Taxonomy of Tobrilidae species from the Alkaline Lakes of the western Nebraska Sandhills.

ABSTRACT: Six distinct COI mitochondrial Haplotype Groups (HG) are morphologically, ecologically, and genetically characterized from the aquatic nematode family Tobrilidae. Collection locations included the extreme habitats of the Alkaline Lakes in the western Nebraska Sandhills and the contaminated stream, Johnson Creek, bordering the AltEn 2021 catastrophic pesticide release near the village of Mead in eastern Nebraska. Maximum likelihood and genetic distance metrics supported the genetic integrity of the haplotype groups. Discriminant function analysis of COI haplotype group datasets of combined morphological characters and soil chemistry attributes for both male and female Tobrilidae were classified correctly in all but one case. Scanning electron microscopy revealed new details about amphid apertures, male supplements, and spicules. Partial 18S gene phylogeny suggests that the genus Semitobrilus may not be a member of the subfamily Neotobrilinae, and three specimens in the 226 tobrilid dataset provide evidence of incongruence between COI and 18S derived phylogenies. Given the strong signal provided by the environmental chemistry data, tobrilid mitochondrial haplotypes may well have value as environmental indicators.

Project description:The Sandhills of Nebraska is a complex ecosystem, covering 50,000 km2 in central and western Nebraska and predominantly of virgin grassland. Grasslands are the most widespread vegetation in the U.S. and once dominated regions are currently cultivated croplands, so it stands to reason that some of the current plant pathogens of cultivated crops originated from grasslands, particularly soilborne plant pathogens. The anamorphic genus Rhizoctonia includes genetically diverse organisms that are known to be necrotrophic fungal pathogens, saprophytes, mycorrhiza of orchids, and biocontrol agents. This study aimed to evaluate the diversity of Rhizoctonia spp. on four native grasses in the Sandhills of Nebraska and determine pathogenicity to native grasses and soybean. In 2016 and 2017, a total of 84 samples were collected from 11 sites in the Sandhills, located in eight counties of Nebraska. The samples included soil and symptomatic roots from the four dominant native grasses: sand bluestem, little bluestem, prairie sandreed, and needle-and-thread. Obtained were 17 Rhizoctonia-like isolates identified, including five isolates of binucleate Rhizoctonia AG-F; two isolates each from binucleate Rhizoctonia AG-B, AG-C, and AG-K, Rhizoctonia solani AGs: AG-3, and AG-4; one isolate of binucleate Rhizoctonia AG-L, and one isolate of R. zeae. Disease severity was assessed for representative isolates of each AG in a greenhouse assay using sand bluestem, needle-and-thread, and soybean; prairie sandreed and little bluestem were unable to germinate under artificial conditions. On native grasses, all but two isolates were either mildly aggressive (causing 5-21% disease severity) or aggressive (21-35% disease severity). Among those, three isolates were cross-pathogenic on soybean, with R. solani AG-4 shown to be highly aggressive (86% disease severity). Thus, it is presumed that Rhizoctonia spp. are native to the sandhills grasslands and an emerging pathogen of crops cultivated may have survived in the soil and originate from grasslands.

Project description:Repeated use of field corn (Zea mays L.) hybrids expressing the Cry3Bb1 and mCry3A traits in Nebraska has selected for field-evolved resistance in some western corn rootworm (WCR; Diabrotica virgifera virgifera LeConte) populations. Therefore, this study was conducted to characterize spatial variation in local WCR susceptibility to Cry3Bb1 and mCry3A traits in Keith and Buffalo counties, Nebraska, and determine the relationship between past management practices and current WCR susceptibility. Adult WCR populations were collected from sampling grids during 2015 and 2016 and single-plant larval bioassays conducted with F1 progeny documented significant variation in WCR susceptibility to Cry3Bb1 and mCry3A on different spatial scales in both sampling grids. At the local level, results revealed that neighboring cornfields may support WCR populations with very different susceptibility levels, indicating that gene flow of resistant alleles from high trait survival sites is not inundating large areas. A field history index, comprised of additive and weighted variables including past WCR management tactics and agronomic practices, was developed to quantify relative selection pressure in individual fields. The field history index-Cry3 trait survivorship relationship from year 1 data was highly predictive of year 2 Cry3 trait survivorship when year 2 field history indices were inserted into the year 1 base model. Sensitivity analyses indicated years of trait use and associated selection pressure at the local level were the key drivers of WCR susceptibility to Cry3 traits in this system. Retrospective case histories from this study will inform development of optimal resistance management programs and increase understanding of plant-insect interactions that may occur when transgenic corn is deployed in the landscape. Results from this study also support current recommendations to slow or mitigate the evolution of resistance by using a multi-tactic approach to manage WCR densities in individual fields within an integrated pest management framework.

Project description:The wood-inhabiting fungi play an integral role in wood degradation and the cycle of matter in the ecological system. They are considered as the "key player" in wood decomposition, because of their ability to produce lignocellulosic enzymes that break down woody lignin, cellulose and hemicellulose. In the present study, four new wood-inhabiting fungal species, Adustochaetealbomarginata, Ad.punctata, Alloexidiopsisgrandinea and Al.xantha collected from southern China, are proposed, based on a combination of morphological features and molecular evidence. Adustochaetealbomarginata is characterised by resupinate basidiomata with cream to buff, a smooth, cracked, hymenial surface, a monomitic hyphal system with clamped generative hyphae and subcylindrical to allantoid basidiospores (12-17.5 × 6.5-9 µm). Adustochaetepunctata is characterised by resupinate basidiomata with cream, a smooth, punctate hymenial surface, a monomitic hyphal system with clamped generative hyphae and subcylindrical to allantoid basidiospores (13.5-18 × 6-8.2 µm). Alloexidiopsisgrandinea is characterised by resupinate basidiomata with buff to slightly yellowish, a grandinioid hymenial surface, a monomitic hyphal system with clamped generative hyphae and allantoid basidiospores (10-12.3 × 5-5.8 µm). Additionally, Alloexidiopsisxantha is characterised by resupinate basidiomata with cream to slightly buff, a smooth hymenial surface, a monomitic hyphal system with clamped generative hyphae and subcylindrical to allantoid basidiospores measuring 20-24 × 5-6.2 µm. Sequences of the internal transcribed spacers (ITS) and the large subunit (nrLSU) of the nuclear ribosomal DNA (rDNA) markers of the studied samples were generated. Phylogenetic analyses were performed with the Maximum Likelihood, Maximum Parsimony and Bayesian Inference methods. The phylogram, based on the ITS+nLSU rDNA gene regions, revealed that four new species were assigned to the genera Adustochaete and Alloexidiopsis within the order Auriculariales, individually. The phylogenetic tree inferred from the ITS sequences highlighted that Ad.albomarginata was retrieved as a sister to Ad.yunnanensis and the species Ad.punctata was sister to Ad.rava. The topology, based on the ITS sequences, showed that Al.grandinea was retrieved as a sister to Al.schistacea and the taxon Al.xantha formed a monophyletic lineage. Furthermore, two identification keys to Adustochaete and Alloexidiopsis worldwide are provided.
