Discovery of Novel Potential Insecticide-Resistance Mutations in Spodoptera frugiperda.

ABSTRACT: The fall armyworm (FAW), Spodoptera frugiperda, is a worldwide agricultural pest that invaded China in 2018, and has developed resistance to multiple insecticides. The evolution of insecticide resistance is facilitated by mutations of target genes responsible for conferring resistance. In this study, amplicon sequencing analyzed 21 sites in six resistance genes. In addition to known mutations, unknown variants were also found, including novel variants: F290C (ace-1 gene, 0.1% frequency), I1040T/V (CHSA gene, 0.1% frequency), A309T (GluCl gene, 0.1% frequency), and I4790T/V (RyR gene, 0.1% frequency). Additionally, molecular docking was employed to investigate the impact of the aforementioned new mutations on insecticide binding to proteins. The analyses indicated that the binding abilities were reduced, similar to the resistance mutations that were reported, implying these novel mutations may confer transitional resistance. This study may provide a foundation for understanding the functions of these novel mutations in the evolutionary processes that drive the emergence of insecticide resistance in this invasive species.

Project description:BackgroundThe fall armyworm (FAW), Spodoptera frugiperda; J.E. Smith (Lepidoptera: Noctuidae), is now an economically important pest that causes huge losses to maize productivity in sub-Saharan Africa. Variations in sub-population genetics and the processes of rapid adaptation underpinning the invasion remain unclear. For this, the genetic identity and diversity of FAW populations in Uganda were revealed by sequencing 87 samples (collected across the country). Based on the partial mitochondrial cytochrome oxidase I (COI) gene polymorphisms, we further examined the mitochondrial haplotype configuration and compared the FAW in Uganda with sequences from other parts of the world. The molecular target for organophosphate and carbamate resistance, acetylcholinesterase, was also investigated.ResultsAnalysis of the partial COI gene sequences showed the presence of both rice (predominant) and corn strain haplotypes, with a haplotype diversity of 0.382. Based on the COI marker, pairwise difference distribution analyses, and neutrality tests, showed that the FAW populations in Uganda and the rest of Africa are evolving neutrally, but those in America and Asia are undergoing expansion. Our findings support observations that invasive FAW populations throughout the rest of Africa and Asia share a common origin. Sequencing of the S. frugiperda ace-1 gene revealed four amino acid substitutions, two of which (A201S and F290V) were previously shown to confer organophosphate resistance in both S. frugiperda and several other insect species. The other two previously reported new variations in positions g-396 and g-768, are presumed to be related to the development of insecticide resistance.ConclusionsThis research has increased our knowledge of the genetics of FAW in Uganda, which is critical for pest surveillance and the detection of resistance. However, due to the low gene polymorphism of COI, more evolutionary studies incorporating the Spodoptera frugiperda whole-genome sequence are required to precisely understand the FAW population dynamics, introduction paths, origin, and subsequent spread.

Project description:BackgroundCyproflanilide is a novel chemical that is already undergoing insecticide registration in China and has been categorized as a member of group 30 by the IRAC. Since it was first detected in 2019, the fall armyworm (FAW), Spodoptera frugiperda, has become a serious pest in China. Our laboratory and field efficacy trials indicated that cyproflanilide exhibits high larvicidal activity against FAW. However, the effect of cyproflanilide against FAW remains unknown. And it is worth exploring further before the cyproflanilide becomes commercially available.ResultsWe found larvae exposed to cyproflanilide had significantly shorter body length and higher death rates compared to control larvae. Additionally, we found surviving larvae had a significantly longer developmental period compared to control larvae. The potential molecular mechanisms of cyproflanilide against FAW were investigated using comparative transcriptomic analyses on larval samples subjected to three insecticide treatments, including cyproflanilide and two other commonly used insecticides against FAW in China, chlorantraniliprole and avermectin. We found that several subunits of the γ-aminobutyric acid receptor (GABAR), a possible target protein of cyproflanilide, were significantly up-regulated at the transcriptional level during cyproflanilide-induced stress. Additionally, between the control and cyproflanilide-treated samples, we identified 131 differentially expressed genes (DEGs) associated with detoxification metabolism. Of these, we found four P450 genes that were significantly up-regulated under cyproflanilide stress but were not DEGs when exposed to chlorantraniliprole and avermectin, or 23 other pesticides from previous reports. Furthermore, we discovered an interesting gene aggregation region for insect cuticle proteins (CPs) on the 18th chromosome, which is likely related to FAW cross-resistance to cyproflanilide and avermectin.ConclusionsOur results contribute to a greater understanding of the mechanisms by which cyproflanilide affects FAW. Additionally, we identified the similarities and differences in transcriptomic profiling of FAW between the novel insecticide cyproflanilide and two other commonly used insecticides.
