Effects of dietary fat source and subtherapeutic levels of antibiotic on the bacterial community in the ileum of broiler chickens at various ages.

ABSTRACT: The effect of dietary fat source (soy oil or a mixture of lard and tallow) and dietary supplementation with antibiotics (a combination of avilamycin at 10 mg kg of feed(-1) and salinomycin at 40 mg kg of feed(-1)) on the bacterial community in the ileum of broiler chickens at different ages (7, 14, 21, and 35 days) was studied using PCR with denaturing gradient gel electrophoresis (DGGE) analysis and bacteriological culture. The bacterial origin of fragments in DGGE profiles was identified by sequencing. Bacterial enumeration results, together with PCR-DGGE profiles, showed that the composition of the microflora was age dependent and influenced by dietary fat source and antibiotic supplementation. An increased incidence of streptococci, enterobacteria, and Clostridium perfringens with age of the chickens was demonstrated. Lactobacilli and C. perfringens were the bacterial groups most strongly affected by the dietary treatments. Moreover, different strains (clonal variants of the alpha-toxin gene) of C. perfringens type A were detected in response to age, dietary fat source, and dietary supplementation with antibiotics.

Project description:BackgroundRestaurant oil in poultry diets increases energy content, reduces production costs, and promotes sustainability within the food supply chain. However, variable oil composition and heating temperatures among restaurant oil sources can impact broiler chicken health due to heat-induced lipid modifications.ObjectivesA 21-d experiment was conducted to evaluate ileal morphology, liver cytokine gene expression, and ileal immune cell populations in broilers fed control or peroxidized lipids with varying chain and saturation characteristics.MethodsDay-old broilers were housed in battery cages (5 birds per cage) and fed diets containing 5% control or peroxidized oils. Eight diets were randomly assigned in a 4 × 2 factorial arrangement consisting of oil source (palm, soybean, flaxseed, or fish) and peroxidation status (control or peroxidized). At day 21, samples were collected for ileal histomorphology [villus height (VH), crypt depth (CrD), and the VH:CrD ratio], and liver cytokine expression (qPCR). Ileum cytokine expression and T-cell markers were analyzed by RNAscope in situ hybridization (ISH). Data were analyzed as a mixed model (SAS 9.4) with fixed effects of lipid source, peroxidation, and lipid × peroxidation interaction.ResultsCD3+ T-cells in the ileum decreased 16.2% due to peroxidation (P = 0.001) with 30.3% reductions observed in birds fed peroxidized flaxseed oil (P = 0.01). Peroxidation increased IL6+ and IL1B+ cells by 62.0% and 40.3%, respectively (P = 0.01). Soybean oil increased IFNG+ cells by 55.1% compared with palm oil, regardless of peroxidation status (P = 0.007). Lipid source and peroxidation did not alter ileal histomorphology or liver cytokine expression.ConclusionsLipid peroxidation increased ileal IL1B and IL6 in broiler chickens, whereas soybean oil diets increased IFNG. Generally, peroxidation decreased overall CD3+ T-cell populations, suggesting impaired T-cell presence or recruitment. These results identify potential immunomodulatory lipid profiles in restaurant oil while supporting RNAscope-ISH as a method to describe avian tissue-level immune responses.

Project description:BackgroundBaicalin and probiotic cocktails are promising feed additives with broad application prospects. While probiotic cocktails are known to enhance intestinal health, the potential synergistic impact of combining baicalin with probiotic cocktails on the gut health of broiler chickens remains largely unexplored. Therefore, this study aims to investigate the influence of the combined administration of baicalin and probiotic cocktails on the composition of ileal and cecal microbiota in broiler chickens to elucidate the underlying mechanisms responsible for the health-promoting effects.ResultsA total of 320 1-day-old male Arbor Acres broilers were divided into 4 groups, each with 8 replicates of 10 chicks per replicate. Over a period of 42 d, the birds were fed a basal diet or the same diet supplemented with 37.5 g/t baicalin (BC), 1,000 g/t probiotic cocktails (PC), or a combination of both BC (37.5 g/t) and PC (1,000 g/t). The results demonstrated that BC + PC exhibited positive synergistic effects, enhancing intestinal morphology, immune function, and barrier function. This was evidenced by increased VH/CD ratio, sIgA levels, and upregulated expression of occludin and claudin-1 (P < 0.05). 16S rRNA analysis indicated that PC potentiated the effects of BC, particularly in the ileum, where BC + PC significantly increased the α-diversity of the ileal microbiota, altered its β-diversity, and increased the relative abundance of Flavonifractor (P < 0.05), a flavonoid-metabolizing bacterium. Furthermore, Flavonifractor positively correlated with chicken ileum crypt depth (P < 0.05). While BC + PC had a limited effect on cecal microbiota structure, the PC group had a very similar microbial composition to BC + PC, suggesting that the effect of PC at the distal end of the gut overshadowed those of BC.ConclusionsWe demonstrated the synergistic enhancement of gut health regulation in broiler chickens by combining baicalin and probiotic cocktails. Probiotic cocktails enhanced the effects of baicalin and accelerated its metabolism in the ileum, thereby influencing the ileal microbiota structure. This study elucidates the interaction mechanism between probiotic cocktails and plant extract additives within the host microbiota. These findings provide compelling evidence for the future development of feed additive combinations.

