Project description:Designing new, verified methodologies with a focus on sustainability, analytical efficiency, simplicity, and the environment has become a major priority for pharmaceutical quality control units. In this way, sustainable and selective separation-based methodologies were designed and validated for the concurrent estimation of amiloride hydrochloride (AML), hydrochlorothiazide (HCT) and timolol maleate (TIM) in their fixed dose formulation (Moducren® Tablets) along with hydrochlorothiazide potential impurities, salamide (DSA) and chlorothiazide (CT). The first method is a high performance thin layer chromatographic method (HPTLC-densitometry). The first developed method employed silica gel HPTLC F254 plates as stationary phase using a chromatographic developing system composed of ethyl acetate-ethanol-water-ammonia (8.5:1:0.5:0.3, by volume). The separated drug bands were densito-metrically measured at 220.0 nm for AML, HCT, DSA and CT and at 295.0 nm for TIM. The linearity was assessed over a wide concentration range, 0.5-10 µg/band, 1.0-16.0 µg/band and 1.0-14 µg/band for AML, HCT and TIM, in order and 0.05-1.0 µg/band for each of DSA and CT. The second method is capillary zone electrophoresis (CZE). The electrophoretic separation was achieved using background electrolyte (BGE), borate buffer 40.0 mM with pH 9.0 ± 0.2, at applied voltage of + 15 kV with on-column diode array detection at 200.0 nm. The method linearity was reached over the concentration range of 20.0-160.0 µg/mL, 10.0-200.0 µg/mL, 10.0-120.0 µg/mL for AML, HCT and TIM, respectively and 10.0-100.0 µg/mL for DSA. The suggested methods were optimized to achieve best performance and validated agreeing with the ICH guidelines. Assessment of methods' sustainability and greenness was performed using different greenness assessment tools.

Project description:In the present work, a comparative study on eco-friendly synthesis of zinc oxide (ZnO) sample 1 and sample 2 with 3.17 and 4.17 M NaOH, respectively, is reported. Sample 2 with 4.17 M NaOH is applied in the photocatalytic degradation of paracetamol (pure and raw both) using the ultraviolet (UV, 280-400 nm) and UV/H2O2 reaction systems. Pure paracetamol (PCM1) and raw paracetamol (PCM2) from tablets are used for photocatalytic degradation by photocatalysis. Our experimental evidence show that ZnO sample 2 was more active in the UV/H2O2 reaction system than under ultraviolet (UV, 280-400 nm) irradiation only in the photocatalytic degradation process. Field emission scanning electron microscopy (FE-SEM) confirms the homogeneous growth of a rod-like structure for sample 1 and brittle and randomly aggregated rod-like and wire-like nanostructures for sample 2. The peaks observed in the region around 440 to 900 cm-1 in the FTIR spectra for sample 1 and sample 2 annealed at 250 °C confirms the presence of ZnO bonds. UV absorption spectroscopy indicates a red shift in the absorption spectra due to the increase in the molar concentration of NaOH to 4.17 M for sample 2. In this study, the band gap values are found to be 3.33 and 3.01 eV for the synthesized ZnO sample 1 and sample 2, respectively, which are 40 and 360 meV less as compared to that of bulk ZnO (3.37 eV). The oxidation rate is increased in the UV/H2O2 reaction system, producing the highest rate for PCM1 drug removal with rate constant 9.7 × 10-3 min-1 and half-life 71.5 min. The kinetic study results for the removal of PCM1 and PCM2 show good results and follow the pseudo-first-order kinetic model with correlation coefficients 0.69556 and 0.90851, respectively, whereas PCM2 follows the pseudo-second-order kinetic model with correlation coefficient 0.9993. The experimental and calculated values of removal capacity (q e) at equilibrium is found close to those of the pseudo-second order kinetic model for the removal of both the paracetamol forms PCM1 and PCM2 with the catalyst ZnO nanostructure. The photostability of ZnO sample 2 is also tested with a reusability test in photocatalytic degradation of paracetamol at least four times. The absence of a maxima peak at 243 of PCM1 in the UV/H2O2 reaction system indicates nearly 100% successful conversion of 20 ppm PCM1 by using synthesized catalyst ZnO sample 2. The comparative results of both reaction systems, i.e., UV and UV/H2O2, show that the hydroxyl radicals, as the active species, are responsible for major degradation of both paracetamol forms (PCM1 and PCM2).

