Mutation in enterovirus 71 nonstructural protein 3A increases genome replication fidelity and exhibits attenuated virulence in mice.

ABSTRACT: Increasing numbers of studies have highlighted the important implications of the replication fidelity of RNA viruses for virus replication, pathogenesis, and the development of antiviral drugs and live-attenuated vaccines. However, most of the research has focused on viral polymerase, and little information is available about the potential role of other viral replicase proteins. Here, we demonstrated that the mutations of 3AV75A and 3DV63A+M393L in enterovirus 71 (EV71) conferred high-fidelity phenotypes through deep sequencing of virus populations in cell culture and animal models of infection. The 3AV75A, 3DV63A+M393L, and 3AV75A-3DV63A+M393L high-fidelity variants were highly attenuated in immunocompetent suckling mice. T
