Project description:Radio frequency ablation and microwave hyperthermia are powerful tools for destroying dysfunctional biological tissues. However, wireless application of these techniques is hindered by their inability to focus the electromagnetic energy to small targets. The use of locally injected radio frequency- or microwave-absorbing nanomaterials can help to overcome this challenge by confining heat production to the injected region. Previous theoretical work suggests that high-aspect-ratio conducting nanomaterials, such as carbon nanotubes, offer powerful radio frequency and microwave absorption. While carbon nanotubes have been demonstrated as radiothermal agents, common solubilization methods may reduce their absorption efficiency, yielding undesirable nonspecific heating in the biological tissue background. In this manuscript, we hypothesize that pristine carbon nanotubes can act as efficient absorbers at radio frequencies, thus providing differential heating over the tissue background. Specifically, we use a sonication-free preparation technique to preserve both the high aspect ratio and local concentration of pristine carbon nanotubes. We validate the differential heating of these samples by 4.5-fold at 2 GHz compared to the heating of saline at a physiological concentration using infrared thermography. In addition, we successfully achieved local heating of pristine carbon nanotubes within a three-dimensional biological tissue phantom. Numerical simulations further aid in producing a temperature map within the phantom and confirming localized heating. Due to their significant differential and local heating, we believe that pristine carbon nanotubes may facilitate region-specific radio frequency ablation and microwave hyperthermia while keeping nonspecific heating to a low level in the normal tissue background.

Project description:In carbon nanotube fibers (CNFs) fabricated by spinning methods, it is well-known that the mechanical and thermal performances of CNFs are highly dependent on the mechanical and thermal properties of the inherent CNTs. Furthermore, long CNTs are usually preferred to assemble CNFs because the interaction and entanglement between long CNTs are effectively stronger than between short CNTs. However, in CNFs fabricated using long CNTs, the interior carbon nanotubes (CNTs) inevitably undergo both tension and torsion loading when they are stretched, which would influence the mechanical and thermal performances of CNFs. Here, molecular dynamics (MD) simulations were carried out to study the mechanical and thermal properties of individual CNTs under tension-torsion loading. As for mechanical properties, it was found that both the fracture strength and Young's modulus of CNTs decreased as the twist angle α increased. Besides, step-wise fracture happened due to stress concentration when the twisted CNTs are stretched. On the other hand, it could be seen that the thermal conductivity of CNTs decreased as α increased. This work presents the systematic investigation of the mechanical and thermal properties of CNTs under tension-torsion loading and provides a theoretical guideline for the design and fabrication of CNFs.

Project description:BackgroundAccumulated evidence shows that the abnormal regulation of long non-coding RNA (lncRNA) is associated with various human diseases. Accurately identifying disease-associated lncRNAs is helpful to study the mechanism of lncRNAs in diseases and explore new therapies of diseases. Many lncRNA-disease association (LDA) prediction models have been implemented by integrating multiple kinds of data resources. However, most of the existing models ignore the interference of noisy and redundancy information among these data resources.ResultsTo improve the ability of LDA prediction models, we implemented a random forest and feature selection based LDA prediction model (RFLDA in short). First, the RFLDA integrates the experiment-supported miRNA-disease associations (MDAs) and LDAs, the disease semantic similarity (DSS), the lncRNA functional similarity (LFS) and the lncRNA-miRNA interactions (LMI) as input features. Then, the RFLDA chooses the most useful features to train prediction model by feature selection based on the random forest variable importance score that takes into account not only the effect of individual feature on prediction results but also the joint effects of multiple features on prediction results. Finally, a random forest regression model is trained to score potential lncRNA-disease associations. In terms of the area under the receiver operating characteristic curve (AUC) of 0.976 and the area under the precision-recall curve (AUPR) of 0.779 under 5-fold cross-validation, the performance of the RFLDA is better than several state-of-the-art LDA prediction models. Moreover, case studies on three cancers demonstrate that 43 of the 45 lncRNAs predicted by the RFLDA are validated by experimental data, and the other two predicted lncRNAs are supported by other LDA prediction models.ConclusionsCross-validation and case studies indicate that the RFLDA has excellent ability to identify potential disease-associated lncRNAs.

