Spiders fluoresce variably across many taxa.

ABSTRACT: The evolution of fluorescence is largely unexplored, despite the newfound occurrence of this phenomenon in a variety of organisms. We document that spiders fluoresce under ultraviolet illumination, and find that the expression of this trait varies greatly among taxa in this species-rich group. All spiders we examined possess fluorophores in their haemolymph, but bright fluorescence appears to result when a spider sequesters fluorophores in its setae or cuticle. By sampling widely across spider taxa, we determine that fluorescent expression is labile and has evolved multiple times. Moreover, examination of the excitation and emission properties of extracted fluorophores reveals that spiders possess multiple fluorophores and that these differ among some families, indicating that novel fluorophores have evolved during spider diversification. Because many spiders fluoresce in wavelengths visible to their predators and prey (birds and insects), we propose that natural selection imposed by predator-prey interactions may drive the evolution of fluorescence in spiders.

Making Nitronaphthalene Fluoresce.

Project description:Nitroaromatic compounds are inherently nonfluorescent, and the subpicosecond lifetimes of the singlet excited states of many small nitrated polycyclic aromatic hydrocarbons, such as nitronaphthalenes, render them unfeasible for photosensitizers and photo-oxidants, despite their immensely beneficial reduction potentials. This article reports up to a 7000-fold increase in the singlet-excited-state lifetime of 1-nitronaphthalene upon attaching an amine or an N-amide to the ring lacking the nitro group. Varying the charge-transfer (CT) character of the excited states and the medium polarity balances the decay rates along the radiative and the two nonradiative pathways and can make these nitronaphthalene derivatives fluoresce. The strong electron-donating amine suppresses intersystem crossing (ISC) but accommodates CT pathways of nonradiate deactivation. Conversely, the N-amide does not induce a pronounced CT character but slows down ISC enough to achieve relatively long lifetimes of the singlet excited state. These paradigms are key for the pursuit of electron-deficient (n-type) organic conjugates with promising optical characteristics.

Polyethylene Glycol 20k. Does It Fluoresce?

Project description:Polyethylene glycol (PEG) is a polyether compound commonly used in biological research and medicine because it is biologically inert. This simple polymer exists in variable chain lengths (and molecular weights). As they are devoid of any contiguous π-system, PEGs are expected to lack fluorescence properties. However, recent studies suggested the occurrence of fluorescence properties in non-traditional fluorophores like PEGs. Herein, a thorough investigation has been conducted to explore if PEG 20k fluoresces. Results of this combined experimental and computational study suggested that although PEG 20k could exhibit "through-space" delocalization of lone pairs of electrons in aggregates/clusters, formed via intermolecular and intramolecular interactions, the actual contributor of fluorescence between 300 and 400 nm is the stabilizer molecule, i.e., 3-tert-butyl-4-hydroxyanisole present in the commercially available PEG 20k. Therefore, the reported fluorescence properties of PEG should be taken with a grain of salt, warranting further investigation.

The study of hidden habitats sheds light on poorly known taxa: spiders of the Mesovoid Shallow Substratum.

Project description:The scarce and biased knowledge about the diversity and distribution of Araneae species in the Iberian Peninsula is accentuated in poorly known habitats such as the Mesovoid Shallow Substratum (MSS). The aim of this study was to characterize the spiders inventory of the colluvial MSS of the Sierra de Guadarrama National Park, and to assess the importance of this habitat for the conservation of the taxon. Thirty-three localities were selected across the high peaks of the Guadarrama mountain range and they were sampled for a year using subterranean traps specially designed to capture arthropods in the MSS. Species accumulation curves were built both for the observed species richness and for the non-parametric richness estimators. The literature was reviewed in order to update the distributional maps of the rarest species. Forty-two species were collected, of which four were species new to science. More than half were represented by one or two individuals which caused the accumulation curves to rise slowly and to end without reaching an asymptote. Almost half of the species showed significant increases in their Iberian distribution ranges. Two species were recorded for the first time in the Iberian Peninsula and 32 species were new additions to the spider checklist of the Sierra de Guadarrama National Park.

Convergent evolution revealed by paraphyly and polyphyly of many taxa of oribatid mites: A molecular approach.

