Project description:Purpose: Chitosan is a natural mucoadhesive polymer with antibacterial activity. In the present study, chitosan (CS) nanoparticles were investigated as a vehicle for delivery of antibiotic, ciprofloxacin hydrochloride. Methods: Ionotropic gelation method was used for preparation chitosan nanoparticles. The effects of various factors including concentration of CS, concentration of tripolyphosphate (TPP), and homogenization rate on the size of nanoparticles were studied. The effects of various mass ratios of CS to ciprofloxacin hydrochloride on the encapsulation efficiency of nanoparticles were assessed. Results: The particles prepared under optimal condition of 0.45% CS concentration, 0.45% TPP concentration and homogenizer rate at 6000 rpm, had 72 nm diameter. In these particles with 1:0.5 mass ratio of CS to ciprofloxacin hydrochloride, the encapsulation efficiency was 23%. The antibacterial activity of chitosan nanoparticles and ciprofloxacin-loaded nanoparticles against E.coli and S.aureus was evaluated by calculation of minimum inhibitory concentration (MIC). Results showed that MIC of ciprofloxacin loaded chitosan nanoparticles was 50% lower than that of ciprofloxacin hydrochloride alone in both of microorganism species. Nanoparticles without drug exhibited antibacterial activity at higher concentrations and MIC of them against E.coli and S.aureus was 177 and 277 µg/ml, respectively. Conclusion: Therefore chitosan nanoparticles could be applied as carrier for decreasing the dose of antibacterial agents in the infections.

Project description:It is common knowledge that pure alginate hydrogel is more likely to have weak mechanical strength, a lack of cell recognition sites, extensive swelling and uncontrolled degradation, and thus be unable to satisfy the demands of the ideal scaffold. To address these problems, we attempted to fabricate alginate/bacterial cellulose nanocrystals-chitosan-gelatin (Alg/BCNs-CS-GT) composite scaffolds using the combined method involving the incorporation of BCNs in the alginate matrix, internal gelation through the hydroxyapatite-d-glucono-δ-lactone (HAP-GDL) complex, and layer-by-layer (LBL) electrostatic assembly of polyelectrolytes. Meanwhile, the effect of various contents of BCNs on the scaffold morphology, porosity, mechanical properties, and swelling and degradation behavior was investigated. The experimental results showed that the fabricated Alg/BCNs-CS-GT composite scaffolds exhibited regular 3D morphologies and well-developed pore structures. With the increase in BCNs content, the pore size of Alg/BCNs-CS-GT composite scaffolds was gradually reduced from 200 μm to 70 μm. Furthermore, BCNs were fully embedded in the alginate matrix through the intermolecular hydrogen bond with alginate. Moreover, the addition of BCNs could effectively control the swelling and biodegradation of the Alg/BCNs-CS-GT composite scaffolds. Furthermore, the in vitro cytotoxicity studies indicated that the porous fiber network of BCNs could fully mimic the extracellular matrix structure, which promoted the adhesion and spreading of MG63 cells and MC3T3-E1 cells on the Alg/BCNs-CS-GT composite scaffolds. In addition, these cells could grow in the 3D-porous structure of composite scaffolds, which exhibited good proliferative viability. Based on the effect of BCNs on the cytocompatibility of composite scaffolds, the optimum BCNs content for the Alg/BCNs-CS-GT composite scaffolds was 0.2% (w/v). On the basis of good merits, such as regular 3D morphology, well-developed pore structure, controlled swelling and biodegradation behavior, and good cytocompatibility, the Alg/BCNs-CS-GT composite scaffolds may exhibit great potential as the ideal scaffold in the bone tissue engineering field.

