Gold-nanoparticle-assisted laser perturbation of chromatin assembly reveals unusual aspects of nuclear architecture within living cells.

ABSTRACT: Chromatin organization within the nucleus is a vital regulator of genome function, yet its mechanical coupling to the nuclear architecture has remained elusive. To directly investigate this coupling, we locally modulated chromatin structure in living cells using nanoparticle-based laser perturbation. Unusual differences in the response of the cell nucleus were observed depending on the nuclear region that was perturbed--the heterochromatin, the euchromatin, and the nuclear envelope. This response varied under different conditions of cellular perturbations such as ATP depletion, apoptosis, and inhibition of histone deacetylases. Our studies implicate heterochromatin organization in imparting mechanical stability to the cell nucleus and suggest that nuclear size and shape are the result of interplay between nuclear and cytoplasmic anchors.

Chemical Printing of Biological Tissue by Gold Nanoparticle-Assisted Laser Ablation.

Project description:A chemical printing method based on gold nanoparticle (AuNP)-assisted laser ablation has been developed. By rastering a thin layer of AuNPs coated on a rat kidney tissue section with a UV laser, biomolecules are extracted and immediately transferred/printed onto a supporting glass substrate. The integrity of the printed sample is preserved, as revealed by imaging mass spectrometric analysis. By studying the mechanism of the extraction/printing process, transiently molten AuNPs were found to be involved in the process, as supported by the color and morphological changes of the AuNP thin film. The success of this molecular printing method was based on the efficient laser-nanomaterial interaction, that is, the strong photoabsorption, laser-induced heating, and phase-transition properties of the AuNPs. It is anticipated that the molecular printing method can be applied to perform site-specific printing, which extracts and transfers biochemicals from different regions of biological tissue sections to different types of supporting materials for subsequent biochemical analysis with the preservation of the original tissue samples.

High-Performance Sample Substrate of Gold Nanoparticle Multilayers for Surface-Assisted Laser Desorption/Ionization Mass Spectrometry.

Project description:The development of a sample substrate with superior performance for desorption and ionization of analyte is the key issue to ameliorate the quality of mass spectra for measurements of small molecules in surface-assisted laser desorption/ionization mass spectrometry (SALDI-MS). Herein, the homogeneous sample substrate of gold nanoparticle multilayers (AuNPs-ML) with hexagonal lattice was successfully prepared by self-assembly technique. With strong surface plasmon resonance absorption and superior photothermal effect, the sample substrate of AuNPs-ML exhibited high signal sensitivity and low background noise for the detection of model analyte of glucose without additional matrixes in SALDI-MS. Furthermore, compared to merchant matrixes of α-cyano-4-hydroxycinnamic acid (CHCA) and 2,5-dihydroxybenzoic acid (DHB), the sample substrate of AuNPs-ML was demonstrated to ameliorate the quality of mass spectra, including signal strength, background interference and signal/noise (S/N) ratio. The sucrose and tryptophan were also measured to show the extensive applications of AuNPs-ML sample substrate for the detections of small molecules in SALDI-MS. Most importantly, the remarkable reproducibility of glucose mass spectra with relative signal of 7.3% was obtained by the use of AuNPs-ML sample substrate for SALDI-MS. The homogeneous sample substrate of AuNPs-ML greatly improved the quality of mass spectra because of its strong absorption of laser energy, low specific heat, high heat conductivity and extraordinary homogeneity. We believe that AuNPs-ML could be a practical sample substrate for small molecule detection in SALDI-MS.

Pulsed laser in liquid grafting of gold nanoparticle-carbon support composites.

Project description:We developed a novel pulsed laser-assisted process for the fabrication of advanced composites of nonequilibrium gold nanoparticles on carbon fiber paper supports. Our one-step process integrates the generation of nanoparticles with their surface attachment and solves longstanding nanoparticle adhesion and electrical contact issues. Irradiation of hydrophilic carbon fiber paper submerged in aqueous HAuCl4 solution by nanosecond laser pulses produced composites with uniform distribution of gold nanoparticles on carbon fibers, taking advantage of the high internal surface area of carbon fiber paper. The pulsed laser-grafted composites exhibited zero measurable charge transfer resistance between gold nanoparticles and the carbon support, leading to superior cathode performance over conventionally prepared electrodes for electrocatalytic hydrogen evolution in aqueous bicarbonate reduction.

Gold nanoparticle mediated laser transfection for efficient siRNA mediated gene knock down.

Project description:Laser based transfection methods have proven to be an efficient and gentle alternative to established molecule delivery methods like lipofection or electroporation. Among the laser based methods, gold nanoparticle mediated laser transfection bears the major advantage of high throughput and easy usability. This approach uses plasmon resonances on gold nanoparticles unspecifically attached to the cell membrane to evoke transient and spatially defined cell membrane permeabilization. In this study, we explore the parameter regime for gold nanoparticle mediated laser transfection for the delivery of molecules into cell lines and prove its suitability for siRNA mediated gene knock down. The developed setup allows easy usage and safe laser operation in a normal lab environment. We applied a 532 nm Nd:YAG microchip laser emitting 850 ps pulses at a repetition rate of 20.25 kHz. Scanning velocities of the laser spot over the sample of up to 200 mm/s were tested without a decline in perforation efficiency. This velocity leads to a process speed of ?8 s per well of a 96 well plate. The optimal particle density was determined to be ?6 particles per cell using environmental scanning electron microscopy. Applying the optimized parameters transfection efficiencies of 88% were achieved in canine pleomorphic adenoma ZMTH3 cells using a fluorescent labeled siRNA while maintaining a high cell viability of >90%. Gene knock down of d2-EGFP was demonstrated and validated by fluorescence repression and western blot analysis. On basis of our findings and established mathematical models we suppose a mixed transfection mechanism consisting of thermal and multiphoton near field effects. Our findings emphasize that gold nanoparticle mediated laser transfection provides an excellent tool for molecular delivery for both, high throughput purposes and the transfection of sensitive cells types.

