Project description:It is highly desirable, while still challenging, to obtain noble metal nanocrystals with custom capping ligands, because their colloidal synthesis relies on specific capping ligands for the shape control while conventional ligand exchange processes suffer from "the strong replaces the weak" limitation, which greatly hinders their applications. Herein, we report a general and effective ligand exchange approach that can replace the native capping ligands of noble metal nanocrystals with virtually any type of ligands, producing flexibly tailored surface properties. The key is to use diethylamine with conveniently switchable binding affinity to the metal surface as an intermediate ligand. As a strong ligand, it in its original form can effectively remove the native ligands; while protonated, it loses its binding affinity and facilitates the adsorption of new ligands, especially weak ones, onto the metal surface. By this means, the irreversible order in the conventional ligand exchange processes could be overcome. The efficacy of the strategy is demonstrated by mutual exchange of the capping ligands among cetyltrimethylammonium, citrate, polyvinylpyrrolidone, and oleylamine. This novel strategy significantly expands our ability to manipulate the surface property of noble metal nanocrystals and extends their applicability to a wide range of fields, particularly biomedical applications.

Project description:Surface modification with functional molecules is essential for introducing various surface properties. As gold nanoparticles (AuNPs) have extraordinary chemical, physical, and optical properties, control of their surface, mainly through modification with mixed alkanethiols via Au-S interactions, has attracted much attention. However, surface modification of AuNPs with mixed alkanethiols to provide a strictly regulated composition remains challenging. Further, there are very few methods that can easily establish the nature of ligands and their replacement with similar molecules at nanoparticle surfaces, limiting precise analyses. Herein, we demonstrate an unfair ligand exchange between oligo(ethylene glycol) (OEG)-attached alkanethiols as a source of unfair surface modification utilizing programable thermo-responsive properties of OEG-alkanethiols-modified AuNPs and fair surface modification with mixed OEG-alkanethiols by minimizing this effect. OEG-alkanethiols-modified AuNPs show an assembly/disassembly behavior in response to the solution temperature. Assembly temperature (T A) changes in the presence of other OEG-alkanethiols, confirming the ligand exchange between alkanethiols in an aqueous solution. Kinetic analyses indicate that the competitive exchange reaction of these two alkanethiols results in an unfair ligand exchange, which leads to gradual changes in surface composition. As this ligand exchange between alkanethiols takes a longer time compared to that from citric acid, which initially covered the AuNPs, exact surface modification of AuNPs with OEG-alkanethiols is performed by moderate reaction conditions (25 °C, several to 24 hours). This insight regarding "more prolonged reaction is not always better" could be widely applied for surface modifications with various thiol-ligands.

Project description:We show the formation of metallic spikes on the surface of gold nanotriangles (AuNTs) by using the same reduction process which has been used for the synthesis of gold nanostars. We confirm that silver nitrate operates as a shape-directing agent in combination with ascorbic acid as the reducing agent and investigate the mechanism by dissecting the contribution of each component, i.e., anionic surfactant dioctyl sodium sulfosuccinate (AOT), ascorbic acid (AA), and AgNO3. Molecular dynamics (MD) simulations show that AA attaches to the AOT bilayer of nanotriangles, and covers the surface of gold clusters, which is of special relevance for the spike formation process at the AuNT surface. The surface modification goes hand in hand with a change of the optical properties. The increased thickness of the triangles and a sizeable fraction of silver atoms covering the spikes lead to a blue-shift of the intense near infrared absorption of the AuNTs. The sponge-like spiky surface increases both the surface enhanced Raman scattering (SERS) cross section of the particles and the photo-catalytic activity in comparison with the unmodified triangles, which is exemplified by the plasmon-driven dimerization of 4-nitrothiophenol (4-NTP) to 4,4'-dimercaptoazobenzene (DMAB).

Project description:Using the previously designed biphenyl-2-ylphosphine ligand, featuring a remote tertiary amino group, the first gold-catalyzed intermolecular hydroalkenylation of alkynes has been developed. Synthetically valuable conjugated dienyl alcohols are formed in moderate to good yields. A range of alkenyltrifluoroborates are allowed as the alkenyl donor, and no erosion of alkene geometry and/or the propargylic configuration are detected. DFT calculations confirm the critical role of the remote basic group in the ligand as a general-base catalyst for promoting this novel gold catalysis with good efficiency.

Project description:Ochratoxin A (OTA) is a type of mycotoxin generated from the metabolism of Aspergillus and Penicillium, and is extremely toxic to humans, livestock, and poultry. However, traditional assays for the detection of OTA are expensive and complicated. Other than OTA aptamer, OTA itself at high concentration can also adsorb on the surface of gold nanoparticles (AuNPs), and further inhibit AuNPs salt aggregation. We herein report a new OTA assay by applying the localized surface plasmon resonance effect of AuNPs and their aggregates. The result obtained from only one single linear calibration curve is not reliable, and so we developed a "double calibration curve" method to address this issue and widen the OTA detection range. A number of other analytes were also examined, and the structural properties of analytes that bind with the AuNPs were further discussed. We found that various considerations must be taken into account in the detection of these analytes when applying AuNP aggregation-based methods due to their different binding strengths.

