Project description:It has been suggested repeatedly that zoo-kept giant anteaters (Myrmecophaga tridactyla) have higher body masses than their free-ranging conspecifics. Here, we assess this hypothesis by comparing body mass data of 184 female and 173 male individuals kept in zoos to published and unpublished data of free-ranging animals (n = 215). In zoos, the average adult body mass of all individuals was 45.9 ± 9.6 kg (range 19.2-72.6 kg), whereas body mass for free-ranging specimens was 33.0 ± 2.3 kg (21-45.5 kg). Among those zoo animals in which a sufficient number of subsequent intra-annual measures had been taken, we assessed visually whether regular, annual fluctuations in body mass were evident. We observed regular, likely seasonal, cycles in body mass, which (i) may be plausibly attributed to ambient temperature changes that affect the body temperature of anteaters, and (ii) would likely not have occurred if the animals were affected by obesity. Additionally, we explain our hypothesis that in the case of giant anteaters, the discrepancy in body mass between natural habitats and zoos most likely is not indicative of a generally obese population in zoos, as is typically concluded in the case of similar comparisons in primates, but represents an unleashing of the full growth potential of the species by conditions of optimal energy and nutrient provision. Future recording of not only body mass but also body dimensions is recommended to better compare the body condition of free-ranging and zoo-managed individuals.

Project description:Spatial ecology data are essential for conservation purposes, especially when extinction risk is influenced by anthropogenic actions. Space use can reveal how individuals use the habitat, how they organize in space, and which components are key resources for the species.We evaluated the space use and multiscale habitat selection of giant anteaters (Myrmecophaga tridactyla), a vulnerable Neotropical mammal, in a Cerrado site within a human-modified landscape in southeastern Brazil.We used GPS transmitters to track eight anteaters in the wild. With the resulting dataset, we estimated home range and core-area sizes and then used two overlap indexes. We assessed habitat selection by compositional analysis and analyzed events of spatio-temporal proximity.The average Brownian bridge kernel estimate of home range size was 3.41 km2 (0.92-7.9). Regarding home range establishment, five individuals showed resident behavior. Males (n = 4) had larger home ranges and were more active than females (n = 4). Despite the spatial overlap of home range (above 40% in four dyads), maximum temporal space sharing was 18%. Giant anteaters were found in proximity. Habitat selection favored savanna, and exotic timber plantation was always avoided. Roads and built-up areas were selected secondarily at the landscape level.The selection of anthropogenic sites denotes behavioral plasticity regarding modified habitats. However, the high selectivity for savanna, at all levels, demonstrates a high dependence on natural habitats, which provide the necessary resources for the species. The recurrent proximity of male-to-female anteaters may indicate reproductive behavior, which is essential for maintaining this isolated population.

Project description:Anaplasma marginale, a well-known cattle pathogen of tropical and subtropical world regions, has been previously molecularly characterized in a giant anteater (Myrmecophaga tridactyla) from Corrientes, Argentina. Ticks or other hematophagous arthropod involved in the wild transmission cycle remained unknown. The aim of the present study was to analyze the simultaneous occurrence of A. marginale in blood samples and ticks from giant anteaters from Corrientes in order to investigate if ticks could be relevant in the transmission among these mammals. Blood samples from 50 giant anteaters collected in different years and 26 ticks Amblyomma dubitatum and A. sculptum were studied through the molecular amplification of two unequivocal species-specific genes from A. marginale: msp5 and msp1β. Twenty five giant anteaters and tick organs (salivary glands, gut and oviduct) from 11 ticks tested positive to the A. marginale DNA amplification. The further molecular characterization through MSP1a tandem repeats analysis revealed the presence of genotypes circulating among giant anteaters that had been previously identified in cattle blood samples from the same geographical region. These results confirm the presence of A. marginale in giant anteaters in Corrientes and suggests that A. dubitatum and A. sculptum ticks could be involved in the transmission among giant anteaters. Future studies will determine the role of these tick species in the wild transmission cycle in the study area and the eventual connection with the domestic cycle.

Project description:Canine distemper outbreak and coinfections in three giant anteaters and in a maned wolf has been described. Three giant anteaters developed respiratory and digestive clinical signs after the introduction of a maned wolf to a Wildlife Rehabilitation Center. The maned wolf and two anteaters died, and one anteater was euthanized. Post mortem and histopathologic exams revealed lesions associated with numerous intraepithelial inclusion bodies, mainly in the respiratory and digestive systems. Infection by distemper virus was confirmed in all animals by RT-PCR and gene sequencing, which revealed the Europe 1/ South America 1 strain, closely related to the strain from Canis familiaris. In addition to distemper, the animals had other comorbidities, such as toxoplasmosis and salmonellosis in the maned wolf and cutaneous candidiasis in an anteater. Considering the chronology of clinical manifestation in both species and the viral characterization, it is possible that the maned wolf was the source of infection to the anteaters. This study demonstrates the importance of implementing biosecurity measures in enclosures that house animals of different species, highlighting the importance of quarantine before introduction of new animals into the same environment.

