Compositional analysis of lignocellulosic feedstocks. 1. Review and description of methods.

ABSTRACT: As interest in lignocellulosic biomass feedstocks for conversion into transportation fuels grows, the summative compositional analysis of biomass, or plant-derived material, becomes ever more important. The sulfuric acid hydrolysis of biomass has been used to measure lignin and structural carbohydrate content for more than 100 years. Researchers have applied these methods to measure the lignin and structural carbohydrate contents of woody materials, estimate the nutritional value of animal feed, analyze the dietary fiber content of human food, compare potential biofuels feedstocks, and measure the efficiency of biomass-to-biofuels processes. The purpose of this paper is to review the history and lineage of biomass compositional analysis methods based on a sulfuric acid hydrolysis. These methods have become the de facto procedure for biomass compositional analysis. The paper traces changes to the biomass compositional analysis methods through time to the biomass methods currently used at the National Renewable Energy Laboratory (NREL). The current suite of laboratory analytical procedures (LAPs) offered by NREL is described, including an overview of the procedures and methodologies and some common pitfalls. Suggestions are made for continuing improvement to the suite of analyses.

Project description:BackgroundThe use of fossil carbon sources for fuels and petrochemicals has serious impacts on our environment and is unable to meet the demand in the future. A promising and sustainable alternative is to substitute fossil carbon sources with microbial cell factories converting lignocellulosic biomass into desirable value added products. However, such bioprocesses require availability of suitable and efficient microbial biocatalysts, capable of utilizing C5 sugars and tolerant to inhibitory compounds generated during pretreatment of biomass. In this study, the performance of a collection of lactic acid bacteria was evaluated regarding their properties with respect to the conversion of lignocellulosic feedstocks. The strains were examined for their ability to utilize xylose and arabinose as well as their resistance towards common inhibitors from pretreated lignocellulosic biomass (furan derivatives, phenolic compounds, weak acids).ResultsAmong 296 tested Lactobacillus and Pediococcus strains, 3 L. pentosus, 1 P. acidilactici and 1 P. pentosaceus isolates were found to be both capable of utilizing xylose and arabinose and highly resistant to the key inhibitors from chemically pretreated lignocellulosic biomass. When tested in broth with commonly found combinations of inhibitors, the selected strains showed merely 4%, 1% and 37% drop in growth rates for sugarcane bagasse, wheat straw and soft wood representatives, respectively, as compared to Escherichia coli MG1655 showing decreased growth rates by 36%, 21% and 90%, respectively, under the same conditions.ConclusionThe study showed that some strains of Lactobacilli and Pediococci have the potential to be used as production platforms for value-added products from pretreated lignocellulosic biomass. Selected Lactobacilli and Pediococci strains were able to tolerate the key inhibitors in higher concentrations compared to E.coli; in addition, as these isolates were also capable of fermenting xylose and arabinose, they constitute good candidates for efficient lignocellulosic feedstock bioconversions.

Project description:Background: Lignocellulosic biomass has many functionalities that hold huge potential for material, energy or chemistry applications. To support advanced applications, the biomass must be milled into ultrafine powder to increase reactivity. This milling unit operation needs to be fully mastered to deliver high-quality standard end-products. Here we studied the relationship between the characteristics of the starting lignocellulosic plant material and the properties of the resulting ultrafine powder in different ball-mill process routes. Methods: Two lignocellulosic biomasses (pine bark and wheat straw) with contrasted compositional and mechanical properties were milled using three ball-mill set-ups delivering different balances of impact force and attrition force. The resulting powders were analysed for particle characteristics (size, agglomeration extent, shape) and powder flow properties (compressibility, cohesion) using a dynamic powder rheometer. Results: Pine bark is more amenable to a fast particle size reduction than the fibrous wheat straw. The resulting pine bark powders appear less compressible but much more cohesive than the straw powders due to particle shape, density and composition factors. The mill set-up working by attrition as dominant mechanical force (vibratory ball mill) produced a mix of large, elongated particles and higher amounts of fines as it acts mainly by erosion, the resulting powder being more prone to agglomerate due to the abundance of fines. The mill set-up working by impact as dominant mechanical force (rotary ball mill) produced more evenly distributed particle sizes and shapes. The resulting powder is less prone to agglomerate due to a preferential fragmentation mechanism. Conclusions: The attrition-dominant mill yields powders with dispersed particle sizes and shapes and the poorest flow properties, while the impact-dominant mill yields more agglomeration-prone powders. The mill set-up working with impact and attrition as concomitant mechanical forces (stirred ball mill) produces powders with better reactivity and flow properties compared to rotary and vibratory mills.