Project description:The advance of nanomaterials has opened new opportunities to develop ever more sensitive sensors owing to their high surface-to-volume ratio. However, it is challenging to achieve intrinsic sensitivities of nanomaterials for ultra-low level detections due to their vulnerability against contaminations. Here we show that despite considerable achievements in the last decade, continuous in situ cleaning of carbon nanotubes with ultraviolet light during gas sensing can still dramatically enhance their performance. For instance in nitric oxide detection, while sensitivity in air is improved two orders of magnitude, under controlled environment it reaches a detection limit of 590 parts-per-quadrillion (ppq) at room temperature. Furthermore, aiming for practical applications we illustrate how to address gas selectivity by introducing a gate bias. The concept of continuous in situ cleaning not only reveals the tremendous sensing potential of pristine carbon nanotubes but also more importantly it can be applied to other nanostructures.

Project description:BackgroundA large body of evidence shows that miRNA regulates the expression of its target genes at post-transcriptional level and the dysregulation of miRNA is related to many complex human diseases. Accurately discovering disease-related miRNAs is conductive to the exploring of the pathogenesis and treatment of diseases. However, because of the limitation of time-consuming and expensive experimental methods, predicting miRNA-disease associations by computational models has become a more economical and effective mean.ResultsInspired by the work of predecessors, we proposed an improved computational model based on random forest (RF) for identifying miRNA-disease associations (IRFMDA). First, the integrated similarity of diseases and the integrated similarity of miRNAs were calculated by combining the semantic similarity and Gaussian interaction profile kernel (GIPK) similarity of diseases, the functional similarity and GIPK similarity of miRNAs, respectively. Then, the integrated similarity of diseases and the integrated similarity of miRNAs were combined to represent each miRNA-disease relationship pair. Next, the miRNA-disease relationship pairs contained in the HMDD (v2.0) database were considered positive samples, and the randomly constructed miRNA-disease relationship pairs not included in HMDD (v2.0) were considered negative samples. Next, the feature selection based on the variable importance score of RF was performed to choose more useful features to represent samples to optimize the model's ability of inferring miRNA-disease associations. Finally, a RF regression model was trained on reduced sample space to score the unknown miRNA-disease associations. The AUCs of IRFMDA under local leave-one-out cross-validation (LOOCV), global LOOCV and 5-fold cross-validation achieved 0.8728, 0.9398 and 0.9363, which were better than several excellent models for predicting miRNA-disease associations. Moreover, case studies on oesophageal cancer, lymphoma and lung cancer showed that 94 (oesophageal cancer), 98 (lymphoma) and 100 (lung cancer) of the top 100 disease-associated miRNAs predicted by IRFMDA were supported by the experimental data in the dbDEMC (v2.0) database.ConclusionsCross-validation and case studies demonstrated that IRFMDA is an excellent miRNA-disease association prediction model, and can provide guidance and help for experimental studies on the regulatory mechanism of miRNAs in complex human diseases in the future.

Project description:Electroconductive materials based on poly(lactic acid) (PLA) electrospinning membranes grafted with carbon nanotubes (CNTs) functionalized with the carboxylic group R-COOH have been obtained. PLA electrospun membranes were modified with sulfuric acid (H2SO4) to oxidize its surface to subsequently graft the CNTs, the treatment time and drying of the membranes before grafting with CNTs being critical, influencing the final properties of the materials. SEM images showed that CNTs presented a uniform distribution on the surface of the PLA nanofibers, while FTIR spectra of PLA-CNTs materials revealed characteristic hydroxyl groups, as evidenced by absorption peaks of CNTs. Thanks to the grafting with CNTs, the resulting PLA-CNTs membranes present an improvement of the mechanical and conductive properties when compared with PLA membranes. On the one hand, grafting with CNTs causes the nanofibers to have greater rigidity, so they are more manipulable and can more easily preserve their conformation when stress is exerted. On the other hand, grafting with CNTs allows elimination of the insulating barrier of the PLA, reducing the resistivity and providing high electrical conductivity to the PLA-CNTs membranes. The incorporation of CNTs into PLA electrospun membranes is expected to offer greater functionalities to electrospun composite nanofibers for medical and industrial applications.

