Zeolite catalyzed solvent-free one-pot synthesis of dihydropyrimidin-2(1H)-ones--a practical synthesis of monastrol.

ABSTRACT: A zeolite-catalyzed, simple, one-pot, solvent-free, cost effective, and environmentally benign process for the synthesis of dihydropyrimidones is described. This reaction is scaleable to multigram scale and the catalyst is recyclable. This methodology has resulted in an efficient synthesis of monastrol, a potent inhibitor of kinesin Eg5.

Project description:Several chiral primary amines, mainly those derived from the cinchona alkaloids, were evaluated as the organocatalysts for the asymmetric Biginelli reaction. With the quinine-derived amine catalyst 1 and after extensive optimization of the reaction conditions, 3,4-dihydropyrimidin-2(1H)-ones were obtained in moderate to good yields and 51-78% ee from a three-component reaction of aryl and aliphatic aldehydes, urea, and acetoacetate.

An Efficient, Eco-friendly and Sustainable One-Pot Synthesis of 3,4-Dihydropyrimidin-2(1H)-ones Directly from Alcohols Catalyzed by Heteropolyanion-Based Ionic Liquids.

Project description:Efficient, eco-friendly and sustainable access to 3,4-dihydropyrimidin-2(1H)-ones directly from alcohols under microwave and solvent-free conditions has been reported. The practical protocol involves heteropolyanion-based catalyzed oxidation of alcohols to aldehydes with NaNO₃ as the oxidant followed by cyclocondensation with dicarbonyl compounds and urea or thiourea in a two-step, one-pot manner. Compatibility with different functional groups, good to excellent yields and reusable catalysts are the main highlights. The utilization of alcohols instead of aldehydes is a valid and green alternative to the classical Biginelli reaction.

A facile hydrothermal synthesis of high-efficient NiO nanocatalyst for preparation of 3,4-dihydropyrimidin-2(1H)-ones.

Project description:The present work introduces a one-step and facile hydrothermal procedure as a green process for the first time to synthesize nickel(II) oxide (NiO) nanoparticles. The as-prepared nanomaterials were used as high efficient, low toxic and cost catalyst for the synthesis of some organic compounds. Ni(NO3)2 and some natural extract were used as a surfactant for the first time to synthesis NiO nanomaterials. A high synthesis yield (91%) was obtained for S2. Rietveld analysis affirmed the cubic crystal system of the obtained NiO nanocatalyst. The morphology studies were carried out with the FESEM method and the images revealed a change from non-homogenous to homogenous spherical particles when the Barberryas was used instead of orange blossom surfactant. Besides, the images revealed that the particle size distribution was in the range of 20 to 60 nm. The synthesized catalysts were used for the first time in Biginelli multicomponent reactions (MCRs) for the preparation of 3,4-dihydropyrimidin-2(1H)-ones (DHPMs) under the present facile reaction conditions. High yield (97%) of the final product was achieved at the optimum condensation reaction conditions (Catalyst: 60 mg; temperature: 90 °C and time: 90 min) when ethyl acetoacetate/methyl acetoacetate (1 mmol), benzaldehyde (1 mmol) and urea (1.2 mmol) were used. A kinetic study affirmed pseudo-first-order model for Biginelli reactions followed the pseudo-first-order model.

Gold-catalyzed one-step practical synthesis of oxetan-3-ones from readily available propargylic alcohols.

Project description:A general solution for the synthesis of various oxetan-3-ones is developed. This reaction uses readily available propargylic alcohols as substrates and proceeds without the exclusion of moisture or air ("open flask"). Notably, oxetan-3-one, a highly valuable substrate for drug discovery, can be prepared in one step from propargyl alcohol in a fairly good yield. The facile formation of the strained oxetane ring provides strong support for the intermediacy of alpha-oxo gold carbenes. This safe and efficient generation of gold carbenes via intermolecular alkyne oxidation offers a potentially general entry into alpha-oxo metal carbene chemistry without using hazardous diazo ketones.

Highly Efficient Synthesis of Substituted 3,4-Dihydropyrimidin-2-(1<i>H</i>)-ones (DHPMs) Catalyzed by Hf(OTf)₄: Mechanistic Insights into Reaction Pathways under Metal Lewis Acid Catalysis and Solvent-Free Conditions.

Project description:In our studies on the catalytic activity of Group IVB transition metal Lewis acids, Hf(OTf)₄ was identified as a highly potent catalyst for "one-pot, three-component" Biginelli reaction. More importantly, it was found that solvent-free conditions, in contrast to solvent-based conditions, could dramatically promote the Hf(OTf)₄-catalyzed formation of 3,4-dihydro-pyrimidin-2-(1H)-ones. To provide a mechanistic explanation, we closely examined the catalytic effects of Hf(OTf)₄ on all three potential reaction pathways in both "sequential bimolecular condensations" and "one-pot, three-component" manners. The experimental results showed that the synergistic effects of solvent-free conditions and Hf(OTf)₄ catalysis not only drastically accelerate Biginelli reaction by enhancing the imine route and activating the enamine route but also avoid the formation of Knoevenagel adduct, which may lead to an undesired byproduct. In addition, ¹H-MMR tracing of the H-D exchange reaction of methyl acetoacetate in MeOH-d₄ indicated that Hf(IV) cation may significantly accelerate ketone-enol tautomerization and activate the β-ketone moiety, thereby contributing to the overall reaction rate.