Project description:Diet has been found to significantly influence gut microbiota throughout various life stages, and gut microbiota have been increasingly shown to influence host physiology, health, and behavior. This study uses 16S rRNA sequencing to examine the effects of six different fat-supplemented diets (canola oil, coconut oil, fish oil, flaxseed oil, lard, and olive oil) on broiler chicken cecal microbial composition and predicted function in comparison with a common and inexpensive fat source (poultry fat). Groups of broilers were fed each of these diets and then evaluated on day 41 and day 55 of age. For both 41- and 55-day samples, Firmicutes and Bacteroidetes phyla were the dominant bacteria in the ceca accounting for 99% of the microbial community. Across the 41- and 55-day samples, treatment time was associated with a stronger and more significant microbiota shift (p < 0.001) than differences in dietary treatment alone (p = 0.117), but dietary treatment combined with treatment time is a significant factor as well (p = 0.047). Sparse partial least squares discriminant analysis was used to explore the more discriminating taxa for each treatment group. For identified species, butyrate production appears to be affected in a diet-specific manner, with many butyrate-producing species being evident for the fish-based diet at day 41 and a few of these species for the flaxseed-based diet at day 55. Predicted functions, as conducted with PICRUSt2, were significant for comparisons between the control and the flaxseed-based dietary treatment group at day 55, with indications of host health benefit for the flaxseed-based diet. Predicted functions found to be significant were for enzymes and pathways such as propionate CoA ligase, aminobutyraldehyde dehydrogenase, vitamin B12-transporting ATPase, thiamine kinase, acetylneuraminate epimerase, and L-tryptophan biosynthesis. This study provides insight surrounding specific dietary fat-based treatments to be investigated further and highlights the importance of polyunsaturated fat sources in poultry feed that may offer a favorable cecal microbial modulation compared to saturated fat sources.

Project description:As an essential micronutrient for animals, vitamin E plays crucial physiological roles in reproduction, antioxidant and immune functions, and lipid metabolism. The objective of this study was to reveal molecular mechanism of vitamin E on intramuscular fat (IMF) deposition through transcriptome sequencing of pectoral muscle in broiler chickens. A total of 240 one-day-old health female chicks were randomly allocated into five dietary treatments with each treatment six replicates. The birds were fed basal diet supplemented with 0 and 100 IU/kg vitamin E in the form of DL-α-tocopheryl acetate, respectively. The body weight, carcass performance and IMF content were measured. Transcriptome profile of pectoral muscle in 35-day-old chickens were sequencing from the control and 100 IU/kg vitamin E treatment. Functional enrichment analyzes of differentially expressed genes (DEGs) based on Gene ontology (GO), KEGG pathway and bio function, and network were performed. Results shown that IMF content of broiler chickens were significantly increased at 12.89% (P < 0.05) between 100 IU/kg vitamin E treatment and control. Transcriptome sequencing results for pectoralis major muscle of 100 IU vitamin E-supplemented and the control groups identified 57 up-regulated and 102 down-regulated DEGs. These DEGs were significantly enriched (FDR corrected P-value < 0.05) in 13 of 236 GO terms involved in muscle development- and lipid metabolism. Pathway functional enrichment analysis revealed that the DEGs were significantly enriched in three signalling pathways (FDR corrected P-value < 0.05). Two of them, MAPK signaling pathway and FoxO signaling pathway, play key roles in muscular and lipid metabolism. It is worth mentioning that 46 DEGs were significantly enriched in 28 skeletal and muscular system development and function categories and 31 DEGs were significantly enriched in 17 lipid metabolism function categories. Moreover, three lipid metabolism and muscular development-related networks of DEGs were also identified. These DEGEs, pathways, function categories and networks identified in this study provide us new insights for the vitamin E regulation of the IMF deposition in broiler chickens.
