Transcriptome-wide analysis of AlbHLH gene family in Atractylodes lancea (Thunb.) DC and functional exploration of AlbHLH3 in regulating terpenoid biosynthesis.

ABSTRACT:

Background

Atractylodes lancea (Thunb.) DC., a medicinal species of Asteraceae with multi-pharmacological applications in rheumatic disorders and gastrointestinal syndromes due to its bioactive terpenoid-enriched essential oils. Although basic helix-loop-helix (bHLH) transcription factors are well known regulators of secondary metabolism biosynthesis pathways, their functional characterization in terpenoid regulation in A. lancea remains unexplored.

Results

176 AlbHLHs were systematically identified through multi-tissue transcriptomic profiling (leaf, stem, rhizome, rhizome buds) and categorized them into 21 evolutionarily conserved subfamilies via phylogenetic analysis with Arabidopsis thaliana bHLH proteins. Conserved motif architecture revealed high intra-subfamily conser
