Mate choice assays and mating propensity differences in natural yeast populations.

ABSTRACT: In sexual microbes, mating occurs by fusion of individual cells. This complete fitness investment suggests that cell behaviour could potentially mediate prezygotic isolation between microbial species, a topic about which very little is known. To investigate this possibility, we conducted individual cell mate choice trials and mass-culture mating propensity assays with isolates from sympatric natural populations of the closely related yeasts Saccharomyces cerevisiae and Saccharomyces paradoxus. Although we found no evidence for active species recognition in mate choice, we observed a marked difference in mating propensity between these two species. We briefly discuss the possibility that this mating propensity difference may contribute to reproductive isolation between S. cerevisiae and S. paradoxus in nature.

Mating behaviour, mate choice and female resistance in the bean flower thrips (Megalurothrips sjostedti).

Project description:Many species of thrips (Thysanoptera) in the family Thripidae form mating aggregations, but the adaptive significance of these aggregations and the extent of male and female mate choice is poorly understood. We studied the mating behaviour of the bean flower thrips Megalurothrips sjostedti (Trybom) (Thysanoptera: Thripidae), which forms male aggregations and occurs across sub-Saharan Africa. We tested whether males choose mates by female age or mating status. No-choice mating bioassays with one male and one female were used to simulate the way males usually encounter only one female at a time in aggregations in the field. Virgin females violently resisted mating attempts by males, but we found no compelling evidence to establish whether this was indiscriminate or was screening suitable males. Younger males (1-2 days old) did not discriminate females by age (1-2 or 7-10 days old), but older males (7-10 days old) avoided mating with older females. Any male choice by female mating status (virgin or mated) was weak or absent. The mating behaviour of M. sjostedti shows broad similarities with that of other thrips species that form aggregations, but also shows some distinct and novel differences, which can help our understanding of the adaptive significance of aggregations.

Facultative pupal mating in Heliconius erato: Implications for mate choice, female preference, and speciation.

Project description:Mating systems have broad impacts on how sexual selection and mate choice operate within a species, but studies of mating behavior in the laboratory may not reflect how these processes occur in the wild. Here, we examined the mating behavior of the neotropical butterfly Heliconius erato in the field by releasing larvae and virgin females and observing how they mated. H. erato is considered a pupal-mating species (i.e., males mate with females as they emerge from the pupal case). However, we observed only two teneral mating events, and experimentally released virgins were almost all mated upon recapture. Our study confirms the presence of some pupal-mating behavior in H. erato, but suggests that adult mating is likely the prevalent mating strategy in this species. These findings have important implications for the role of color pattern and female mate choice in the generation of reproductive isolation in this diverse genus.

Effects of past mating behavior versus past ejaculation on male mate choice and male attractiveness.

Project description:Past reproductive effort allows males to assess their ability to acquire mates, but it also consumes resources that can reduce their future competitive ability. Few studies have examined how a male's reproductive history affects his subsequent mate choice, and, to date, no study has determined the relative contribution of past mating behavior and past ejaculate production because these two forms of investment are naturally highly correlated. Here, we disentangled the relative effects of past mating behavior and past ejaculate production in mosquitofish (Gambusia holbrooki) by experimentally preventing some males from ejaculating when trying to mate. We assessed the effect of mating behavior on mate choice by comparing males that had previously been with or without access to females and male rivals for 8 and 16 weeks and assessed the effect of ejaculation on mate choice by comparing males that either could or could not ejaculate when they had access to females for 16 weeks. Reproductive treatment did not affect male attractiveness, but it did affect male mate choice. Somewhat surprisingly, in five of the six treatment-by-age at testing combinations, males preferred a female in the vicinity of a male rival over a solitary female. This preference was marginally stronger for males that had previously engaged in mating behavior but were unaffected by past ejaculate production. We discuss the potential benefits to males of associating with another male when seeking mates. This is the first study to quantify the relative influence of pre- and post-copulatory reproductive investment on male mate choice.

Kin recognition: Neurogenomic response to mate choice and sib mating avoidance in a parasitic wasp.