Project description:We report the successful microencapsulation of multi-walled carbon nanotubes suspended in a 5-ethylidene-2-norbornene (5E2N) self-healing monomer, into poly melamine urea formaldehyde shells through in situ polymerization. The average size of the microcapsules, their size-distribution, shell wall structural integrity and thickness are characterized by optical and scanning electron microscopy. The presence of carbon nanotubes (CNTs) inside the core liquid content, as well as their release after breaking is confirmed by microscopy and spectroscopy analyses. A small amount of CNTs inside the microcapsules is found to have no significant impact on the thermal stability of the system, as determined by thermogravimetric analysis and differential scanning calorimetry. Both the mechanical and the electrical properties of CNT-based self-healing materials can be restored up to 80% when CNT/5E2N microcapsules are incorporated into polymer composites, thus making them highly suitable for applications in aerospace.

Project description:The estimation of a postmortem interval (PMI) is particularly important for forensic investigations. The aim of this study was to assess the succession of bacterial communities associated with the decomposition of mouse cadavers and determine the most important biomarker taxa for estimating PMIs. High-throughput sequencing was used to investigate the bacterial communities of gravesoil samples with different PMIs, and a random forest model was used to identify biomarker taxa. Redundancy analysis was used to determine the significance of environmental factors that were related to bacterial communities. Our data showed that the relative abundance of Proteobacteria, Bacteroidetes and Firmicutes showed an increasing trend during decomposition, but that of Acidobacteria, Actinobacteria and Chloroflexi decreased. At the genus level, Pseudomonas was the most abundant bacterial group, showing a trend similar to that of Proteobacteria. Soil temperature, total nitrogen, NH4+-N and NO3--N levels were significantly related to the relative abundance of bacterial communities. Random forest models could predict PMIs with a mean absolute error of 1.27 days within 36 days of decomposition and identified 18 important biomarker taxa, such as Sphingobacterium, Solirubrobacter and Pseudomonas. Our results highlighted that microbiome data combined with machine learning algorithms could provide accurate models for predicting PMIs in forensic science and provide a better understanding of decomposition processes.

Project description:This study aims to investigate the price changes in the carbon trading market and the development of international carbon credits in-depth. To achieve this goal, operational principles of the international carbon credit financing mechanism are considered, and time series models were employed to forecast carbon trading prices. Specifically, an ARIMA(1,1,1)-GARCH(1,1) model, which combines the Generalized Autoregressive Conditional Heteroskedasticity (GARCH) and Autoregressive Integrated Moving Average (ARIMA) models, is established. Additionally, a multivariate dynamic regression Autoregressive Integrated Moving Average with Exogenous Inputs (ARIMAX) model is utilized. In tandem with the modeling, a data index system is developed, encompassing various factors that influence carbon market trading prices. The random forest algorithm is then applied for feature selection, effectively identifying features with high scores and eliminating low-score features. The research findings reveal that the ARIMAX Least Absolute Shrinkage and Selection Operator (LASSO) model exhibits high forecasting accuracy for time series data. The model's Mean Squared Error, Root Mean Squared Error, and Mean Absolute Error are reported as 0.022, 0.1344, and 0.1543, respectively, approaching zero and surpassing other evaluation models in predictive accuracy. The goodness of fit for the national carbon market price forecasting model is calculated as 0.9567, indicating that the selected features strongly explain the trading prices of the carbon emission rights market. This study introduces innovation by conducting a comprehensive analysis of multi-dimensional data and leveraging the random forest model to explore non-linear relationships among data. This approach offers a novel solution for investigating the complex relationship between the carbon market and the carbon credit financing mechanism.
