Scent of danger: floc formation by a freshwater bacterium is induced by supernatants from a predator-prey coculture.

ABSTRACT: We investigated predator-prey interactions in a model system consisting of the bacterivorous flagellate Poterioochromonas sp. strain DS and the freshwater bacterium Sphingobium sp. strain Z007. This bacterial strain tends to form a subpopulation of grazing-resistant microscopic flocs, presumably by aggregation. Enhanced formation of such flocs could be demonstrated in static batch culture experiments in the presence of the predator. The ratio of aggregates to single cells reached >0.1 after 120 h of incubation in an oligotrophic growth medium. The inoculation of bacteria into supernatants from cocultures of bacteria and flagellates (grown in oligotrophic or in rich media) also resulted in a substantially higher level of floc formation than that in supernatants from bacterial monocultures only. After separation of supernatants on a C(18) cartridge, the aggregate-inducing activity could be assigned to the 50% aqueous methanolic fraction, and further separation of this bioactive fraction could be achieved by high-pressure liquid chromatography. These results strongly suggest the involvement of one or several chemical factors in the induction of floc formation by Sphingobium sp. strain Z007 that are possibly released into the surrounding medium by flagellate grazing.

Project description:Camera traps are a unique survey tool used to monitor a wide variety of mammal species. Camera trap (CT) data can be used to estimate animal distribution, density, and behaviour. Attractants, such as scent lures, are often used in an effort to increase CT detections; however, the degree which the effects of attractants vary across species is not well understood. We investigated the effects of scent lure on mammal detections by comparing detection rates between 404 lured and 440 unlured CT stations sampled in Alberta, Canada over 120 day survey periods between February and August in 2015 and 2016. We used zero-inflated negative binomial generalized linear mixed models to test the effect of lure on detection rates for a) all mammals, b) six functional groups (all predator species, all prey, large carnivores, small carnivores, small mammals, ungulates), and c) four varied species of management interest (fisher, Pekania pennanti; gray wolf, Canis lupus; moose, Alces alces; and Richardson's ground squirrel; Urocitellus richardsonii). Mammals were detected at 800 of the 844 CTs, with nearly equal numbers of total detections at CTs with (7110) and without (7530) lure, and variable effects of lure on groups and individual species. Scent lure significantly increased detections of predators as a group, including large and small carnivore sub-groups and fisher specifically, but not of gray wolf. There was no effect of scent lure on detections of prey species, including the small mammal and ungulate sub-groups and moose and Richardson's ground squirrel specifically. We recommend that researchers explicitly consider the variable effects of scent lure on CT detections across species when designing, interpreting, or comparing multi-species surveys. Additional research is needed to further quantify variation in species responses to scent lures and other attractants, and to elucidate the effect of attractants on community-level inferences from camera trap surveys.

Project description:BackgroundBacterial floc formation plays a central role in the activated sludge (AS) process. The formation of AS flocs has long been known to require exopolysaccharide biosynthesis. We had demonstrated that both expolysaccharides and PEP-CTERM (a short C-terminal domain includes a near-invariant motif Pro-Glu-Pro (PEP)) proteins were required for floc-forming in Zoogloea resiniphila MMB, a dominant AS bacterium. However, the PEP-CTERM proteins are not encoded in the genome of AS bacterium Shinella zoogloeoides ATCC 19623 (formerly known as Zoogloea ramigera I-16-M) and other sequenced AS bacteria strains. The mechanism underlying floc formation of Shinella and related AS bacteria remained largely unclear.ResultsIn this study, we have sequenced and annotated the complete genome of S. zoogloeoides ATCC 19623 (aka I-16-M), previously isolated in USA and treated as the neotype for the AS floc-forming bacterium Zoogloea ramigera I-16-M, and another AS strain XJ20 isolated in China. Mariner transposon mutagenesis had been conducted to isolate floc-forming-deficient mutants in the strain ATCC 19623 as previously performed by using Tn5 transposon three decades ago. The transposon insertional sites of multiple mutants were mapped to the gene cluster for bacterial cellulose synthesis (bcs) and secretion, and the role played by these genes in floc-formation had been further confirmed by genetic complementation. Interestingly, the restriction map of this bcs locus-flanking region was highly similar to that of the previously identified DNA fragment required for floc-formation in 1980s. Cellulase treatment abolished the floc-forming phenotype of S. zoogloeoides ATCC 19623 but not that of Z. resiniphila MMB strain. The FTIR spectral analyses revealed that the samples extracted from S. zoogloeoides ATCC 19623 were cellulose polymer.ConclusionOur results indicated that we have largely reproduced and completed the unfinished pioneering work on AS floc-formation mechanism, demonstrating that the floc-formation and flocculating capability of Shinella were mediated by extracellular cellulose polymers.

Project description:Small mammal abundances are frequently limited by resource availability, but predators can exert strong lethal (mortality) and nonlethal (e.g., nest abandonment) limitations. Artificially increasing resource availability for uncommon small mammals provides a unique opportunity to examine predator-prey interactions. We used remote cameras to monitor 168 nest platforms placed in the live tree canopy (n = 23 young forest stands), primarily for arboreal red tree voles (tree voles; Arborimus longicaudus), over 3 years (n = 15,510 monitoring-weeks). Tree voles frequently built nests and were detected 37% of monitoring-weeks, whereas flying squirrels (Glaucomys oregonensis) built nests infrequently but were detected 45% of monitoring-weeks. Most nest predators were detected infrequently (<1% of monitoring-weeks) and were positively correlated with tree vole presence. Weasels (Mustela spp.) were highly effective predators of tree voles (n = 8 mortalities; 10% of detections) compared to owls (n = 1), flying squirrels (n = 2), and Steller's jays (n = 1). Tree vole activity decreased from 84.1 (95% confidence interval [CI]: 56.2, 111.9) detections/week 1-week prior to a weasel detection to 4.7 detections/week (95% CI: 1.7, 7.8) 1-week postdetection and remained low for at least 12 weeks. Interpretations of predator-prey interactions were highly sensitive to how we binned continuously collected data and model results from our finest bin width were biologically counter-intuitive. Average annual survival of female tree voles was consistent with a previous study (0.14; 95% CI: -0.04 [0.01], 0.32) and high compared to many terrestrial voles. The relative infrequency of weasel detections and inefficiency of other predators did not provide strong support for the hypothesis that predation per se limited populations. Rather, predation pressure, by reducing occupancy of already scarce nest sites through mortality and nest abandonment, may contribute to long-term local instability of tree vole populations in young forests. Additional monitoring would be needed to assess this hypothesis.