Project description:As the genome carries the historical information of a species' biotic and environmental interactions, analyzing changes in genome structure over time by using powerful statistical physics methods (such as entropic segmentation algorithms, fluctuation analysis in DNA walks, or measures of compositional complexity) provides valuable insights into genome evolution. Nucleotide frequencies tend to vary along the DNA chain, resulting in a hierarchically patchy chromosome structure with heterogeneities at different length scales that range from a few nucleotides to tens of millions of them. Fluctuation analysis reveals that these compositional structures can be classified into three main categories: (1) short-range heterogeneities (below a few kilobase pairs (Kbp)) primarily attributed to the alternation of coding and noncoding regions, interspersed or tandem repeats densities, etc.; (2) isochores, spanning tens to hundreds of tens of Kbp; and (3) superstructures, reaching sizes of tens of megabase pairs (Mbp) or even larger. The obtained isochore and superstructure coordinates in the first complete T2T human sequence are now shared in a public database. In this way, interested researchers can use T2T isochore data, as well as the annotations for different genome elements, to check a specific hypothesis about genome structure. Similarly to other levels of biological organization, a hierarchical compositional structure is prevalent in the genome. Once the compositional structure of a genome is identified, various measures can be derived to quantify the heterogeneity of such structure. The distribution of segment G+C content has recently been proposed as a new genome signature that proves to be useful for comparing complete genomes. Another meaningful measure is the sequence compositional complexity (SCC), which has been used for genome structure comparisons. Lastly, we review the recent genome comparisons in species of the ancient phylum Cyanobacteria, conducted by phylogenetic regression of SCC against time, which have revealed positive trends towards higher genome complexity. These findings provide the first evidence for a driven progressive evolution of genome compositional structure.

Project description:Soil mineral compositions are often complex and spatially diverse, with each mineral exhibiting characteristic chemical properties that determine the intrinsic total concentration of soil nutrients and their phyto-availability. Defining soil mineral-nutrient relationships is therefore important for understanding the inherent fertility of soils for sustainable nutrient management, and data-driven approaches such as cluster analysis allow for these relations to be assessed in new detail. Here the fuzzy-c-means clustering algorithm was applied to an X-ray powder diffraction (XRPD) dataset of 935 soils from sub-Saharan Africa, with each diffractogram representing a digital signature of a soil's mineralogy. Nine mineralogically distinct clusters were objectively selected from the soil mineralogy continuum by retaining samples exceeding the 75% quantile of the membership coefficients in each cluster, yielding a dataset of 239 soils. As such, samples within each cluster represented mineralogically similar soils from different agro-ecological environments of sub-Saharan Africa. Mineral quantification based on the mean diffractogram of each cluster illustrated substantial mineralogical diversity between the nine groups with respect to quartz, K-feldspar, plagioclase, Fe/Al/Ti-(hydr)oxides, phyllosilicates (1:1 and 2:1), ferromagnesians, and calcite. Mineral-nutrient relationships were defined using the clustered XRPD patterns and corresponding measurements of total and/or extractable (Mehlich-3) nutrient concentrations (B, Mg, K, Ca, Mn, Fe, Ni, Cu and Zn) in combination with log-ratio compositional data analysis. Fe/Al/Ti/Mn-(hydr)oxides and feldspars were found to be the primary control of total nutrient concentrations, whereas 2:1 phyllosilicates were the main source of all extractable nutrients except for Fe and Zn. Kaolin minerals were the most abundant phyllosilicate group within the dataset but did not represent a nutrient source, which reflects the lack of nutrients within their chemical composition and their low cation exchange capacity. Results highlight how the mineral composition controls the total nutrient reserves and their phyto-availability in soils of sub-Saharan Africa. The typical characterisation of soils and their parent material based on the clay particle size fraction (i.e. texture) and/or the overall silica component (i.e. acid and basic rock types) alone may therefore mask the intricacies of mineral contributions to soil nutrient concentrations.