A facile, rapid, one-pot regio/stereoselective synthesis of 2-iminothiazolidin-4-ones under solvent/scavenger-free conditions.

Project description:A rapid and efficient one pot solvent/scavenger-free protocol for the synthesis of 2-iminothiazolidin-4-ones has been developed. Interestingly, the regio/stereoselective synthesis affords the regioisomeric (Z)-3-alkyl/aryl-2-(2-phenylcyclohex-2-enylimino)thiazolidin-4-one as the sole product in good yield. The selectivities observed have been rationalized based on the relative magnitude of the allylic strains developed during the course of the reaction. This is the first report wherein the impact of allylic strains in directing the regiocyclization has been noted.

Discovery and structural optimization of 4-(4-(benzyloxy)phenyl)-3,4-dihydropyrimidin-2(1H)-ones as RORc inverse agonists.

Project description:AimRetinoic acid receptor-related orphan nuclear receptors (RORs) are orphan nuclear receptors that show constitutive activity in the absence of ligands. Among 3 subtypes of RORs, RORc is a promising therapeutic target for the treatment of Th17-mediated autoimmune diseases. Here, we report novel RORc inverse agonists discovered through structure-based drug design.MethodsBased on the structure of compound 8, a previously described agonist of RORa, a series of 4-(4-(benzyloxy)phenyl)-3,4-dihydropyrimidin-2(1H)-one derivatives were designed and synthesized. The interaction between the compounds and RORc was detected at molecular level using AlphaScreen assay. The compounds were further examined in 293T cells transfected with RORc and luciferase reporter gene. Thermal stability shift assay was used to evaluate the effects of the compounds on protein stability.ResultsA total of 27 derivatives were designed and synthesized. Among them, the compound 22b was identified as the most potent RORc inverse agonist. Its IC50 values were 2.39 μmol/L in AlphaScreen assay, and 0.82 μmol/L in inhibition of the cell-based luciferase reporter activity. Furthermore, the compound 22b displayed a 120-fold selectivity for RORc over other nuclear receptors. Moreover, a molecular docking study showed that the structure-activity relationship was consistent with the binding mode of compound 22b in RORc.Conclusion4-(4-(Benzyloxy)phenyl)-3,4-dihydropyrimidin-2(1H)-one derivatives are promising candidates for the treatment of Th17-mediated autoimmune diseases, such as rheumatoid arthritis, psoriasis, and multiple sclerosis.

An Automated Process for a Sequential Heterocycle/Multicomponent Reaction: Multistep Continuous Flow Synthesis of 5-(Thiazol-2-yl)-3,4-Dihydropyrimidin-2(<i>1H</i>)-ones.

Project description:The first example of a sequential heterocycle formation/multicomponent reaction using an automated continuous flow microreactor assembly is reported. Consecutive Hantzsch thiazole synthesis, deketalization, and Biginelli multicomponent reaction provides rapid and efficient access to highly functionalized, pharmacologically significant 5-(thiazol-2-yl)-3,4-dihydropyrimidin-2(1H)-ones without isolation of intermediates. These complex small molecules are generated in reaction times less than 15 min and in high yields (39-46%) over three continuous chemical steps.

The Biginelli-based hit optimization process enabled a thoughtful exploration of the structure-activity and structure-selectivity relationships for this chemotype, enabling the identification of ligands that combine structural simplicity with excellent hA2B AdoR affinity and remarkable selectivity profiles.

Project description:Based on the Biginelli reaction of β-ketoesters, arylaldehydes, and urea/thiourea, we created a green radical synthesis procedure for 3,4-dihydropyrimidin-2-(1H)-ones/thiones. A single-electron redox mediator was applied to a solution of ethanol in an air environment, at room temperature, and with blue LEDs as a renewable energy source in order to create. The objective of this research is to create a halide perovskite that is widely available, affordable, recyclable, and economically feasible. A factor mentioned in the discussion is that the procedure tolerates a variety of donating and withdrawing functional groups while still offering a very fast rate and excellent yields. The range of yields is quite uniform (86-94%, average: 90.4%), and the range of reaction times is very quick (4-8 min, average: 5.8 min). Furthermore, gram-scale cyclization shows that it is applicable for use in industry. Additionally, CsPbBr3 is quite stable and can be used six times in a row without experiencing significant structural changes or activity loss, which has been extremely helpful in meeting industrial needs and environmental issues.