Project description:Aquatic environments face contamination by pharmaceuticals, prompting concerns due to their toxicity even at low concentrations. To combat this, we developed an ecologically sustainable biosurfactant derived from a microorganism and integrated it into bacterial cellulose (BC). This study aimed to evaluate BC's efficacy, with and without the biosurfactant, as a sorbent for paracetamol and 17α-ethinylestradiol (EE2) in water. We cultivated BC membranes using Gluconacetobacter xylinus ATCC 53582 and synthesized the biosurfactant through pre-inoculation of Bacillus subtilis in a synthetic medium. Subsequently, BC membranes were immersed in the biosurfactant solution for incorporation. Experiments were conducted using contaminated water, analyzing paracetamol concentrations via spectrophotometry and EE2 levels through high-performance liquid chromatography. Results indicated BC's superior adsorption for EE2 over paracetamol. Incorporating the biosurfactant reduced hormone adsorption but enhanced paracetamol sorption. Notably, original and freeze-dried BC exhibited better adsorption efficacy than biosurfactant-infused BC. In conclusion, BC showed promise in mitigating EE2 contamination, suggesting its potential for environmental remediation. Future research could focus on optimizing biosurfactant concentrations to enhance sorption capabilities without compromising BC's inherent effectiveness.

Project description:Graphene oxide (GO) was synthesized and characterized by scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDX), atomic force microscopy (AFM), X-ray diffraction (XRD), Fourier transform-infrared spectroscopy (FT-IR) and thermogravimetric analysis (TGA). GO was then electrochemically reduced and used for electrochemical study of mycophenolate mofetil (MMF). The electrochemically reduced graphene oxide (ERGO) film on glassy carbon electrode (GCE) showed enhanced peak current for electrooxidation of MMF. MMF exhibited two irreversible oxidation peaks at 0.84 V (peak a1) and 1.1 V (peak a2). Effects of accumulation time, pH and scan rate were studied and various electrochemical parameters were calculated. A differential pulse voltammetric method was developed for the determination of MMF in bulk samples and pharmaceutical formulations. Linear relationship was observed between the peak current and concentration of MMF in the range of 40 nM-15 μM with a limit of detection of 11.3 nM. The proposed method is simple, sensitive and inexpensive and, hence, could be readily adopted in clinical and quality control laboratories.

Project description:Hand sanitizer usage has proven to be a common and practical method for reducing the spread of infectious diseases which can be caused by many harmful pathogens. There is a need for alcohol-free hand sanitizers because most hand sanitizers on the market are alcohol-based, and regular use of them can damage the skin and can be hazardous. India is the world's largest producer of fruits and one of the major problems after fruit consumption is their peels, causing waste management problems and contributing to the formation of greenhouse gases leading to air pollution and adding to the problem of climate change. Valorization of such wastes into other value-added products and their incorporation into formulations of eco-friendly alcohol-free hand sanitizers would solve these issues, save the environment, benefit the society, and help in achieving the sustainable development goals. Thus, this research focuses on formulating an effective natural alcohol-free hand sanitizer that harnesses the antimicrobial properties of the various types of bioactive components found in fruit peels of pomegranate, sweet lime, and lemon. The peel extracts and the formulated sanitizer proved considerable antimicrobial activity against the pathogenic Escherichia coli and hand microflora. Molecular docking was also applied to examine ligand-protein interaction patterns and predict binding conformers and affinity of the sanitizer phytocompounds towards target proteins in COVID-19, influenza, and pneumonia viruses. The binding affinities and the protein-ligand interactions virtual studies revealed that the sanitizer phytocompounds bind with the amino acids in the target proteins' active sites via hydrogen bonding interactions. As a result, it is possible to formulate a natural, alcohol-free hand sanitizer from fruit peels that is effective against pathogenic germs and viruses using the basic structure of these potential findings.

Project description:Four precise, accurate, selective, and sensitive UV-spectrophotometric methods were developed and validated for the simultaneous determination of a binary mixture of Oxytetracycline HCl (OXY) and Flunixin Meglumine (FLU). The first method, dual wavelength (DW), depends on measuring the difference in absorbance (ΔA 273.4-327 nm) for the determination of OXY where FLU is zero while FLU is determined at ΔA 251.7-275.7 nm. The second method, first-derivative spectrophotometric method (1D), depends on measuring the peak amplitude of the first derivative selectively at 377 and 266.7 nm for the determination of OXY and FLU, respectively. The third method, ratio difference method, depends on the difference in amplitudes of the ratio spectra at ΔP 286.5-324.8 nm and ΔP 249.6-286.3 nm for the determination of OXY and FLU, respectively. The fourth method, first derivative of ratio spectra method (1DD), depends on measuring the amplitude peak to peak of the first derivative of ratio spectra at 296.7 to 369 nm and 259.1 to 304.7 nm for the determination of OXY and FLU, respectively. Different factors affecting the applied spectrophotometric methods were studied. The proposed methods were validated according to ICH guidelines. Satisfactory results were obtained for determination of both drugs in laboratory prepared mixture and pharmaceutical dosage form. The developed methods are compared favourably with the official ones.

