ABSTRACT: Multimodal analgesia and rational polypharmacy have emerged as modern pain management strategies, offering synergistic pain relief while mitigating the adverse effects of high-dose monotherapy. A novel antinociceptive fixed-dose combination tablet containing tramadol, ibuprofen, and caffeine exemplifies this approach by integrating a centrally acting weak opioid tramadol, a peripherally acting nonsteroidal anti-inflammatory drugs ibuprofen, and a central nervous system stimulant adjuvant caffeine into a single formulation.
