Project description:Selenium (Se) supplement was combined with different LED light qualities to investigate mutual effects on the growth, nutritional quality, contents of glucosinolates and mineral elements in broccoli sprouts. There were five treatments: CK:1R1B1G, 1R1B1G+Se (100 μmol L-1 Na2SeO3), 1R1B+Se, 1R2B+Se, 2R1B+Se, 60 μmol m-2 s-1 PPFD, 12 h/12 h (light/dark). Sprouts under a combination of selenium and LED light quality treatment exhibited no remarkable change fresh weight, but had a shorter hypocotyl length, lower moisture content and heavier dry weight, especially with 1R2B+Se treatment. The contents of carotenoid, soluble protein, soluble sugar, vitamin C, total flavonoids, total polyphenol and contents of total glucosinolates and organic Se were dramatically improved through the combination of Se and LED light quality. Moreover, heat map and principal component analysis showed that broccoli sprouts under 1R2B+Se treatment had higher nutritional quality and health-promoting compound contents than other treatments. This suggests that the Se supplement under suitable LED lights might be beneficial to selenium-biofortified broccoli sprout production.

The present study aimed to identify the regulatory mechanisms of red, blue, and white light on carotenoid biosynthesis in maize sprouts. Determinations of carotenoid, chlorophyll and phytohormone profiles, as well as relative gene expression, were explored. The results identified enhancement of carotenoid and chlorophyll production as well as gene expression. Most notably, the expression levels of CRY, HY5, and beta-carotene 3-hydroxylase genes peaked under blue light. Photomorphogene-related hormone, auxins and strigolactone production was also altered under different lights and might have a role in carotenoid metabolism. Gibberellins competed with carotenoids for the precursor geranylgeranyl diphosphate and were hindered by certain light characteristics, probably via DELLA-PIF4 signalling. ERF021 and MYB68 were negative regulators of carotenoid biosynthesis in maize sprouts. These findings provide new insights into the light-regulated mechanism and biofortification of carotenoids in maize sprouts.
Project description:Heat stress has been proved to increase the content of melatonin in plants. In the present study, a combination of methods including physiological and biochemical, gene transcription and proteomic were used to investigate the melatonin accumulation mechanisms in mustard sprouts under heat treatment during the germination period. It was revealed that the heat treatment can significantly affect sprout growth, antioxidant enzyme activity and melatonin content in mustard sprouts. Meanwhile, the expression of melatonin key synthase genes, such as tryptophan decarboxylase genes (BjTDC 1, BjTDC 2) and serotonin N-acetyltransferase genes (BjSNAT 1), were significantly induced by heat stress, which coincided with the trend of melatonin content. Under the heat stress, a total of 172 differential abundance proteins were confidently identified in mustard sprouts and participated in many physiological processes. Functional classification analysis showed that the defense/pressure, energy and nucleotide metabolism, protein biosynthesis, signal transduction and transcription etc. were largely induced. Heat treatment stimulated a defense response at the protein level by regulating the growth and physiological metabolism in mustard sprouts. The results in this work provided novel deep insights into the proteins' response to heat stress, which will certainly promote further understanding of the heat-tolerance mechanism of mustard sprouts.
Project description:Light is an important factor influencing melatonin synthesis in response to cadmium treatment in rice. However, the effects of light quality on, and the involvement of phytochrome light receptors in, melatonin production have not been explored. In this study, we used light-emitting diodes (LEDs) to investigate the effect of light wavelength on melatonin synthesis, and the role of phytochromes in light-dependent melatonin induction in rice. Upon cadmium treatment, peak melatonin production was observed under combined red and blue (R + B) light, followed by red (R) and blue light (B). However, both far-red (FR) LED light and dark treatment (D) failed to induce melatonin production. Similarly, rice seedlings grown under the R + B treatment showed the highest melatonin synthesis, followed by those grown under B and R. These findings were consistent with the results of our cadmium treatment experiment. To further confirm the effects of light quality on melatonin synthesis, we employed rice photoreceptor mutants lacking functional phytochrome genes. Melatonin induction was most inhibited in the phytochrome A mutant (phyA) followed by the phyB mutant under R + B treatment, whereas phyB produced the least amount of melatonin under R treatment. These results indicate that PhyB is an R light receptor. Expression analyses of genes involved in melatonin biosynthesis clearly demonstrated that tryptophan decarboxylase (TDC) played a key role in phytochrome-mediated melatonin induction when rice seedlings were challenged with cadmium.

We examined how photic and non-photic time cues may be combined by the human circadian system by assessing the phase advancing effects of one evening dose of exogenous melatonin, alone and in combination with one session of morning bright light exposure.DesignRandomized placebo-controlled double-blind circadian protocol. The effects of four conditions, dim light (∼1.9 lux, ∼0.6 Watts/m(2))-placebo, dim light-melatonin (5 mg), bright light (∼3000 lux, ∼7 Watts/m(2))-placebo, and bright light-melatonin on circadian phase was assessed by the change in the salivary dim light melatonin onset (DLMO) prior to and following treatment under constant routine conditions. Melatonin or placebo was administered 5.75 h prior to habitual bedtime and 3 h of bright light exposure started 1 h prior to habitual wake time.SettingSleep and chronobiology laboratory environment free of time cues.ParticipantsThirty-six healthy participants (18 females) aged 22 ± 4 y (mean ± SD).ResultsMorning bright light combined with early evening exogenous melatonin induced a greater phase advance of the DLMO than either treatment alone. The ability to combine circadian time cues has important implications for understanding fundamental physiological principles of the human circadian timing system. Knowledge of such principles is important for designing effective countermeasures for phase-shifting the human circadian clock to adapt to jet lag, shift work, and for designing effective treatments for circadian sleep-wakefulness disorders.
