Abundance, characterization, and health risk evaluation of microplastics in borehole water in Birnin Kebbi, Nigeria.

ABSTRACT: Microplastic pollution has become a global menace, and water, being a major "sink" for pollutants, represents a significant source of human exposure. This study aimed to assess the safety of borehole water in Birnin Kebbi, Nigeria, specifically concerning microplastic pollution. Water samples were collected from boreholes in selected areas, including Bayan Kara, Malali, Rafin Atiku, Aliero Quarters, GwadanGaji, FUBK Takeoff Site, Kalgo Market, and Tarasa. Microplastics were extracted from the water samples through filtration using glass fiber filter papers, and were subsequently subjected to spectroscopy and microscopy to determine concentrations, shapes, and polymer types. Health risks associated with the microplastics were also calculated. The results revealed that the samples from Tarasa exhibited the highest concentrations of microplastics (96.967 particles/L), followed by Bayan Kara (92.70 particles/L), Rafin Atiku (92.33 particles/L), GwadanGwaji (92.30 particles/L), FUBK Takeoff Site (91.07 particles/L), Aliero Quarters (90.43 particles/L), Kalgo Market (88.00 particles/L), and Malali (86.40 particles/L). The most dominant shape was fibers (73 %), followed by fragments (16 %), foams (6 %), and filaments (5 %). Polyethylene and polyamide, in that order, were the most dominant polymers, while polystyrene was the least common. The majority of risk scores were classified as III. It can be inferred from the results that microplastic pollution in borehole water poses a health hazard in the city. Consumers of borehole water in the studied areas are advised to treat the water before consumption to mitigate potential health risks.

Project description:Mycotoxins in drinking water are neglected pollutants that have serious health implications when ingested. Common mycotoxins with health concerns include deoxynivalenol (DON), ochratoxin A (OTA), and zearalenone (ZEN). This study considers the distribution and apparent health risks of these mycotoxins in drinking water sources (groundwater, surface water, bottled water, sachet water) in three Southwest Nigeria States: Osun, Oyo, and Lagos States, using the UHPLC-ESI-QTOF. ZEN and DON were found in all 95 water samples across all three states. ZEN in sachet water samples has the highest mean concentrations, with those from Osun, Oyo, and Lagos States having concentrations of 14.96 ± 4.46, 8.59 ± 3.86, and 10.56 ± 2.84 μg L-1, respectively. In contrast, the mean concentrations of all three mycotoxins (± Standard Error of Mean) in bottled water samples are the lowest across the three States. The mean concentrations of OTA in sachet water samples (2.93 ± 0.79, 1.24 ± 0.40, and 3.01 ± 1.50 μg L-1) are slightly higher than those in bottled water (1.47 ± 0.28, 1.53 ± 0.31, and 0.75 ± 0.31 μg L-1) for Osun, Oyo, and Lagos States, respectively. Groundwater samples across the three States had the lowest average pH values, below the WHO's lowest limit of 6.5. Principal Component Analysis studies indicate that all three mycotoxins in water samples are closely associated by source. Possible human exposure values for ZEN suggest some health concerns, especially with the use of sachet water type based on the estrogenicity of this mycotoxin. However, OTA values for all water samples analyzed, are much lower than the tolerable daily intake (TDI) of 4.73 μg kg-1 bw per day provided by European Food Safety Auhtority (EFSA). The levels of DON in all water sample types were of no serious health concern. However, human exposure levels to all three mycotoxins in bottled water fall within safe limits of health-based guidance values (HBGV) indicating that this drinking water type is better treated, unlike sachet water. Nevertheless, stakeholders need to re-examine water quality with respect to these mycotoxins and adopt stringent guidelines and new water treatment strategies to provide consumers with safe drinking water in line with the UN SDG #6.

