 sampling sites across Croatia as a part of the implementation of the Water Framework Directive. This survey is the first to present a comprehensive floristic catalogue of the freshwater bryoflora, along with an analysis of the distribution and diversity patterns on a national level. In all, 83 bryophyte species (68 mosses and 15 liverworts) were recorded in the 228 sites, with average species richness of 4.17 species per site. The most frequent species were Fontinalisantipyretica, Rhynchostegiumriparioides, Leptodictyumriparium and Cratoneuronfilicinum. The majority of the species encountered were rarely found, with over 70% of species recorded on less than 10 sampling sites and the majority of the species not being truly aquatic, rather being classified as facultative aquatics. The Dinaric Ecoregion, characterised by clean, cold, fast-flowing karstic rivers, especially in the Continental Subecoregion, supported higher freshwater bryophyte diversity than the lowland Pannonian Ecoregion, with mostly slow, eutrophic lowland watercourses with unstable sandy and gravelly alluvial sediments. Chorological comparison of Croatian eco- and subecoregions revealed the expected dominance of circumpolar and European elements, i.e. temperate chorotypes, as well as some biogeographical differences. The most frequent life forms were aquatic trailings and turfs. Amongst the recorded species, perennials and colonists were the most represented life strategies. The analysis of both the life-form and life-strategy spectra showed some differences amongst the Croatian regions, supporting the fact that the Dinaric Ecoregion provides more truly aquatic habitats and microhabitats suitable for the freshwater bryophytes, while in the Pannonian Ecoregion freshwater bryophytes dominantly inhabit the periodically submerged riparian zones, for example shaded lowland forest streams and rivulets or gently sloping margins of rivers and lakes.