Effects of macrophage polarization on gold nanoparticle-assisted plasmonic photothermal therapy.

Project description:Tumor associated macrophages (TAM) are key pathogenic factors in neoplastic diseases. They are known to have plasticity and can polarize into two opposing phenotypes, including the tumoricidal M1 and the protumoral M2 phenotypes with high prevalence of M2-phentoypes in patients with poor prognosis. Strategies for targeting M2-TAM may consequently increase the efficacy of therapeutic strategies for cancer treatment. Gold nanorod-assisted plasmonic photothermal therapy (PPTT) has emerged as a promising treatment for cancer but the effects of macrophage polarization parameters in the performance of this new treatment modality is still unknown. Herein, human monocytic THP-1 cells were polarized into two opposite phenotypic macrophages (M1-TAM and M2-TAM) and their response to PPTT was examined. M2-TAM exhibits a three-fold increase in AuNP uptake compared to M1-TAM. Laser irradiation results in selective killing of pro-tumoral M2-TAM after treatment with AuNPs with limited effects on anti-tumoral M1-TAM. A positive correlation between the expression of CD206 marker and the AuNP uptake may indicate the role of CD206 in facilitating AuNP uptake. Our findings also suggest that the differences in AuNP avidity and uptake between the M1-TAM and M2-TAM phenotypes may be the rationale behind the effectiveness of PPTT in the treatment of solid tumors.

A DNAzyme-gold nanoparticle probe for uranyl ion in living cells.

Project description:DNAzymes have shown great promise as a general platform for detecting metal ions, as many metal-specific DNAzymes can be obtained using in vitro selection. While DNAzyme-based metal sensors have found many applications in the extracellular environment, no intracellular application of DNAzyme sensors has yet been reported. Here, we demonstrate a novel type of metal ion sensor for intracellular metal ion detection. The probe consists of a 13 nm gold nanoparticle (AuNP) core functionalized with a shell consisting of a uranyl-specific 39E DNAzyme whose enzyme strand contains a thiol at the 3' end for conjugation to the AuNP, and whose substrate strand is modified with a Cy3 fluorophore at the 5' end and a molecular quencher at the 3' end. In the absence of uranyl, the fluorescence of the Cy3 is quenched by both AuNP and the molecular quencher. In the presence of uranyl, the DNAzyme cleaves the fluorophore-labeled substrate strand, resulting in release of the shorter product strand containing the Cy3 and increased fluorescence. We demonstrate that this DNAzyme-AuNP probe can readily enter cells and can serve as a metal ion sensor within a cellular environment, making it the first demonstration of DNAzymes as intracellular metal ion sensors. Such a method can be generally applied to the detection of other metal ions using other DNAzymes selected through in vitro selection.

Tunable-wavelength nanosecond laser tailoring of plasmon resonance spectra of gold nanoparticle colloids.

Project description:Metal nanoparticles have applications across a range of fields of science and industry. While there are numerous existing methods to facilitate their large-scale production, most face limitations, particularly in achieving reproducible processes and minimizing undesirable impurities. Common issues are varying particle sizes and aggregates with unfavorable spectral properties. Researchers are currently developing methods to separate or modify nanoparticle sizes and shapes post-synthesis and to eliminate impurities. One promising approach involves laser light irradiation and enables the changing of nanoparticle sizes and shapes while controlling crucial spectral parameters. In this work, we present a novel extension of this method by irradiating nanoparticle colloids with variable-wavelength nanosecond laser pulses on both sides of the extinction band. Our results demonstrate the use of gradual laser wavelength tuning to optimize the photothermal reshaping of gold nanorods and achieve precise control over the plasmon resonance band. By irradiating both sides of the plasmon resonance band, we execute a multistep tuning process, controlling the band's width and spectral position. A statistical analysis of SEM images reveals differences in the nanorod morphology when irradiated on the long- or short-wavelength side of the plasmon resonance band. The fine-tuning of plasmonic spectral properties is desirable for various applications, including the development of sensors and filters and the exploitation of the photothermal effect. The findings of this study can be extended to other plasmonic nanostructures.

Shear-Assisted Laser Transfer of Metal Nanoparticle Ink to an Elastomer Substrate.

Project description:Selective laser sintering of metal nanoparticle ink is an attractive technology for the creation of metal layers at the microscale without any vacuum deposition process, yet its application to elastomer substrates has remained a highly challenging task. To address this issue, we introduced the shear-assisted laser transfer of metal nanoparticle ink by utilizing the difference in thermal expansion coefficients between the elastomer and the target metal electrode. The laser was focused and scanned across the absorbing metal nanoparticle ink layer that was in conformal contact with the elastomer with a high thermal expansion coefficient. The resultant shear stress at the interface assists the selective transfer of the sintered metal nanoparticle layer. We expect that the proposed method can be a competent fabrication route for a transparent conductor on elastomer substrates.