Project description:A reliable phylogeny is crucial for understanding the evolution and radiation of animal taxa. Phylogenies based on morphological data may be misleading due to frequent convergent evolution of traits-a problem from which molecular phylogenies suffer less. This may be particularly relevant in oribatid mites, an ancient soil animal taxon with more than 11,000 species, where the classification of species into high-ranking taxa such as superfamilies is equivocal. Here, we present a molecular phylogeny of 317 oribatid, 4 astigmatid and 17 endeostigmatid mite species/taxa based on 18S rDNA sequences. We aimed at testing the validity of the 41 superfamilies of oribatid mites recognized by Norton and Behan-Pelletier (in Krantz and Walter, A manual of Acarology, 3rd ed., Texas Technical University Press, Lubbock, 2009). The results indicate that 17 of the 41 oribatid mite superfamilies are monophyletic but that 18 superfamilies are paraphyletic or polyphyletic (5 superfamilies were only included with one species and Microzetoidea were not included). Our findings point to the importance of convergent evolution in polyphyletic oribatid mite taxa. Convergent evolution and the old age of mites likely resulted in a mosaic-like distribution of morphological characters impeding phylogenetic reconstructions based on morphology, calling for molecular approaches to improve oribatid mite systematics.

ASTRAL-II: coalescent-based species tree estimation with many hundreds of taxa and thousands of genes.

Project description:MotivationThe estimation of species phylogenies requires multiple loci, since different loci can have different trees due to incomplete lineage sorting, modeled by the multi-species coalescent model. We recently developed a coalescent-based method, ASTRAL, which is statistically consistent under the multi-species coalescent model and which is more accurate than other coalescent-based methods on the datasets we examined. ASTRAL runs in polynomial time, by constraining the search space using a set of allowed 'bipartitions'. Despite the limitation to allowed bipartitions, ASTRAL is statistically consistent.ResultsWe present a new version of ASTRAL, which we call ASTRAL-II. We show that ASTRAL-II has substantial advantages over ASTRAL: it is faster, can analyze much larger datasets (up to 1000 species and 1000 genes) and has substantially better accuracy under some conditions. Phylogenomics of North American cybaeid spiders (Araneae, Cybaeidae), including the description of new taxa from the Klamath Mountains Geomorphic Province.

Project description:The systematics of humble-in-appearance brown spiders ("marronoids"), within a larger group of spiders with a modified retrolateral tibial apophysis (the RTA Clade), has long vexed arachnologists. Although not yet fully settled, recent phylogenomics has allowed the delimitation and phylogenetic relationships of families within marronoids to come into focus. Understanding relationships within these families still awaits more comprehensive generic-level sampling, as the majority of described marronoid genera remain unsampled for phylogenomic data. Here we conduct such an analysis in the family Cybaeidae Banks, 1892. We greatly increase generic-level sampling, assembling ultraconserved element (UCE) data for 18 of 22 described cybaeid genera, including all North American genera, and rigorously test family monophyly using a comprehensive outgroup taxon sample. We also conduct analyses of traditional Sanger loci, allowing curation of some previously published data. Our UCE phylogenomic results support the monophyly of recognized cybaeids, with strongly supported internal relationships, and evidence for five primary molecular subclades. We hypothesize potential morphological synapomorphies for most of these subclades, bringing a robust phylogenomic underpinning to cybaeid classification. A new cybaeid genus Siskiyu gen. nov. and species Siskiyuarmilla sp. nov. is discovered and described from far northern California and adjacent southern Oregon and a new species in the elusive genus Cybaeozyga, C.furtiva sp. nov., is described from far northern California.

Fluorescence confocal microscopy for margin assessment in prostatectomy: IP8-FLUORESCE study protocol.

Project description:BackgroundRadical prostatectomy (RP) represents the cornerstone of surgical treatment for prostate cancer. Assessing surgical margin status intraoperatively with current techniques remains challenging due to high costs in the context of an already stretched pathology workforce. Fluorescence confocal microscopy (FCM) is a promising technique to detect margins in prostate cancer surgery not bound by such limitations.Study designThe Imperial Prostate 8 - Fluorescence Confocal Microscopy for Rapid Evaluation of Surgical Cancer Excision (IP8-FLUORESCE) study is a multicentre, prospective, ex vivo, 'blinded', comparative cohort study. It aims to assess the accuracy of digital FCM for detection of prostate cancer at surgical margins compared to traditional histopathology.EndpointsThe primary endpoint is the accuracy of digital FCM for detection of prostate cancer at surgical margins on a per-patient level, reported with sensitivity, specificity, positive and negative predictive values.Patients and methodsA total of 153 patients with localised prostate cancer undergoing robot-assisted RP across three UK National Health Service tertiary referral centres will be recruited. Following RP, prostate specimens will undergo immediate immersion in Acridine Orange solution, scanning 'en face' with FCM using the Histolog® Scanner, and subsequent formalin fixation and paraffin embedding. Two independent, 'blinded' uro-pathologists will report both the FCM images and the histopathology slides. Recruitment commenced on 17 August 2023.