Project description:Sib mating increases homozygosity, which therefore increases the risk of inbreeding depression. Selective pressures have favoured the evolution of kin recognition and avoidance of sib mating in numerous species, including the parasitoid wasp Venturia canescens. We studied the female neurogenomic response associated with sib mating avoidance after females were exposed to courtship displays by i) unrelated males or ii) related males or iii) no courtship (controls). First, by comparing the transcriptional responses of females exposed to courtship displays to those exposed to controls, we saw a rapid and extensive transcriptional shift consistent with social environment. Second, by comparing the transcriptional responses of females exposed to courtship by related to those exposed to unrelated males, we characterized distinct and repeatable transcriptomic patterns that correlated with the relatedness of the courting male. Network analysis revealed 3 modules of specific 'sib-responsive' genes that were distinct from other 'courtship-responsive' modules. Therefore, specific neurogenomic states with characteristic brain transcriptomes associated with different behavioural responses affect sib mating avoidance behaviour.

Variation in human mate choice: simultaneously investigating heritability, parental influence, sexual imprinting, and assortative mating.

Project description:Human mate choice is central to individuals' lives and to the evolution of the species, but the basis of variation in mate choice is not well understood. Here we looked at a large community-based sample of twins and their partners and parents ([Formula: see text] individuals) to test for genetic and family environmental influences on mate choice, while controlling for and not controlling for the effects of assortative mating. Key traits were analyzed, including height, body mass index, age, education, income, personality, social attitudes, and religiosity. This revealed near-zero genetic influences on male and female mate choice over all traits and no significant genetic influences on mate choice for any specific trait. A significant family environmental influence was found for the age and income of females' mate choices, possibly reflecting parental influence over mating decisions. We also tested for evidence of sexual imprinting, where individuals acquire mate-choice criteria during development by using their opposite-sex parent as the template of a desirable mate; there was no such effect for any trait. The main discernible pattern of mate choice was assortative mating; we found that partner similarity was due to initial choice rather than convergence and also at least in part to phenotypic matching.

Assortative mating and mate-choice contributes to the maintenance of a developmental dimorphism in Streblospio benedicti.

Project description:Assortative mating, where individuals non-randomly mate with respect to phenotype or genotype, can occur when preferences between potential mates have evolved. When such mate preferences occur in a population it can drive evolutionary and phenotypic divergence. But the extent to which assortative mating, mate preference, and development are evolutionarily linked remains unclear. Here we use Streblospio benedicti, a marine annelid with a rare developmental dimorphism, to investigate if mate-choice could contribute to developmental evolution. For S. benedicti two types of ecologically and phenotypically similar adults persist in natural populations, but they give rise to distinctly different offspring with alternative life-histories. This dimorphism persists despite the absence of post-zygotic reproductive barriers, where crosses between the developmental types can produce phenotypically intermediate offspring. How this life-history strategy evolved remains unknown, but assortative mating is a typical first step in evolutionary divergence. Here we investigate if female mate-choice is occurring in this species. We find that mate preferences could be contributing to the maintenance of alternative developmental and life-history strategies.

Sex Differences in Mate Choice Preference Characteristics of <i>Aequidens rivulatus</i>.

Project description:The mating roles of males and females, to a certain extent, are dynamic and variable. Several factors influence the mate choice process. Nonetheless, the main preference features have not yet been fully understood in Aequidens rivulatus. In this study, because of its natural pairing characteristics, A. rivulatus was selected to explore the mate choice preferences of different sexes. Specifically, male and female behavioral performances were described and quantified through a "no-choice paradigm" during mate choice. A total of 12 behavioral performances were defined in male mate choice (experiment 1), whereas 14 behavioral performances were defined in female mate choice (experiment 2). According to the obtained results, unselected females did not display any proactive behaviors in experiment 1, whereas unselected males exhibited proactive behaviors in experiment 2, including quivering, nipping, tail beating, swimming up and down, and aggression. It was also found that both male and female individuals tend to express dislike rather than like. Those behaviors with higher frequencies (e.g., quivering) often mean less energy expenditure, thus easier repeatability. Moreover, principal component analysis (PCA) was employed to extract and identify mate choice preference features. Preliminary results indicated that male preferences for a mate were mainly associated with body size, behavioral intention, and appearance, whereas the intensity of female preferences was in the order of body size, appearance, and behavioral intention. In addition, sex hormone levels were associated with mate choices.