Project description:A non-labeled, portable plasmonic biosensor-based device was developed to enable the ultra-sensitive and selective detection of Salmonella typhimurium in pork meat samples. Specifically, a plasmonic sensor, using the self-assembly of gold nanoparticles (AuNPs) to achieve a regulated diameter of 20 nm for the AuNP monolayers, was used to conduct high-density deposition on a transparent substrate, which produced longitudinal wavelength extinction shifts via a localized surface plasmon resonance (LSPR) signal. The developed aptamers conjugated to the LSPR sensing chips revealed an ultra-sensitive upper limit of detection (LOD) of approximately 104 cfu/mL for S. typhimurium in pure culture under the optimal assay conditions, with a total analysis time of 30-35 min. When the LSPR sensing chips were applied on artificially contaminated pork meat samples, S. typhimurium in the spiked pork meat samples was also detected at an LOD of 1.0 × 104 cfu/mL. The developed method could detect S. typhimurium in spiked pork meat samples without a pre-enrichment step. Additionally, the LSPR sensing chips developed against S. typhimurium were not susceptible to any effect of the food matrix or background contaminant microflora. These findings confirmed that the developed gold nanoparticle-aptamer-based LSPR sensing chips could facilitate sensitive detection of S. typhimurium in food samples.

Project description:The influence of heat generation on the conventional process of LSPR based sensing has not been explored thus far. Therefore, a need exists to draw attention toward the heat generation issue during LSPR sensing as it may affect the refractive index of the analyte, leading to incorrect sensory conclusions. This manuscript addresses the connection between the photo-thermal effect and LSPR. We numerically analyzed the heat performance of a gold cladded nanotip. The numerical results predict a change in the micro-scale temperature in the microenvironment near the nanotip. These numerical results predict a temperature increase of more than 20 K near the apex of the nanotip, which depends on numerous factors including the input optical power and the diameter of the fiber. We analytically show that this change in the temperature influences a change in the refractive index of the microenvironment in the vicinity of the nanotip. In accordance with our numerical and analytical findings, we experimentally show an LSPR shift induced by a change in the input power of the source. We believe that our work will bring the importance of temperature dependence in nanotip based LSPR sensing to the fore.

Project description:In the past decade, there has been considerable interest in radiosensitization using gold nanoparticles that accumulate specifically in cancerous tissue while sparing normal tissues. Despite this interest, it remains unclear which nanoparticle morphologies, cellular uptake, or cytoplasmic distribution elicit optimal radiosensitization. We introduce gold nanotriangles (AuNTs) as a possible X-ray radiotherapy sensitizer. In this study, we first explored a large-scale synthetic method for the production of high quality monodisperse AuNTs. Second, we conducted in vitro and in vivo experiments to evaluate the effect of PEGylated AuNTs (pAuNTs) on cellular uptake, cytotoxicity, bio-distribution, and radiosensitization on radiation-resistant human Glioblastoma Multiforme (GBM) cells. Our results suggest that the new scale up synthesis methodology consistently produced high quality AuNTs and pAuNTs which had nonspecific cellular uptake without any obvious cytotoxicity and exhibited excellent radiosensitization.

Project description:We report an efficient, seed-mediated method for the synthesis of gold nanotriangles (NTs) which can be used for controlled self-assembly. The main advantage of the proposed synthetic protocol is that it relies on using stable (over the course of several days) intermediate seeds. This stability translates into increasing time efficiency of the synthesis and makes the protocol experimentally less demanding ('fast addition' not required, tap water can be used in the final steps) as compared to previously reported procedures, without compromising the size and shape monodispersity of the product. We demonstrate high reproducibility of the protocol in the hands of different researchers and in different laboratories. Additionally, this modified seed-mediated method can be used to produce NTs with edge lengths between ca. 45 and 150 nm. Finally, the high 'quality' of NTs allows the preparation of long-range ordered assemblies with vertically oriented building blocks, which makes them promising candidates for future optoelectronic technologies.

Project description:In this study, thin films composed of gold nanoparticles embedded in a copper oxide matrix (Au:CuO), manifesting Localized Surface Plasmon Resonance (LSPR) behavior, were produced by reactive DC magnetron sputtering and post-deposition in-air annealing. The effect of low-power Ar plasma etching on the surface properties of the plasmonic thin films was studied, envisaging its optimization as gas sensors. Thus, this work pretends to attain the maximum sensing response of the thin film system and to demonstrate its potential as a gas sensor. The results show that as Ar plasma treatment time increases, the host CuO matrix is etched while Au nanoparticles are uncovered, which leads to an enhancement of the sensitivity until a certain limit. Above such a time limit for plasma treatment, the CuO bonds are broken, and oxygen is removed from the film’s surface, resulting in a decrease in the gas sensing capabilities. Hence, the importance of the host matrix for the design of the LSPR sensor is also demonstrated. CuO not only provides stability and protection to the Au NPs but also promotes interactions between the thin film’s surface and the tested gases, thereby improving the nanocomposite film’s sensitivity. The optimized sensor sensitivity was estimated at 849 nm/RIU, which demonstrates that the Au-CuO thin films have the potential to be used as an LSPR platform for gas sensors.