Project description:The implementation of inorganic adsorbents for the removal of heavy metals from industrial effluents generates secondary waste. Therefore, scientists and environmentalists are looking for environmentally friendly adsorbents isolated from biobased materials for the efficient removal of heavy metals from industrial effluents. This study aimed to fabricate and characterize an environmentally friendly composite bio-sorbent as an initiative toward greener environmental remediation technology. The properties of cellulose, chitosan, magnetite, and alginate were exploited to fabricate a composite hydrogel bead. The cross linking and encapsulation of cellulose, chitosan, alginate, and magnetite in hydrogel beads were successfully conducted through a facile method without any chemicals used during the synthesis. Energy-dispersive X-ray analysis verified the presence of element signals of N, Ca, and Fe on the surface of the composite bio-sorbents. The appearance and peak's shifting at 3330-3060 cm-1 in the Fourier transform infrared spectroscopy analysis of the composite cellulose-magnetite-alginate, chitosan-magnetite-alginate, and cellulose-chitosan-magnetite-alginate suggested that there are overlaps of O-H and N-H and weak interaction of hydrogen bonding with the Fe3O4 particles. Material degradation, % mass loss, and thermal stability of the material and synthesized composite hydrogel beads were determined through thermogravimetric analysis. The onset temperature of the composite cellulose-magnetite-alginate, chitosan-magnetite-alginate, and cellulose-chitosan-magnetite-alginate hydrogel beads were observed to be lower compared to raw-material cellulose and chitosan, which could be due to the formation of weak hydrogen bonding resulting from the addition of magnetite Fe3O4. The higher mass residual of cellulose-magnetite-alginate (33.46%), chitosan-magnetite-alginate (37.09%), and cellulose-chitosan-magnetite-alginate (34.40%) compared to cellulose (10.94%) and chitosan (30.82%) after degradation at a temperature of 700 °C shows that the synthesized composite hydrogel beads possess better thermal stability, owing to the addition of magnetite and the encapsulation in the alginate hydrogel beads.

Project description:Due to their high entrapment efficiency, anodized titanium nanotubes (TiO2-NTs) are considered effective reservoirs for loading/releasing strong antibiotics whose systemic administration is associated with diverse and severe side-effects. In this study, TiO2-NTs were synthesized by anodic oxidation of titanium foils, and the effects of electrolyte percentage and viscosity on their dimensions were evaluated. It was found that as the water content increased from 15 to 30%, the wall thickness, length, and inner diameter of the NTs increase from 5.9 to 15.8 nm, 1.56 to 3.21 µm, and 59 to 84 nm, respectively. Ciprofloxacin, a highly potent antibiotic, was loaded into TiO2-NTs with a high encapsulation efficiency of 93%, followed by coating with different chitosan layers to achieve a sustained release profile. The prepared formulations were characterized by various techniques, such as scanning electron microscopy, differential scanning calorimetry, and contact measurement. In vitro release studies showed that the higher the chitosan layer count, the more sustained the release. Evaluation of antimicrobial activity of the formulation against two endodontic species from Peptostreptococcus and Fusobacterium revealed minimum inhibitory concentrations (MICs) of 1 µg/mL for the former and the latter. To summarize, this study demonstrated that TiO2-NTs are promising reservoirs for drug loading, and that the chitosan coating provides not only a sustained release profile, but also a synergistic antibacterial effect.

Project description:Cannabinoids present in Cannabis sativa are increasingly used in medicine due to their therapeutic potential. Moreover, the synergistic interaction between different cannabinoids and other plant constituents has led to the development of full-spectrum formulations for therapeutic treatments. In this work, the microencapsulation of a full-spectrum extract via vibration microencapsulation nozzle technique using chitosan-coated alginate is proposed to obtain an edible pharmaceutical-grade product. The suitability of microcapsules was assessed by their physicochemical characterization, long-term stability in three different storage conditions and in vitro gastrointestinal release. The synthetized microcapsules contained mainly ∆9-tetrahydrocannabinol (THC)-type and cannabinol (CBN)-type cannabinoids and had a mean size of 460 ± 260 µm and a mean sphericity of 0.5 ± 0.3. The stability assays revealed that capsules should be stored only at 4 °C in darkness to maintain their cannabinoid profile. In addition, based on the in vitro experiments, a fast intestinal release of cannabinoids ensures a medium-high bioaccessibility (57-77%) of therapeutically relevant compounds. The full characterization of microcapsules indicates that they could be used for the design of further full-spectrum cannabis oral formulations.

