Spatial- and Phospho-Proteomic Profiling Reveals Pancreatic and Hepatic Dysfunction in a Rat Model of Lethal Insulin Overdose.

ABSTRACT: Insulin, a pivotal hormone synthesized by the pancreas and regulated through hepatic first-pass metabolism, plays an essential role in the management of diabetes. However, non-therapeutic exposure to insulin can lead to life-threatening hypoglycemia. The postmortem diagnosis of fatalities resulting from exogenous insulin presents numerous forensic challenges, including the disruption of pharmacokinetic evidence due to the rapid degradation of insulin after death and the lack of pathognomonic histopathological markers. These factors create significant obstacles in establishing medicolegal causality. Furthermore, the mechanisms underlying insulin overdose-induced injury to the pancreas and liver are poorly understood. This study aims to address these gaps by integrating standardized histopat
