Project description:So far, there have been 4 methods to control chirality including the use of chiral auxiliaries, reagents, solvents, and catalysts documented in literature and textbooks. Among them, asymmetric catalysts are normally divided into homogeneous and heterogeneous catalysis. In this report, we present a new type of asymmetric control-asymmetric catalysis via chiral aggregates that would not belong to the above categories. This new strategy is represented by catalytic asymmetric dihydroxylation reaction of olefins in which chiral ligands are aggregated by taking advantage of typical aggregation-induced emission systems containing tetrahydrofuran and H2O cosolvents. It was proven that the chiral induction can be enhanced from er of 78:22 to 97:3 simply by changing the ratios of these 2 cosolvents. The formation of chiral aggregates of asymmetric dihydroxylation ligands, (DHQD)2PHAL and (DHQ)2PHAL, has been proven by aggregation-induced emission and a new analytical tool-aggregation-induced polarization established by our laboratory. In the meanwhile, chiral aggregates were found to be formed either by adding NaCl into tetrahydrofuran/H2O systems or by increasing concentrations of chiral ligands. The present strategy also showed promising reverse control of enantioselectivity in the Diels-Alder reaction. This work is anticipated to be extended broadly to general catalysis, especially to asymmetric catalysis in the future.

Project description:A new chiral aggregate-based tool for asymmetric synthesis has been developed by taking advantage of chiral aggregates of GAP (Group-Assisted Purification) reagents, N-phosphonyl imines. This tool was proven to be successful in the asymmetric GAP synthesis of functionalized 2,3-dihydrobenzofurans by reacting salicyl N-phosphonyl imines with dialkyl bromomalonates in various cosolvent systems. The chiral induction can be controlled by differentiating between two asymmetric directions simply by changing the ratios of cosolvents which are commonly adopted in AIE (aggregation-induced emission) systems. The formation of chiral aggregates was witnessed by a new analytical tool-aggregation-induced polarization (AIP). The present synthetic method will be broadly extended for general organic synthesis, particularly, for asymmetric synthesis and asymmetric catalysis in the future.

Project description:Compartmentalization of chemical reactions inside cells are a fundamental requirement for life. Encapsulins are self-assembling protein-based nanocompartments from the prokaryotic repertoire that present a highly attractive platform for intracellular compartmentalization of chemical reactions by design. Using single-molecule Förster resonance energy transfer and 3D-MINFLUX analysis, we analyze fluorescently labeled encapsulins on a single-molecule basis. Furthermore, by equipping these capsules with a synthetic ruthenium catalyst via covalent attachment to a non-native host protein, we are able to perform in vitro catalysis and go on to show that engineered encapsulins can be used as hosts for transition metal catalysis inside living cells in confined space.

Project description:In this paper, we report the synthesis and self-assembly behavior of coil-rod-coil molecules, consisting of three biphenyls linked through a vinylene unit as a conjugated rod segment and poly(ethylene oxide) (PEO) with a degree of polymerization (DP) of 7, 12 and 17, incorporating lateral methyl groups between the rod and coil segments as the coil segment. Self-organized investigation of these molecules by means of differential scanning calorimetry (DSC), thermal polarized optical microscopy (POM) and X-ray diffraction (XRD) reveals that the lateral methyl groups attached to the surface of rod and coil segments, dramatically influence the self-assembling behavior in the liquid-crystalline mesophase. Molecule 1 with a relatively short PEO coil length (DP=7) self-assembles into rectangular and oblique 2-dimensional columnar assemblies, whereas molecules 2 and 3 with DP of 12 and 17 respectively, spontaneously self-organize into unusual 3-dimensional hexagonal close-packed or body-centered tetragonal assemblies.

Project description:A series of bent-shaped molecules, consisting of dibenzo[a,c]phenazine and phenyl groups connected together as a rod segment, and poly(ethylene oxide) (PEO) with a degree of polymerization (DP) of 6 as the coil segment, were synthesized. The self-assembling behavior of these molecules by differential scanning calorimetry (DSC), thermal optical polarized microscopy (POM), small-angle X-ray scattering spectroscopy (SAXS), atomic force microscopy (AFM), and transmission electron microscopy (TEM), revealed that carboxyl or butoxy carbonyl groups at the 11 position of dibenzo[a,c]phenazine noticeably influence self-organization of molecules into supramolecular aggregates in bulk and aqueous solutions. Molecules 1 and 2 with chiral or non-chiral PEO coil chains and the carboxyl group at the rod segments self-organize into a hexagonal perforated lamellar structure and a hexagonal columnar structure in the solid state. In aqueous solution, molecules 1 and 2 self-assemble into diverse lengths of nanofibers, whereas molecules 3 and 4 with butoxy carbonyl groups exhibit a self-organizing capacity to form diverse sizes of spherical aggregates.