Project description:Detergent formulations for cleaning a carbonized soil-degreasers-typically comprise surfactants, organic solvents, phosphate-based cleaning agents, and alkaline agents, which results in high pH values (>11) that raise human and environmental risks. It is important to develop eco-friendly and safer degreasers, while maintaining their cleaning efficiency. In this work, simple degreaser formulations, with a pH below 11 and without phosphates, were developed by using a mixture of solvent, surfactant, and water to remove carbonized soil. The efficiency of the new degreaser formulations (with 5 wt% solvent, 5 wt% nonionic or ionic surfactant, and 90 wt% water) was evaluated by an abrasion test in the removal of carbonized soil from ceramic and stainless steel surfaces and compared with a commercial product. The results obtained show that the formulations comprising isopropylene glycol (IPG) with C11-C13 9EOs and diethylene glycol butyl ether (BDG) with octyltrimethylammonium octanoate ([N1118][C8O2]) present the best cleaning efficiency for both surfaces. The composition of these formulations was optimized for each surface using a mixture design. The resulting formulations, despite having a simpler composition, a pH lower than 11, and being phosphate-free, presented a cleaning efficiency equal or slightly higher than the commercial control. These results show that it is possible to design degreasers that are much less aggressive to the environment and user, while simultaneously fulfilling the market requirements.

Project description:Chlorphenoxamine hydrochloride is a chemical that has attracted interest because of its notable anti-histaminic and anti-cholinergic characteristics. Moreover, it has been recognized as a highly efficient tool in the fight against several lethal viral diseases, such as Severe Acute Respiratory Syndrome Coronavirus. In this study, five efficient and straightforward univariate spectrophotometric approaches are proposed for accurately measuring the quantities of Caffeine and Chlorphenoxamine HCl in bulk forms and combined pharmaceutical formulations. Notably, these methods based on an advanced approach using the factorized response spectrum and not require any initial processing.They have been classified into three spectrophotometric platform windows. The study of Window I focuses on absorption spectra of substances in their original states(zero-order). It includes, absorbance resolution method (AR), extended absorbance difference method (EAD), and factorized zero order method (FZM). Window II focuses on the factorized derivative method (FDM), while Window III focuses on factorized ratio difference method (FRM). These approaches successfully measured concentration of Caffeine and Chlorphenoxamine HCl within a range of 3-35.0 and 3-45.0 μg/mL, respectively. The factorized response spectrum's exclusivity stems from its capacity to fully separate the mentioned components in mixture and recover the pure spectra. Validation of the suggested approaches has been conducted according to guidelines established by International Council for Harmonization, which demonstrated acceptable levels of accuracy and precision. The scope of this work has been expanded to include verification of content uniformity of dosage units according to recommendations outlined in United States Pharmacopoeia. Greenness profile of the proposed approaches has been properly assessed using state-of-the-art software metrics, in comparison to the reported one. Finally, the proposed methods demonstrated strong compliance with the recently established principles of white field of analytical chemistry.

Project description:Considering the green chemistry perspective and improving the environmental impact of quality control labs; two direct techniques with less hazardous solvents, less waste production and less energy consumption were developed for simultaneous analysis of Aspirin and Metoclopramide in bulk powder and pharmaceutical formulation. The ratio between the two drugs in their co-formulated preparation is very challenging; (90: 1, Aspirin: Metoclopramide). The first technique is spectrophotometry using simple mathematical operations; ratio difference and derivative ratio-zero crossing. The second technique is high-performance thin-layer chromatography (HPTLC) -densitometry which used a mobile phase consisting of cyclo-hexane: methanol: methylene chloride in a ratio of (1:4:1, v/v/v). The greenest solvents which give acceptable resolution were chosen. Following the International Conference on Harmonization (ICH) guidelines, the methods were found to be accurate, precise, and selective. Those methods were statistically compared to the reported spectrophotometric method and the results proved that there is no significant difference in accuracy and precision. Furthermore, the developed methods were assessed using the Analytical Eco-scale, Green Analytical Procedure Index (GAPI) and the Analytical Greenness calculator (AGREE), which gave a full image about their greenness profile. The spectrophotometry was found to be an excellent green technique compared to HPTLC with was considered an acceptable green one. The developed HPTLC-densitometric method was used for the first time for the analysis of this binary mixture. The two proposed spectrophotometric methos have advantages over the published methods as they used easy manipulation steps and are applied on the market pharmaceutical formulation. Owing to the advantages of the developed techniques; being green, do not require expensive sophisticated equipment or large volume of solvents; they could be used for routine analysis in quality control aspects.