Past and present resource availability affect mating rate but not mate choice in <i>Drosophila melanogaster</i>.

Project description:The choices of when, where, and with whom to mate represent some of the most important decisions an individual can make to increase their fitness. Several studies have shown that the resources available to an individual during development can dramatically alter their mating rate later in life, and even the choice of mate. However, an individual's surroundings and available resources can change rapidly, and it is not clear how quickly the redistribution of resources towards reproduction can change. To address this important question, we measured mating rate and mate choice among Drosophila melanogaster males that were manipulated in terms of both past resources (control vs. starvation) and the resources available during mate choice (food vs. no food). We found that males given access to ample resources prior to mate choice showed higher mating rates than those that were starved, in agreement with previous studies. However, we also found that this effect can be reversed quickly, as starved males given the opportunity to mate in a high-quality environment mated at frequencies equivalent to their fed counterparts. Although past and present resources affected mating rate, they did not affect mate choice, as males mated with high-quality females at high frequencies regardless of their condition and environment. Our results show that both current condition as well as the promise of future resources can dramatically influence individuals' investment into reproduction and that such mating decisions are extremely plastic and reliant on environmental cues.

No evidence for size-assortative mating in the wild despite mutual mate choice in sex-role-reversed pipefishes.

Project description:Size-assortative mating is a nonrandom association of body size between members of mating pairs and is expected to be common in species with mutual preferences for body size. In this study, we investigated whether there is direct evidence for size-assortative mating in two species of pipefishes, Syngnathus floridae and S. typhle, that share the characteristics of male pregnancy, sex-role reversal, and a polygynandrous mating system. We take advantage of microsatellite-based "genetic-capture" techniques to match wild-caught females with female genotypes reconstructed from broods of pregnant males and use these data to explore patterns of size-assortative mating in these species. We also develop a simulation model to explore how positive, negative, and antagonistic preferences of each sex for body size affect size-assortative mating. Contrary to expectations, we were unable to find any evidence of size-assortative mating in either species at different geographic locations or at different sampling times. Furthermore, two traits that potentially confer a fitness advantage in terms of reproductive success, female mating order and number of eggs transferred per female, do not affect pairing patterns in the wild. Results from model simulations demonstrate that strong mating preferences are unlikely to explain the observed patterns of mating in the studied populations. Our study shows that individual mating preferences, as ascertained by laboratory-based mating trials, can be decoupled from realized patterns of mating in the wild, and therefore, field studies are also necessary to determine actual patterns of mate choice in nature. We conclude that this disconnect between preferences and assortative mating is likely due to ecological constraints and multiple mating that may limit mate choice in natural populations.

Mate choice is affected by parasite infestation rate of the choosing individual as well as of potential mating partners

Project description:Abstract Parasites are known to be a key driving force in mate choice and are important for the expression and evolution of ornaments and behavioral traits being used. However, there is little experimental evidence on how the parasite’s burden of the choosing individual is integrated into the mate-choice process and how it affects decision-making, especially in relation to parasite infestation of potential mates. Thus, the aim of our study was to determine whether female house sparrows Passer domesticus adjust their mate preference according to their own as well as the parasite load of prospective partners. To do this, we experimentally manipulated female parasite load and determined their mate preferences prior to and after parasite treatment. We manipulated the chronic coccidian parasite burden of females either by initiating the acute infection phase via re-infecting them with coccidian or by temporally reducing the parasite load of coccidia. We then measured the effect of this manipulation on mate preference by presenting females with a choice of four stimuli: three males with similar ornaments, but unmanipulated, naturally varying chronic coccidiosis levels, and an unmanipulated control female. Additionally, we recorded some males’ behavior in relation to their infection status pointing toward an increased or reduced interest in mating. We found that females preferred highly infested males prior to manipulation, regardless of their own infestation level. However, after manipulation, infested females avoided highly infested males probably in response to the deterioration of their health condition by parasites. Our study suggests that mate-choice decisions are more complex when they are mediated by parasites. The implications of parasites for evolutionary theories of sexual signaling and mate choice are discussed.