Project description:Asymmetric amplification induced in the Soai autocatalytic reaction by chiral initiators that are enantiomeric only by virtue of an isotope, e.g., -CH3 vs CD3-, is examined by spectroscopic, kinetic, and DFT modeling studies to help understand requirements for the emergence of biological homochirality. We find that the initiator inhibits the autocatalytic pathway at the outset of the reaction but ultimately provides the imbalance required for asymmetric amplification. This work provides clues in the ongoing search for prebiotically plausible versions of asymmetric autocatalysis.

Project description:From their realization just over a decade ago, nanopores in silicon nitride membranes have allowed numerous transport-based single-molecule measurements. Here we report the use of these nanopores as subzeptoliter mixing volumes for the controlled synthesis of metal nanoparticles. Particle synthesis is controlled and monitored through an electric field applied across the nanopore membrane, which is positioned so as to separate electrolyte solutions of a metal precursor and a reducing agent. When the electric field drives reactive ions to the nanopore, a characteristic drop in the ion current is observed, indicating the formation of a nanoparticle inside the nanopore. While traditional chemical synthesis relies on temperature and timing to monitor particle growth, here we observe it in real time by monitoring electrical current. We describe the dynamics of gold particle formation in sub-10 nm diameter silicon nitride pores and the effects of salt concentration and additives on the particle's shape and size. The current versus time signal during particle formation in the nanopore is in excellent agreement with the Richards growth curve, indicating an access-limited growth mechanism.

Project description:We report the conformational and assembly behavior of a thermoresponsive triblock biohybrid conjugate under aqueous conditions. The triblock comprises of poly(diethylene glycol methyl ether methacrylate) (PDEGMEMA) conjugated to the ends of a triple-helix forming collagen-like peptide. The circular dichroism (CD) experiment confirms the ability of the collagen-like peptide middle block to assemble as a triple helix in the hybrid conjugate. Above the LCST (~35 °C), the collapse of the thermoresponsive PDEGMEMA polymer at the ends of the peptide domain resulted in a concomitant increase in the conformational stability of the peptide domain towards thermal denaturation. Upon cooling back, the kinetic conformational refolding behavior was still observed for the peptide domain in the hybrid conjugate. Static light scattering (SLS) experiments suggested the formation of supramolecular structures upon increasing solution temperatures to above the LCST. The scattering intensity increased with increasing temperature, until at 75 °C then it was found to decrease. Cryogenic scanning electron microscopy and regular transmission electron microscopy suggested the formation of spherical aggregates that increased in size with increasing temperature up to 65 °C and a morphological transformation into fibrils was also observed at 75 °C. The synergistic effect of dual thermoresponsive behavior from the peptide and the polymer block in the triblock hybrid is suggested for the observed conformational and assembly behaviors.

Project description:Sorption mechanism of uranyl by poly(bis[2-(methacryloyloxy)ethyl] phosphate) (PB2MP) functionalised polyvinylidene fluoride (PVDF) track-etched membranes, PB2MP-g-PVDF, was investigated. It was found that uranyl sorption obeyed Langmuir isotherm model giving a maximum U(VI) membrane uptake of 6.73 μmol g-1 and an affinity constant of 9.85 ⋅ 106 L mol-1. XPS and TRPL measurements were performed to identify sorbed uranyl oxidation state and its environment. Uranyl was found to be mainly in its hexavalent state, i.e. U(VI), showing that the trapping inside the PB2MP-g-PVDF nanoporous membranes did not change the ion speciation. Two sorbed uranyl life-times (τ1 = 8.8 μs and τ2 = 102.8 μs) were measured by TRPL which pointed out different complexations taking place inside the nanopores. Uranyl sorption by PB2MP-g-PVDF membranes was also found to be pH dependent demonstrating the highest performance at circumneutral pH. In addition, TRPL was demonstrated to be not only a remarkable technique for U(VI) characterization, but also an alternative to voltammetry detection for trace on-site uranyl monitoring using PB2MP-g-PVDF nanoporous membranes.

Project description:The diffusion of coil-rod-coil triblock copolymers in entangled coil homopolymers is experimentally measured and demonstrated to be significantly slower than rod or coil homopolymers of the same molecular weight. A model coil-rod-coil triblock was prepared by expressing rodlike alanine-rich α-helical polypeptides in E. coli and conjugating coillike poly(ethylene oxide) (PEO) to both ends to form coil-rod-coil triblock copolymers. Tracer diffusion through entangled PEO homopolymer melts was measured using forced Rayleigh scattering at various rod lengths, coil molecular weights, and coil homopolymer concentrations. For rod lengths, L, that are close to the entanglementh length, a, the ratio between triblock diffusivity and coil homopolymer diffusivity decreases monotonically and is only a function of L/a, in quantitative agreement with previous simulation results. For large rod lengths, diffusion follows an arm retraction scaling, which is also consistent with previous theoretical predictions. These experimental results support the key predictions of theory and simulation, suggesting that the mismatch in curvature between rod and coil entanglement tubes leads to the observed diffusional slowing.