Project description:In general, large mammal species with highly specialized feeding behavior and solitary habits are expected to suffer genetic consequences from habitat loss and fragmentation. To test this hypothesis, we analyzed the genetic diversity distribution of the threatened giant anteater inhabiting a human-modified landscape. We used 10 microsatellite loci to assess the genetic diversity and population structure of 107 giant anteaters sampled in the Brazilian Central-Western region. No genetic population structuring was observed in this region suggesting no gene flow restriction within the studied area. On the other hand, the moderate level of genetic diversity (Ho = 0.54), recent bottleneck detected and inbreeding (Fis, 0.13; p ≤ 0.001) signatures suggest potential impacts on the genetic variation of this Xenarthra. Additionally, a previous demographic reduction was suggested. Thus, considering the increased human-promoted impacts across the entire area of distribution of the giant anteater, our results can illustrate the potential effects of these disturbances on the genetic variation, allowing us to request the long-term conservation of this emblematic species.

Project description:The giant anteater (Myrmecophaga tridactyla, Pilosa, Linnaeus 1758) belongs to the mammalian order Pilosa and presents a large distribution along South America, occupying a great variety of habitats. It is listed in the IUCN Red List of threatened species as Vulnerable. Despite threatened, there is a lack of studies regarding its genetic variability. The aim of this study was to examine the genetic diversity and patterns of genetic structure within remaining populations. We analyzed 77 individuals from seven different populations distributed in four biomes across Brazil: Cerrado, Pantanal, Atlantic Forest and Amazon Forest. We sequenced two mitochondrial markers (control region and Cyt-b) and two nuclear markers (AMELY and RAG2). We found high genetic diversity within subpopulations from National Parks of Serra da Canastra and Emas, both within the Cerrado biome, with signs of population expansion. Besides, we found a notable population structure between populations from the Cerrado/Pantanal and Amazon Forest biomes. This data is a major contribution to the knowledge of the evolutionary history of the species and to future management actions concerning its conservation.

Project description:Canine morbillivirus, also known as canine distemper virus (CDV), induces a contagious multisystemic disease caused by an enveloped RNA virus belonging to the genus Morbillivirus within the family Paramyxoviridae. CDV replicates readily in epithelial, nerve and lymphoid tissues; it is excreted in urine, feces, saliva, oral and nasal discharge; and its major route of entry for infection is through the respiratory system. Although the virus was originally believed to infect domestic dogs, new studies have shown that it can also naturally or experimentally infect non-domestic hosts. A recent blood test performed on a giant anteater (Myrmecophaga tridactyla) found Lentz inclusions in the animal's leucocytes. A rapid CDV test, an RT-PCR assay and pathology findings confirmed this report of canine morbillivirus in this species, which corresponds to the second report of CDV infection in the order Pilosa, family Myrmecophagidae in central west Brazil.

Project description:Habitat loss is the main threat to biodiversity conservation worldwide. Some species may be particularly susceptible to the effects of fragmentation and the isolation of populations. The impacts of human activity on wild animal populations may be understood through relationships between individual genetic data and spatial landscape variables, particularly when considering local population dynamics influenced by fragmented habitats. Thus, the objective of this study was to analyze the population structure and genetic diversity of the giant anteater (Myrmecophaga tridactyla) using an individual sampling scheme (ISS) on a regional geographic scale. Data were collected from 41 specimens from twenty different locations in São Paulo State, Brazil, and six polymorphic microsatellite loci were genotyped. Our results indicate that barriers to gene flow exist and have segregated individuals of the farther away areas into two spatially structured clusters. The populations were also found to have high genetic diversity. The experimental sampling approach used herein enabled an analysis of the population dynamics of the giant anteater on a regional scale, as well as the identification of priority populations for genetic resource conservation for this species. The results reflect the need for adequate management plans. The efficacy of the sampling scheme may vary based on the study model used, but we argue that the use of an ISS combined with suitable molecular markers and statistical methods may serve as an important tool for initial analyses of threatened or vulnerable species, particularly in anthropized regions where populations are small or hard to characterize.

Project description:Microsporidia are emerging, opportunistic fungi that infect a diverse population of vertebrates and invertebrates. Mammals of the superorder Xenarthra can harbor and transmit several pathogens, acting as important sources of infection for spreading various zoonoses. Microsporidia have not yet been described in this group of animals, the aim of this study was to analyze the occurrence of the zoonotic microsporidian Encephalitozoon intestinalis, Encephalitozoon cuniculi and Enterocytozoon bieneusi in the feces of giant anteater (Myrmecophaga tridactyla) and armadillo species (Priodontes maximus, Euphractus sexcinctus, Dasypus novemcinctus, Cabassous squamicaudis) monitored by Wild Animal Conservation Institute (ICAS) in Brazil. Fecal samples (n=127) were subjected to DNA extraction with the QIAamp Fast DNA Stool Mini Kit. Amplification by PCR was performed with generic primers and the product generated from this reaction was subjected to nested PCR with specific primers. Eleven samples tested positive for E. intestinalis, two from M. tridactyla (2/56, 3.6%), seven from E. sexcinctus (7/51, 13.7%), two from P. maximus (2/11, 18%) and one from C. squamicaudis (1/3, 33.3%). There was a predominance of positive results in adult animals, of both sexes across and in the Pantanal and Cerrado biomes. In conclusion, the prevalence in Xenarthra was 9.4%, with a higher occurrence in armadillos than in anteaters. Therefore, the species of wild mammals studied here should be considered reservoirs of microsporidian pathogens and have a relevant role in the concept of One Health.
