ABSTRACT: In the title compound {systematic name: N-[(4R,5R)-3,11-dioxo-10-oxa-6-thia-2-aza-tricyclo-[6.3.0.0(2,5)]undec-1(8)-en-4-yl]-2-phenyl-acetamide}, C(16)H(14)N(2)O(4)S, the four- and five-membered rings adopt planar conformations (with r.m.s. deviations of 0.0349 and 0.0108 Å respectively) while the six-membered ring adopts a half-chair, or envelope-like, conformation with the S atom in the flap position. In the crystal, mol-ecules are linked by N-H⋯O hydrogen bonds.

Novel Conversions of a Multifunctional, Bio-sourced Lactone Carboxylic Acid.

Project description:The plant-derived compounds furfuryl alcohol and itaconic anhydride are known to undergo a Diels-Alder reaction at room temperature and in bulk to efficiently give an alkene-containing lactone carboxylic acid. Reported here is the conversion of this substance to a variety of derivatives via hydrogenation, epoxidation, or halolactonization reactions. Most notable is the formation of a set of three related acrylate or methacrylate esters (see graphical abstract) produced by direct acylative ring opening of ether bonds using Sc(OTf)3 and (meth)acrylic anhydride. These esters are viewed as promising candidates for use as biorenewable monomers in reversible addition-fragmentation chain transfer (RAFT) polymerization reactions.

Project description:The title compound, C(12)H(9)NO(2)S, belongs to the nitro-gen-containing group of heterocyclic organic compounds and crystallized with two mol-ecules per asymmetric unit. In the crystal, both molecules form inversion dimers linked by pairs of O-H-O hydrogen bonds. Weak symmetry-related C-H-O inter-actions link the carboxyl dimers along b axis. The dihedral angle between the two aromatic rings in the two mol-ecules are 55.75 (14) and 58.33 (13)°.

5-Amino-1-phenyl-1H-pyrazole-4-carboxylic acid.

Project description:In the mol-ecule of the title compound, C(10)H(9)N(3)O(2), the pyrazole ring is approximately coplanar with the amino and carboxyl groups. The phenyl group is twisted by 48.13 (3)° relative to this plane. An intra-molecular N-H⋯O hydrogen bond stabilizes the planar conformation of the mol-ecule. The mol-ecules are linked into two-dimensional sheets by two strong inter-molecular N-H⋯N and O-H⋯O hydrogen bonds. The latter forms the classic carboxylic acid dimer motif.

Project description:In the mol-ecule of the title compound, C(10)H(10)N(2)O(5), intra-molecular C-H⋯O inter-actions result in the formation of a five- and a six-membered ring. The five-membered ring is planar and is oriented at a dihedral angle of 0.34 (3)° with respect to the plane of the aromatic ring, while the six-membered ring has a twist conformation. In the crystal structure, inter-molecular C-H⋯O inter-actions link the mol-ecules into chains.

Discovery of antiproliferative and anti-FAK inhibitory activity of 1,2,4-triazole derivatives containing acetamido carboxylic acid skeleton.

Project description:Small molecule inhibitors of the focal adhesion kinase are regarded as promising tools in our armamentarium for treating cancer. Here, we identified four 1,2,4-triazole derivatives that inhibit FAK kinase significantly and evaluated their therapeutic potential. Most tested compounds revealed potent antiproliferative activity in HepG2 and Hep3B liver cancer cells, in which 3c and 3d were the most potent (IC50 range; 2.88 ~ 4.83 µM). Compound 3d possessed significant FAK inhibitory activity with IC50 value of 18.10 nM better than the reference GSK-2256098 (IC50 = 22.14 nM). The preliminary mechanism investigation by Western blot analysis showed that both 3c and 3d repressed FAK phosphorylation comparable to GSK-2256098 in HepG2 cells. As a result of FAK inhibition, 3c and 3d inhibited the pro-survival pathways by decreasing the phosphorylation levels of PI3K, Akt, JNK, and STAT3 proteins. This effect led to apoptosis induction and cell cycle arrest. Taken together, these results indicate that 3d could serve as a potent preclinical candidate for the treatment of cancers.

5-Methyl-1-phenyl-1H-1,2,3-triazole-4-carboxylic acid.

Project description:The title compound, C(10)H(9)N(3)O(2), was synthesized from azido-benzene and ethyl acetyl-acetate. A pair of hydrogen bonds [2.617?(2)?Å] inter-connects a pair of the carboxyl groups, forming an R(2) (2)(8) inversion dimer, a frequent motif in carboxylic acids. In the title structure, the bonding H atom in the aforementioned O-H?O hydrogen bond is significantly shifted towards the acceptor O atom [the donor and acceptor O-H distances are 1.25?(4) and 1.38?(4)?Å, respectively]. A plot of the O?O versus O-H distances in compounds with paired carboxyl groups shows that the title structure belongs to the group of structures with abnormally long O-H distances with regard to the O?O contacts. The displacement of the bonding H atom towards the centre of the hydrogen bond is concomitant with more equal C-O bonding distances in the carboxyl group.

The packing of the mol-ecules in the crystal structure is governed by C-H⋯O and O-H⋯O hydrogen-bonding inter-actions.

Project description:In the title compound, C(11)H(8)F(3)N(3)O(2), there are two mol-ecules in the asymmetric unit wherein the phenyl rings make dihedral angles of 65.3 (2) and 85.6 (2)° with the pyrazole rings. In the crystal, pairs of mol-ecules are held together by O-H⋯O hydrogen bonds between the carboxyl groups, forming a centrosymmetric dimer with an R(2) (2)(8) motif. Intra-molecular N-H⋯O inter-actions are also present.

Project description:The title compound, C(14)H(15)NO(5), was obtained from the reaction of acetanilide with dimethyl acetyl-enedicarboxyl-ate in the presence of potassium carbonate. The C=C double bond adopts an E configuration and the geometry around the amide N atom is almost planar rather than pyramidal (mean deviation of 0.0032 Å from the C(3)N plane). The packing of the mol-ecules in the crystal structure is stabilized by inter-molecular C-H⋯O hydrogen bonds.