Project description:Pea sprouts have rich nutrition and are considered good for heart health. In this study, the kaspa peas and black-eyed peas were chosen to clarify the effect of different LED spectral combinations on the growth, yield, and nutritional quality of pea sprouts under long photoperiod (22 h light/2 h dark). The results showed that the two pea varieties responded differently to light spectral combinations. Black-eyed pea sprouts had higher plant height, fresh weight per plant, dry weight per plant, soluble sugar content, and lower malondialdehyde (MDA) content than kaspa peas under the same light treatment. Compared with white light, red-to-blue ratio of 2:1 significantly increased peroxidase (POD) and superoxide dismutase (SOD) activity, soluble sugar and soluble protein content of kaspa pea sprouts, and decreased MDA content of black-eyed pea sprouts. Blue light was negatively correlated with the plant height of pea sprouts and positively correlated with SOD activity, vitamin C, soluble sugar, and soluble protein content. Antioxidant capacity, yield, and nutritional quality of black-eyed pea sprouts were higher than those of kaspa pea sprouts under the same light treatment. Blue light improved the nutritional quality of pea sprouts. Compared with other light treatments, the red-to-blue ratio of 2:1 was more conducive to improving the antioxidant capacity and nutritional quality of pea sprouts under long photoperiod.
Project description:Indole-3-carbinol (I3C), an important secondary metabolite with strong anti-cancer ability, is widely found in cruciferous plants. Light and phytohormones are one of the most important external and internal signals, respectively, that control the growth, development, and secondary metabolism of the plant life cycle. However, there are few studies about the influence of the blue light and salicylic acid (SA) on the regulation of I3C accumulation. In this study, a negative correlation was found between the content of I3C and SA in different species. Among this, broccoli and Arabidopsis thaliana were chosen for further studies. We observed that blue light treatment increased the accumulation of I3C, and exogenous SA treatment significantly inhibited the accumulation of I3C in broccoli sprouts. Based on the RNA sequence, the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis indicated that blue light promoted the enrichment of differentially expressed genes (DEGs) in plant hormone signal transduction pathways. More specifically, downregulated expression of genes related to SA biosynthesis and upregulated expression of I3C genes related to metabolic pathway were observed under blue light. Taken together, these results suggested that SA negatively regulates blue light-induced I3C accumulation in broccoli sprouts.
Project description:This study aimed to identify the regulatory mechanisms of white, blue, red lights on carotenoid and tocochromanol biosynthesis in mung bean sprouts. Results showed that three lights stimulated the increase of the predominated lutein (3.2-8.1 folds) and violaxanthin (2.1-6.1 folds) in sprouts as compared with dark control, as well as β-carotene (20-36 folds), with the best yield observed under white light. Light signals also promoted α- and γ-tocopherol accumulation (up to 1.8 folds) as compared with dark control. The CRTISO, LUT5 and DXS (1.24-6.34 folds) exhibited high expression levels under light quality conditions, resulting in an overaccumulation of carotenoids. The MPBQ-MT, TC and TMT were decisive genes in tocochromanol biosynthesis, and were expressed up to 4.19 folds as compared with control. Overall, the results could provide novel insights into light-mediated regulation and fortification of carotenoids and tocopherols, as well as guide future agricultural cultivation of mung bean sprouts.
Project description:Light treatment is an innovative method to enhance the synthesis of secondary metabolites in plants and improve the quality of plant-based food ingredients. This study investigated the effects of red light treatment on the physiological and biochemical changes during wheat germination, aiming to produce high-quality wheat sprouts with strong antioxidant capacity. Using response surface methodology, the study optimized the conditions for phenolic accumulation in wheat sprouts under red light treatment and explored the molecular mechanisms behind the enhancement of total phenolic content (TPC) and quality. The results indicated that red light treatment significantly increased the TPC in wheat sprouts. The highest TPC, reaching 186.61 μg GAE/sprout, was observed when wheat sprouts were exposed to red light at an intensity of 412 μmol/m²/s for 18.2 h/d over four days. Compared to no light, red light treatment significantly increased the content of photosynthetic pigments (chlorophyll and carotenoids). Red light treatment notably heightened the levels of both free and bound phenolic in the germinating wheat. Red light treatment markedly boosted the activities and relative gene expression levels of enzymes related to phenolic biosynthesis, including phenylalanine ammonia-lyase, cinnamate-4-hydroxylase, and 4-coumarate-CoA ligase. Additionally, red light treatment enhanced the antioxidant capacity of wheat sprouts by improving the activity and gene expression of four key antioxidant enzymes, thereby promoting growth and germination. This research suggested that red light treatment is an effective strategy for stimulating total phenolic biosynthesis, enhancing antioxidant capacity, and producing highly nutritious wheat sprouts, thus laying the groundwork for developing total phenolic-enriched wheat sprouts as valuable food ingredients in the future.
Project description:Anthocyanin is one important nutrition composition in Tartary buckwheat (Fagopyrum tataricum) sprouts, a component missing in its seeds. Although anthocyanin biosynthesis requires light, the mechanism of light-induced anthocyanin accumulation in Tartary buckwheat is unclear. Here, comparative transcriptome analysis of Tartary buckwheat sprouts under light and dark treatments and biochemical approaches were performed to identify the roles of one B-box protein BBX22 and ELONGATED HYPOCOTYL 5 (HY5). The overexpression assay showed that FtHY5 and FtBBX22 could both promote anthocyanin synthesis in red-flower tobacco. Additionally, FtBBX22 associated with FtHY5 to form a complex that activates the transcription of MYB transcription factor genes FtMYB42 and FtDFR, leading to anthocyanin accumulation. These findings revealed the regulation mechanism of light-induced anthocyanin synthesis and provide excellent gene resources for breeding high-quality Tartary buckwheat.
