Statistical optimization of process variables for enhanced serratiopeptidase production from soil Serratia marcescens VS56.

ABSTRACT: Serratiopeptidase was recognized as a clinically valuable proteolytic enzyme due to its potent anti-inflammatory and analgesic properties, widely employed in pharmaceutical formulations for the treatment of arthritis, sinusitis, and post-operative swelling. The present study focused on enhancing the production and purification of serratiopeptidase from Serratia marcescens VS56 (OQ859929). Initially, a One-Factor-At-a-Time (OFAT) approach was used to evaluate the effects of production parameters including carbon and nitrogen sources, pH, and temperature on enzyme production. This was followed by statistical optimization using Response Surface Methodology (RSM) and the Box-Behnken design. Glucose and beef extract, at pH 7 ± 0.3 and 37 °C, were identified as the optimal conditions, under whic