Project description:BackgroundHydroxy-α-Sanshool (HAS) possesses various pharmacological properties, such as analgesia and regulating gastrointestinal function. However, the low oral bioavailability of HAS has limited its oral delivery in clinical application.Methods and resultsTo enhance its oral bioavailability, a nanocomposite delivery system based on chitosan (CH, as the polycation) and sodium alginate (SA, as the polyanion) was prepared using a layer-by-layer coating technique. The morphology, thermal behavior and Fourier transform infrared spectrum (FTIR) showed that the obtained sodium alginate/chitosan-coated HAS-loaded liposomes (SA/CH-HAS-LIP) with core-shell structures have been successfully covered with polymers. When compared with HAS-loaded liposomes (HAS-LIP), SA/CH-HAS-LIP displayed obvious pH sensitivity and a sustained-release behavior in in vitro studies, which fitted well to Weibull model. In vivo, the half-life of HAS from SA/CH-HAS-LIP remarkably extended after oral administration compared to the free drug. Additionally, it allowed a 4.6-fold and 4.2-fold increase in oral bioavailability, respectively, compared with free HAS and HAS-LIP.ConclusionsSA/CH-HAS-LIP could be a promising release vehicle for the oral delivery of HAS to increase its oral bioavailability.

Project description:Polymeric ultrathin membranes that are compatible with cells offer tremendous advantages for tissue engineering. In this article, we report a free-standing nanomembrane that was developed using a layer-by-layer self-assembly technique with a safe and sacrificial substrate method. After ionization, two oppositely charged polyelectrolytes, alginate and chitosan, were alternately deposited on a substrate of a solidified gelatin block to form an ultrathin nanomembrane. The space between the two adjacent layers was ∼200 nm. The thickness of the nanomembrane was proportional to the number of layers. The temperature-sensitive gelatin gel served as a sacrificial template at 37°C. The free-standing nanomembrane promoted bone marrow stem cell adhesion and proliferation. Fluorescence-activated cell sorting was used to analyze green-fluorescent-protein-positive mesenchymal stem cells from the wounds, which showed a significantly high survival and proliferation from the nanomembrane when cells were transplanted to mouse dorsal skin that had a full-thickness burn. The bone-marrow-stem-cell-loaded nanomembrane also accelerated wound contraction and epidermalization. Therefore, this methodology provides a fast and facile approach to construct free-standing ultrathin scaffolds for tissue engineering. The biocompatibility and free-standing nature of the fabricated nanomembrane may be particularly useful for stem cell delivery and wound healing.

Project description:Glucose oxidase (GOD) is widely used as an important oxidoreductase in various fields. However, maintaining the vitality and stability of GOD under environmental stress is a challenge. To improve the thermal and storage stability of GOD, this study constructed sodium alginate-carboxymethyl cellulose sodium gel particles (SA/CMC) and chitosan-coated SA/CMC gel particles (CS/SA/CMC) of GOD. The encapsulation efficiency (EE), gel particle structure, stability, and release behavior of GOD were evaluated. The results showed that the thermal stability of GOD encapsulated in SA/CMC and CS/SA/CMC gel particles was improved by approximately 2.8-fold and 4.3-fold compared with the free enzyme at 85 °C, respectively. In addition, CS/SA/CMC gel particles enhanced the enzyme activity retention rate of GOD to over 80% during storage at 4 °C for four weeks. Both SA/CMC and CS/SA/CMC gel particles loaded with GOD had more than 70% of the enzymes released during the simulated gastrointestinal experiment. The results demonstrated that encapsulating GOD in SA/CMC and CS/SA/CMC gel particles could improve its thermal stability and storage stability, which is conducive to further expanding the application of GOD in food, pharmaceutical and feed industries.

Project description:This study explores the combined delivery of doxorubicin and quercetin using a gelatin-oxidized alginate-based hydrogel as a promising strategy for localized breast cancer therapy. Our approach involves the incorporation of doxorubicin within the hydrogel matrix and loading quercetin into chitosan-coated zein nanoparticles. The hydrogel exhibited self-healing properties attributed to Schiff base cross-linking and demonstrated injectability. Characterization of its microstructural, mechanical, and textural properties revealed a porous and flexible structure, demonstrating its suitability for drug release applications. Both drugs exhibited distinct in vitro release profiles at pH 6.8 (typical of tumor tissue), with doxorubicin at 81.2% and quercetin at 9.7%. After 72 h of release, the cytotoxicity against MCF-7 breast cancer cells was assessed. The hydrogel formulation containing doxorubicin increased the cytotoxic action by 4.66-fold, whereas the hydrogel composite, containing both doxorubicin and quercetin-loaded nanoparticles, enhanced it by 20.7-fold compared with doxorubicin alone. Thus, the findings of our study highlight the enhancing effect of the dual release system, thereby expanding the utility of gelatin-oxidized alginate-based hydrogels as advanced drug delivery systems, as exemplified by the combined delivery of doxorubicin and quercetin.