Project description:IntroductionDespite the benefits of periodic evaluation of the vaccine safety surveillance system, no formal assessment, to our knowledge, has been conducted in Nigeria. Hence, this study evaluated the surveillance system for adverse events following immunization (AEFI) to ascertain the system's functionality to inform vaccine safety considerations and guide communication strategies for demand generation.Materials and methodsThe study employed a mixed-method approach. Survey questionnaires were administered to 274 routine immunization service providers in Kebbi State, Northern Nigeria, and data were analyzed descriptively using SPSS. In this study, 10 Key Informant Interviews and two Focus Group Discussions were conducted with senior officers and managers at sub-national and national levels within the immunization and surveillance landscape in Nigeria. The interview recordings were cleaned minimally, transcribed, and manually analyzed thematically. Finally, methodological triangulation was done to improve research rigor and provide a better understanding of the phenomena under investigation.ResultsOf the respondents, 201(73.4%) reported that the surveillance system can inform vaccine safety considerations while 170(62%) reported that the AEFI surveillance system can determine the magnitude of AEFI within the population. Further, 173(63%) reported that the surveillance system can provide timely feedback about causality assessment. However, 158(58%) of the respondents stated that the surveillance system is competent in informing communication strategies to improve immunization demand. Triangulation was done which showed dissonance in AEFI surveillance and vaccine safety considerations but partial agreement in immunization demand generation. Further, AEFI surveillance system attributes' triangulation revealed agreements (convergence) on simplicity and timeliness; partial agreements on acceptability, data quality, sensitivity, flexibility, and completeness; dissonance on representativeness and silence on stability, indicating a sub-optimal performance of the AEFI surveillance system in the study setting. Finally, the study unearthed some underlying health system factors impeding the AEFI surveillance system from fully fulfilling its objectives.ConclusionThe AEFI surveillance system in Northern Nigeria is well established but functioning sub-optimally. Based on the study findings, the capacity to provide information on vaccine safety exists but it is not robust enough to generate sufficient and convincing vaccine safety data and guide communication strategies for vaccine demand generation, especially for new vaccines and those under emergency authorization use.

Project description:Validation of analytical methods for measurements of microplastics (MP) is severely hampered because of a general lack of reference materials, RM. There is a great need to develop such reference materials. This study presents a concept of three-component kit with immobilised MP in solid NaCl, a surfactant and clean water that can be applied for the production of many types of MP RMs. As proof of concept, an RM for polyethylene terephthalate (PET) particles in water was prepared and evaluated for its homogeneity. The particles ranged from 30 μm (Feretmin) to about 200 μm adapted by wet sieving. A specific number of PET particles were immobilized in about 0.29 g of solid NaCl by freeze-drying 1 mL of a NaCl suspension. By using manual and automated counting, twenty reconstituted 1-L water samples were evaluated for homogeneity with respect to number of PET particles from 30 μm to > 200 μm/L of water. The number of particles was 730 ± 120 (mean ± one standard deviation (SD); n = 10) and 865 ± 155 particles (n = 10) obtained by optical microscopy in two independent laboratories. This corresponded to relative SDs of 16.4 and 17.9% and a mean of 797 ± 151 particles (18.9% RSD, for n = 20). Homogeneity studies of the NaCl carrier without reconstitution resulted in 794 ± 60 particles (7.5% RSD). The homogeneity of PET in the salt carrier was also evaluated directly with respect to mass of PET per vial using an ultra-micro balance. An average mass of 293 ± 41 μg of PET was obtained (14, % RSD for n = 14). Micrographs were recorded to demonstrate the absence of major sources of contamination of the RM components. Information about the particle size distribution and particle shapes was obtained by laser diffraction (LD) and dynamic image analysis (DIA). In addition, the identity of the PET polymer was confirmed by Raman and FT-IR spectroscopy. The RM was developed for a large-scale inter-laboratory comparison of PET particles in water (ILC). Based on the homogeneity results, the material was found to be sufficiently homogeneous to be of meaningful use in the ILC. In a 3-day process, more than 500 samples of PET particles in the NaCl carrier were prepared with good potential for further upscaling with respect to the number of vials or with other kinds of polymers. The stability of PET was not evaluated but it was deemed to be stable for the duration of the ILC.
